Verfahren zur Herstellung des neuen Furfuryloxy-carbonylfluorids
Durch die Arbeit von Losse und Mitarbeitern (H. Jeschkeit, G. Losse und K. Neubert, Chem. Ber.
Bd. 99 [1966], S. 2803) ist mit demFurfuryloxycarbonylrest eineAminoschutzgruppe, die sich besonders leicht acidolytisch abspalten lässt, für die Peptidchemie verfügbar geworden.
Eine kritische Untersuchung über die Brauchbarkeit dieser Gruppe bei der Synthese von Peptiden wurde bisher vor allem durch das relativ umständliche Herstellungsverfahren für die Furfuryloxycarbonyl- aminosäuren erschwert. Da das Furfuryloxycarbonylchlorid bisher nicht dargestellt werden konnte bzw. instabil ist, haben Jeschkeit, Losse und Neubert diese Derivate über die Isocyanatofettsäureester durch Umsetzen mit Furfurylalkohol und anschliessende Verseifung der Ester dargestellt. Nach diesem mehrstufigen und daher zeitraubenden Verfahren waren bisher nur die Furfuryloxycarbonylderivate der bi- funktionellen Aminosäuren zugänglich.
Bei den trifunktionellen Aminosäuren versagte das Verfahren infolge von Nebenreaktionen der dritten funktionellen Gruppe während der Umsetzung mit Phosgen oder bei der anschliessenden Verseifung.
Es wurde nun gefunden, dass das bisher unbekannte Furfuryloxycarbonylfluorid in fast quantitativer Ausbeute bei Temperaturen von 0 bis -700C aus Carbonylchloridfluorid und Furfurylalkohol in einem inerten Lösungsmittel und in Gegenwart von säurebindenden Mitteln, wie Metalloxyden bzw. Hydroxyden oder tert. Basen, vorzugsweise Pyridin, erhalten werden kann. Es ist in Lösung bei Temperaturen unter- halb -300C längere Zeit haltbar. Schon bei 0 C färbt sich die Lösung allmählich dunkel, während bei Zimmertemperatur Zersetzung unter kräftiger CC-Entwicklung erfolgt.
Das Carbonylchloridfluorid wird nach bekannten Methoden durch Fluorierung von Phosgen erhalten.
Es kann als Rohware, die unverändertes Phosgen und Fluorphosgen enthält, eingesetzt werden.
Das Furfuryloxycarbonylfluorid eignet sich hervorragend zur Synthese der Furfuryloxycarbonylaminosäuren, die bequem als Dicyclohexylammoniumsalze kristallisiert und isoliert werden können.
In der folgenden Tabelle sind einige Beispiele für die Herstellung von Furfuryloxycarbonylaminosäuren mittels Furfuryloxycarbonylfluorid bei stationärem pH-Wert angeführt.
Wie man aus dieser Tabelle entnehmen kann, sind auf diesem Wege auch die Derivate der trifunk- tionellen Aminosäuren bequem zugänglich. Als Aminosäurekomponenten können natürliche oder synthetische, racemische und optisch aktive Aminosäuren oder Iminosäuren oder Aminosulfonsäuren mit und ohne Seitenketten verwendet werden, die noch mindestens ein ersetzbaresWasserstoffatom an derAminobzw. Iminogruppe enthalten. Wie die Beispiele zeigen, können die Seitenketten auch noch zusätzliche funktionelle Gruppen enthalten. Beim Histidin erhält man ein Bisacylderivat ; ebenso lässt sich auch die phenolische Hydroxylgruppe von Tyrosin acylieren.

In einem 500 ems Dreihalskolben mit Rührer, Thermometer und einem auf -700C gehaltenen Rück- flusskühler werden 75 cm3 absolutes Methylenchlorid vorgelegt und anschliessend 31g 80% iges Carbonylchloridfluorid (0, 3 Mol)'bei einer Badtemperatur von -700C aufkondensiert.
Innerhalb l'h lässt man ein Gemisch von 24, 5 g (0, 25 Mol) frisch destilliertem Furfurylalkohol, 20 g (0, 25 Mol) absolutem Pyridin und 75 ml absolutem Methylenchlorid langsam unter Rühren zutropfen (Kühltemperatur-70 C, Sumpftemperatur-70 C). Im Verlauf von etwa 1/2 h lässt man die Reaktionstemperatur auf -250C ansteigen und destilliert dann dasMethylenchlorid bei einer Badtemperatur von -250C im Vakuum vollständig ab.
Den Rückstand versetzt man mit 150 ml absolutem Äther und saugt vom festen Pyridinhydrochlorid ab.

Die Verbindung zeigte keine Alkoholbande mehr im IR-Spektrum ; die Carbonylbande liegt bei 1820 cm-l.
Die Verbindung ist nur in Lösung (Methylenchlorid) bei Temperaturen unter -300C längere Zeit haltbar. Bei Zimmertemperatur zersetzt sich die Substanz in wenigen Minuten unter COz-Entwicklung.

- 2. Verfahren nach Anspruch l, dadurch gekennzeichnet, dass man als säurebindendes Mittel Metalloxyde, Hydroxyde oder tert. Basen, vorzugsweise Pyridin, verwendet.
