   Die Erfindung betrifft ein Verfahren zur Herstellung von neuen   D-2-subst.-6-Methyl-8-cyanomethyl-   ergolinen der allgemeinen Formel

   Das erfindungsgemässe Verfahren besteht in seinem Wesen darin, dass man eine Verbindung der allgemeinen Formel

 worin   R'Cl,   Br, J,   O-SO-CHg   oder   O-SO-p-Tolyl   bedeutet, mit einem Halogenierungsmittel umsetzt, wobei eine Verbindung der allgemeinen Formel

 gebildet wird, worin   R'und   X die oben angegebene Bedeutung haben, und man dann diese Verbindung mit einem anorganischen Cyanid in einem inerten Lösungsmittel umsetzt und gewünschtenfalls die erhaltene Verbindung in ein Salz überführt. 



   Die neuen Verbindungen können somit durch Einwirkung eines positiven Halogenierungsmittels auf D-6Methyl-8-halomethylergolin und anschliessenden Ersatz des Halogenatoms der Halogenmethylgruppedurch eine Cyanogruppe unter Verwendung von Natriumcyanid oder einem ähnlichen Reagens hergestellt werden. 



  Auf   ähnliche Welse ergibt die Halogenierung   von   D-6- Methyl-S-mesyloxymethyl (oder S-tosyloxymethyl) -ergo-     lininder 2-Stellung   leicht ein Zwischenprodukt, das mit Natriumcyanid oder andern ähnlichen anorganischen Cyaniden in einem inerten Lösungsmittel reagiert, wobei Verbindungen der obigen Formel   (1)   gebildet werden. 



   DieVorsilbe "D" bedeutetbei der Bezeichnung der Verbindungen der obigen Formel, dass die Stereochemie des Ergolinderivats identisch ist mit der der D-Lysergsäure, der natürlich vorkommenden Form. 



   Salze der Verbindungen der Formel (1) können mit organischen und anorganischen, pharmazeutisch verwendbaren Säuren gebildet werden. In diesem Falle sind die Salze löslich und werden durch Entfernung des Lösungsmittels durch Eindampfen im Vakuum gewonnen. Der entstehende Rückstand kann, wenn er nicht kristallin ist, aus Äthanol oder andern geeigneten Lösungsmitteln kristallisiert werden. 



   Die Verbindungen der Formel   (I)   sind wertvolle Gonadotropin-Inhibitoren. Als solche inhibieren sie die Lactation und sind insbesondere Prolaetininhibitoren. Die Verbindungen sind bei der Behandlung von unerwünschter Lactation, wie   postpartum-Laotatlon   und Galaktorrhoe, nützlich.

   Die Verbindungen können ebenfalls verwendet werden, um   Prolactin-abhängige   Adenocarcinoma und Prolaetin-abscheidende pituitäre Tumore, wie auch die folgenden Krankheiten zu behandeln : Forbes-Albright-Syndrom, Chiari-Frommel-Syndrom,   Gynäkomastie selbstund   Gynäkomastie, die als Ergebnis von der Verabreichung von östrogenen Stero-   idenfürdieprostatischeHypertrophieverursacht wird, fibrocystische Krankheiten derBrust (benigne Noduli),    prophylaktische Behandlung von Brustkrebs und Brustentwicklung, die durch Verabreichung von psychotropischen Arzneimitteln, beispielsweise Thorazin, verursacht wird.

Aus der obigen Tabelle ist ersichtlich, dass die beiden Verbindungen, die von der Formel (1) umfasst werden, die Prolactin-Gehalte stark vermindern und somit die Prolactin-Sekretion in den lactierenden (= Milch absondernden) weiblichen Ratten vermindern. 



   Es wurden Versuche entwickelt, um die Aktivität der   erfindungsgemäss erhältlichen Verbindungen   zu zeigen, das Wachstum von Adenocarcinoma In weiblichen Säugetieren zu unterdrücken. Beispielsweise wurde der folgende Versuch durchgeführt, bei dem zur Erläuterung Ratten als Beispiele für weibliche Säugetiere, die gegenüber Adenocarcinoma empfindlich sind, verwendet wurden. Die Bestimmung erfolgte folgendermassen :
Brusttumore wurden bei Ratten durch eine einzige orale Fütterung von 20 mg 7,12-Dimethylbenzanthracen (DMBA) induziert. Die Brustdrüsen wurden in wöchentlichen Intervallen auf Tumore abgetastet. Die Ratten wurden für den Versuch ausgewählt, wenn sie mindestens einen messbaren Tumor mit einem Durchmesser von ungefähr   0, 5 cm   hatten.

   Die Tiere mit Tumoren wurden In Gruppen von 5 eingeteilt und täglich mit unterschiedlichen Dosen an   D-2-Halogen-6-methyl-8-cyanomethylergolin,   suspendiert In Maisöl, behandelt. 



  Eine Gruppe von Vergleichsratten wurde nur mit Maisöl behandelt. Die Ergebnisse sind In Tabelle   n   aufgeführt. Die erfindungsgemäss erhältlichen Verbindungen zeigen eine bemerkenswerte Rückbildung der Brusttumore, wenn sie entweder subkutan oder oral verabreicht werden, wie aus dem obigen Versuch hervorgeht. 



   Die   erfindungsgemäss erhältlichen   Verbindungen besitzen weiterhin eine stark verminderte Toxizität, verglichen mit den bekannten Verbindungen, in denen die 2-Stellung unsubstituiert ist. Beispielsweise zeigen

 dieakute Toxizität inthylergolin einen LD50 -Wert von etwa 100 mg/kg bei Mäusen. Als Prolactin-Inhibitoren sind die erfindungsgemäss hergestellten 2-halogensubstituierten Verbindungen gleich wirksam wie die bekannten unsubstituierten   D-6-Methyl-8-cyanomethylergoline.   



   Das folgende Beispiel erläutert die Erfindung, ohne sie zu beschränken. 



   Beispiel :HerstellungvonD-2-Brom-6-methyl-8-cyanomethylergolin. 



   Eine Lösung aus 955 mg D-6-Methyl-8-brommethylergolin in 50 ml Dioxan wurde auf 60 bis   650C   unter   Stickstoffatmosphäre   erwärmt. Eine Lösung aus 600 mg   N-Bromsuccinimid   in 70 ml Dioxan wurde tropfenweise zugegeben. Die Reaktionsmischung wurde eine weitere 1/2 h auf 60 bis   650C   erwärmt, wonach das gesamte   N-Bromsuccinimid   zugegeben war. Die Reaktionsmischung wurde abgekühlt und wässerige Weinsäure

   wurde zugegeben, wobei wasserlösliche Tartratsalze der vorhandenen Ergolinbasen gebildet wurden. Die entstehende Mischung wurde mit Chloroform extrahiert und die Chloroformschicht wurde verworfen. Die wässerige Schicht wurde filtriert und dann wurde dazu festes Natriumbicarbonat gegeben, bis die Lösung basisch reagierte.

   In der basischen Reaktionsmischung waren die Ergolinbasen unlöslich. Die wässerige Schicht wurde mit Chloroform extrahiert und die Chloroformschicht wurde abgetrennt und getrocknet und das Chloroform durch Verdampfen im Vakuum entfernt. Chromatographie des entstehenden Rückstandes über Florisil unter Verwendung von Chloroform als Eluierungsmittel ergab Fraktionen, die, wie durch Dünn- schichtchromatographie gezeigt wurde, hauptsächlich ein Material enthielten, das sich von dem Ausgangsmaterial unterschied. Diese Fraktionen wurden vereinigt, das Lösungsmittel wurde durch Verdampfen im Vakuum entfernt und der entstehende Rückstand wurde aus Äther umkristallisiert, wobei man das bei der obigen Umsetzung gebildete D-2-Brom-6-methyl-8-brommethylergolin erhielt. Die Verbindung schmolz bei etwa 2150C (Zers.).

   90 mg D-2-Brom-6-methyl-8-brommethylergolin, hergestellt wie oben beschrieben, werden in 10 ml Dimethylsulfoxyd (DMSO) gelöst. 100 mg Natriumcyanid werden zugefügt und die entstehende Mischung wird auf   1050C   unter Stickstoffatmosphäre während etwa 45 min erwärmt. Die Reaktionsmischung wird gekühlt,   mit Wasser verdünntundfiltriert. Der Filterkuchen wird in   einer Lösungsmittelmischung aus Äthanol/Chloroform gelöst und die Lösungsmittel werden durch Verdampfen im Vakuum entfernt. Umkristallisation des Rückstandes aus Äther ergibt das   D-2-Brom-6-methyl-S-cyanomethylergolin,   das bei der obigen Umsetzung gebildet wurde. Die Verbindung schmilzt bei etwa 240 bis   243 C   (Zers.). 



   Entsprechend dem oben beschriebenen Verfahren kann das D-6-Methyl-8-mesyloxymethylergolin (hergestellt gemäss b)) in der 2-Stellung entsprechend dem Verfahren gemäss a) halogeniert werden, wobei man   das D-2-Chlor-6-methyl-S-mesyloxymethylergolin,   das D-2-Brom-6-methyl-8-mesyloxymethylergolin oder das   D-2-J od-6-methyl-S-mesyloxymethylergolin   erhält. 



   Die 8-Tosyloxymethylderivate, die hergestellt werden, wenn man das Methansulfonylehlorid bei dem Verfahren gemäss b) durch   p-Toluolsulfonylehlorid   ersetzt, können in der 2-Stellung halogeniert werden. 



   Herstellung der Salze :
Eine Lösung, die 560 mg   D-2-Brom-6-methyl-8-cyanomethylergolin   in etwa 40 ml Tetrahydrofuran (THF) enthält, wird hergestellt. Etwa 10 ml einer Lösung, die man herstellt, indem man 1 g Maleinsäure in 50 ml THF löst, werden unter Rühren zu der Lösung der Ergolinbase zugefügt. Etwa 200 ml Äther werden zugegeben und der entstehende Niederschlag wird durch Filtration abgetrennt. Das so hergestellte saure   D-2-Brom-6-methyl-S-cyanomethylergolinmaleat   schmilzt bei etwa 207 bis   209 C (Zers.).   



   Entsprechend dem obigen Verfahren wird saures   D-2-Chlor-6-methyl-8-cyanomethylergolinmaleat   hergestellt, das bei etwa 204 bis 206 C unter Zersetzung schmilzt. 



   Entsprechend dem obigen Verfahren werden   320mg D-2-Chlor-6-methyl-S-cyanomethylergolin   in 15 ml THF gelöst. Zu dieser Lösung fügt man eine Lösung aus Methansulfonsäure in THF, jeweils 1 Tropfen, bis die Zugabe eines weiteren Tropfens keinen weiteren Niederschlag mehr ergibt. Die THF-Lösung wird mit Äther verdünnt und die entstehende Mischung wird filtriert, wobei man das Methansulfonsäuresalz des D-2-   Chlor-6-methyl-8-eyanomethylergolins   erhält, das bei etwa 2950C unter Zersetzung nach Umkristallisation aus einer Lösungsmittelmischung aus Äthanol/Äther schmilzt. 



   Analyse :   ber. :   C = 54,61% H = 5, 60% N = 10, 61% Cl = 8, 95%   S = 8, 10%

Die Lösung wurde dann in gesättigtes wässeriges Natrium- bicarbonat gegossen und das organische Material wurde mit Chloroform extrahiert.

   Die Chloroformschicht wurde dann mit gesättigter wässeriger Weinsäure behandelt, dabei bildeten sich wasserlösliche Tartratsalze der vorhandenen Ergolinbasen. Die Wasserschicht wurde abgetrennt, die Chloroformschicht wurde verwor- fen. Die Wasserschicht wurde dann mit festem Natriumbicarbonat bis zur basischen Reaktion versetzt. Die ) Ergolinbase, die in der wässerigen alkalischen Lösung unlöslich war, wurde abgetrennt und in Chloroform gelöst. Die Chloroformschicht wurde abgetrennt und getrocknet. Eindampfen des Chloroforms im Vakuum ergab einen Rückstand aus   D-6-Methyl-8-brommethylergolin,   das bei der obigen Umsetzung gebildet wurde.

   Eine weitere Menge an D-6-Methyl-
8-mesyloxymethylergolin wurde durch Extraktion des Filtrats mit Äthylacetat, Abtrennung der   Äthylacetat-   schicht und Entfernung des Äthylacetats durch Eindampfen im Vakuum erhalten. Umkristallisation des D-6-   Methyl-8-mesyloxymethylergolins,   das wie oben beschrieben hergestellt wurde, aus Äthanol ergab Material mit einem Fp. von etwa 192 bis 1940C   (Zers.).

   The invention relates to a process for the preparation of new D-2-substituted-6-methyl-8-cyanomethyl-ergolines of the general formula

   The essence of the process according to the invention consists in that a compound of the general formula

 wherein R'Cl, Br, J, O-SO-CHg or O-SO-p-tolyl, is reacted with a halogenating agent, a compound of the general formula

 is formed, in which R 'and X have the meaning given above, and this compound is then reacted with an inorganic cyanide in an inert solvent and, if desired, the compound obtained is converted into a salt.



   The new compounds can thus be prepared by the action of a positive halogenating agent on D-6-methyl-8-halomethylergoline and then replacement of the halogen atom of the halomethyl group with a cyano group using sodium cyanide or a similar reagent.



  Similarly, halogenation of D-6-methyl-S-mesyloxymethyl (or S-tosyloxymethyl) -ergoline in the 2-position readily gives an intermediate which reacts with sodium cyanide or other similar inorganic cyanides in an inert solvent, compounds of the Formula (1) above can be formed.



   The prefix "D" in the designation of the compounds of the above formula means that the stereochemistry of the ergoline derivative is identical to that of D-lysergic acid, the naturally occurring form.



   Salts of the compounds of formula (1) can be formed with organic and inorganic, pharmaceutically acceptable acids. In this case the salts are soluble and are obtained by removing the solvent by evaporation in vacuo. The resulting residue can, if it is not crystalline, be crystallized from ethanol or other suitable solvents.



   The compounds of the formula (I) are valuable gonadotropin inhibitors. As such, they inhibit lactation and, in particular, are prolaetin inhibitors. The compounds are useful in the treatment of undesirable lactate ion such as postpartum laotatlon and galactorrhea.

   The compounds can also be used to treat prolactin-dependent adenocarcinomas and prolaetin-secreting pituitary tumors, as well as the following diseases: Forbes-Albright syndrome, Chiari-Frommel syndrome, gynecomastia itself and gynecomastia resulting from the administration of estrogenic steroids for prostatic hypertrophy, fibrocystic diseases of the breast (benign nodules), prophylactic treatment of breast cancer and breast development caused by administration of psychotropic drugs such as thorazine.



   The inhibition of prolaetin secretion by the compounds obtainable according to the invention can be illustrated with the following experiment: Groups of lactating female postpartum rats are fed the medicaments 4 to 8 days postpartum (the day of birth is designated as 0). It can be seen from the table above that the two compounds encompassed by the formula (1) greatly reduce the prolactin content and thus reduce the prolactin secretion in the lactating (= milk-secreting) female rats.



   Attempts were developed to show the activity of the compounds obtainable according to the invention in suppressing the growth of adenocarcinoma in female mammals. For example, the following experiment was carried out using rats as examples of female mammals susceptible to adenocarcinoma for illustration. The determination was made as follows:
Breast tumors were induced in rats by a single oral feeding of 20 mg 7,12-dimethylbenzanthracene (DMBA). The mammary glands were palpated for tumors at weekly intervals. The rats were selected for the experiment if they had at least one measurable tumor approximately 0.5 cm in diameter.

   The animals with tumors were divided into groups of 5 and treated daily with different doses of D-2-halo-6-methyl-8-cyanomethylergoline suspended in corn oil.

The compounds obtainable according to the invention show a remarkable regression of the breast tumors when they are administered either subcutaneously or orally, as can be seen from the above experiment.



   The compounds obtainable according to the invention also have a greatly reduced toxicity compared with the known compounds in which the 2-position is unsubstituted. For example show

   The following example explains the invention without restricting it.



   Example: Preparation of D-2-bromo-6-methyl-8-cyanomethylergoline.



   A solution of 955 mg of D-6-methyl-8-bromomethylergoline in 50 ml of dioxane was heated to 60 to 650 ° C. under a nitrogen atmosphere. A solution of 600 mg of N-bromosuccinimide in 70 ml of dioxane was added dropwise. The reaction mixture was heated to 60 ° to 65 ° C. for a further 1/2 hour, after which time all of the N-bromosuccinimide had been added. The reaction mixture was cooled and aqueous tartaric acid

   was added, whereby water-soluble tartrate salts of the ergoline bases present were formed. The resulting mixture was extracted with chloroform and the chloroform layer was discarded. The aqueous layer was filtered and then solid sodium bicarbonate was added until the solution became basic.

   The ergoline bases were insoluble in the basic reaction mixture. The aqueous layer was extracted with chloroform and the chloroform layer was separated and dried and the chloroform was removed by evaporation in vacuo. Chromatography of the resulting residue over Florisil using chloroform as the eluent gave fractions which, as shown by thin layer chromatography, mainly contained a material different from the starting material. These fractions were combined, the solvent was removed by evaporation in vacuo and the resulting residue was recrystallized from ether to give the D-2-bromo-6-methyl-8-bromomethylergoline formed in the above reaction. The compound melted at about 2150C (decomp.).

   90 mg of D-2-bromo-6-methyl-8-bromomethylergoline, prepared as described above, are dissolved in 10 ml of dimethyl sulfoxide (DMSO). 100 mg of sodium cyanide are added and the resulting mixture is heated to 1050 ° C. under a nitrogen atmosphere for about 45 minutes. The reaction mixture is cooled, diluted with water, and filtered. The filter cake is dissolved in a solvent mixture of ethanol / chloroform and the solvents are removed by evaporation in vacuo. Recrystallization of the residue from ether gives the D-2-bromo-6-methyl-S-cyanomethylergoline, which was formed in the above reaction. The compound melts at about 240 to 243 C (decomp.).



   According to the process described above, the D-6-methyl-8-mesyloxymethylergoline (prepared according to b)) can be halogenated in the 2-position according to the process according to a), the D-2-chloro-6-methyl-S -mesyloxymethylergoline, which contains D-2-bromo-6-methyl-8-mesyloxymethylergoline or D-2-I od-6-methyl-S-mesyloxymethylergoline.



   The 8-tosyloxymethyl derivatives, which are prepared when the methanesulfonyl chloride in the process according to b) is replaced by p-toluenesulfonyl chloride, can be halogenated in the 2-position.



   Production of the salts:
A solution containing 560 mg of D-2-bromo-6-methyl-8-cyanomethylergoline in about 40 ml of tetrahydrofuran (THF) is prepared. About 10 ml of a solution which is prepared by dissolving 1 g of maleic acid in 50 ml of THF are added to the solution of the ergoline base with stirring. About 200 ml of ether are added and the resulting precipitate is separated off by filtration. The acidic D-2-bromo-6-methyl-S-cyanomethylergoline maleate produced in this way melts at about 207 to 209 ° C. (decomp.).



   Acid D-2-chloro-6-methyl-8-cyanomethylergoline maleate, which melts at about 204 to 206 ° C. with decomposition, is prepared according to the above process.



   According to the above procedure, 320 mg of D-2-chloro-6-methyl-S-cyanomethylergoline are dissolved in 15 ml of THF. A solution of methanesulfonic acid in THF is added to this solution, 1 drop at a time, until the addition of a further drop results in no further precipitate. The THF solution is diluted with ether and the resulting mixture is filtered, the methanesulfonic acid salt of D-2-chloro-6-methyl-8-eyanomethylergoline being obtained, which decomposes at about 2950C after recrystallization from a solvent mixture of ethanol / ether melts.

Evaporation of the chloroform in vacuo gave a residue of D-6-methyl-8-bromomethylergoline which was formed in the above reaction.

   A suspension of 10 g of D-6-methyl-8-hydroxymethylergoline in 200 ml of pyridine was prepared. A solution which contained 6.0 ml of methanesulfonyl chloride and 200 ml of pyridine was slowly added to this suspension. The resulting mixture was stirred at room temperature under a nitrogen atmosphere for about 1/2 hour and then poured into 2.51 saturated aqueous sodium bicarbonate solution. The alkaline aqueous

Recrystallization from ethanol of the D-6-methyl-8-mesyloxymethylergoline, prepared as described above, gave material with a m.p.
