Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer 1, 4-Benzodiazepine der allgemeinen Formel

in denen R1 ein Fluor-, Chlor-, Brom-, Jodatom oder eine Nitrogruppe, R2 ein Wasserstoffatom, einen gegebenenfalls durch 3 Halogenatome substituierten niederen Alkylrest oder eine Dialkylaminoalkylgruppe, Rg ein Wasserstoff-, Fluor-, Chlor- oder Bromatom, R. ein Wasserstoffatom, einen niederen aliphatischen Acyl-oder niederen Alkylrest und die beiden Reste R 5 niedere Alkylreste bedeuten, und deren physiologisch verträglicher Säureadditionssalze mit anorgani- schen oder organischen Säuren und deren Salze mit Alkalibasen, falls R ein Wasserstoffatom darstellt.
Die Verbindungen der obigen allgemeinen Formeln (I) und (Ia) besitzen wertvolle pharmakologische Eigenschaften, insbesondere eine sedative, tranquillisierende, muskelrelaxierende und antikonvulsive Wirkung.
Ausserdem stellen sie neue Zwischenprodukte zur Synthese von weiteren pharmakologisch wirksamen 1, 4-Benzodiazepinen dar, welche insbesondere sedative, tranquillisierende, muskelrelaxierende und antikonvulsi- ve Eigenschaften aufweisen.
Die neuen Verbindungen lassen sich nach folgendem Verfahren herstellen :

B. bei Temperaturen zwischen 100 und 200 C, in eine Verbindung der allgemeinen Formel (I), in der R4 den entsprechenden niederen Alkylrest dar- stellt, übergeführt werden kann.
Die Überführung einer Verbindung der allgemeinen Formel (1) in ihr Derivat der allgemeinen Formel (Ia) erfolgt zweckmässigerweise in einem inerten Lösungsmittel, z. B. inTetrahydrofuran oder Dimethylformamid, vorzugsweise jedoch in einem Überschuss des verwendeten Orthoesters der allgemeinen Formel (III), in Ge- genwart des entsprechenden Alkohols und von Ammoniumnitrat und bei Temperaturen zwischen 0 und 1500C. vorzugsweise jedoch beim Siedepunkt des verwendeten Orthoameisensäureesters.
Die so erhaltenen Verbindungen der allgemeinen Formeln (1) und (Ia) können in ihre physiologisch ver- träglichen Salze mit den wasserfreien Lösungen anorganischer oder organischer Säuren, z. B. in Tetrahydrofuran, Äther, Dioxan oder Äthanol, übergeführt werden. Als Säuren kommen beispielsweise Salzsäure, Brom- wasserstoffsäure, Schwefelsäure, Phosphorsäure, Milchsäure, Weinsäure oder Maleinsäure in Betracht.
Die als Ausgangsstoffe verwendeten Verbindungen der allgemeinen Formel (II) sind Gegenstand der deutschen Offenlegungsschrift 2234150 und lassen sich nach den dort beschriebenen Verfahren herstellen.
Wie bereits eingangs erwähnt, weisen die neuen Verbindungen der allgemeinen Formeln (I) und (Ia) wertvolle pharmakologische Eigenschaften auf, insbesondere eine sedative, tranquillisierende, muskelrelaxierende und antikonvulsive Wirkung. Ausserdem stellen sie neue Zwischenprodukte zur Herstellung von weiteren pharmakologisch wirksamen 1, 4-Benzodiazepinen dar, welche insbesondere sedative, tranquillisierende, muskelrelaxierende und antikonvulsive Eigenschaften aufweisen.
Beispielsweise wurden folgende Substanzen auf ihre biologischen Wirkungen untersucht : A = Natriumsalz des 7-Chlor-5- (2-chlor-phenyl)-1, 3-dihydro-3-hydroxymethylen-l-methyl-2H-l, 4-ben- zodiazepin-2-ons, B = 7-Brom-5- (2-chlor-phenyl)-1, 3-dihydro-3-hydroxymethylen-l-methyl-2H-l, 4-benzodiazepin-2-on- hydrochlorid und C = Natriumsalz des 7-Brom-l, 3-dihydro-5- (2-fluor-phenyl)-3-hydroxymethylen-l-methyl-2H-l, 4-ben- zodiazepin-2-ons.
1. Muskelrelaxierende und sedierende Wirkung bei Mäusen :
Diese wurde nach der Methode vonYoung und Lewis (Seience Bd. 105 [1947], S. 368) an weiblichen NMRIMäusen eigener Zucht mit einem Körpergewicht zwischen 20 und 26 g mittels sich langsam drehender, um 300 gegen die Vertikale geneigter Drahtzylinder (Länge : 43 cm ; Durchmesser : 22 cm ; Maschenweite des Drahtgeflechtes : 0, 6 cm) untersucht. Nach peroraler Applikation der zu prüfenden Substanz in 1%piger Tylosesuspension an Gruppen von 10 Mäusen/Dosis wurde deren Haltefähigkeit in den langsam rotierenden Zylindern (2 Drehungen/min) gegenüber einer Kontrollgruppe geprüft.
Es wurde die Dosis (ED50) graphisch ermittelt, bei welcher nach den verschiedenen Zeiten 50% der Tiere herausgefallen waren :

2. Anitkonvulsive Wirkung bei Mäusen :
Die antikonvulsive Wirkung wurde als Schutzwirkung gegen den maximalen Elektroschock-Krampf bei männlichen NMRI-Mäusen eigener Zucht mit einem Körpergewicht zwischen 20 und 26 g in Anlehnung andie Methode von Swinyard, Brown and Goodman (J. Pharmacol. exp. Therap. Bd. 106 [1952], S. 319) untersucht.
Hiebei wurden die Tiere einem Wechselstrom von 50 Hz und 50 mA bei einer Reizdauer von 0, 2 sec ausgesetzt, wobei das Auftreten des tonischen Streckkrampfes als positiv gewertet wurde. Nach peroraler Applikation der zu prüfenden Substanzen in 1%piger Tylosesuspension wurde die Dosis (ED50) graphisch ermittelt, bei welcher nach verschiedenen Zeiten 50% der Tiere gegen die tonische Extensorkomponente der Hinterex- tremitäten beim Krampf geschützt waren :

Ergänzend sei hier noch erwähnt, dass die Substanzen praktisch untoxisch sind ; so konnten beispielsweise bei allen Dosierungen keinerlei toxische Nebenwirkungen beobachtet werden.
Die neuen Verbindungen der allgemeinen Formeln (1) und (Ia) lassen sich zur pharmazeutischen Anwendung in die üblichen pharmazeutischen Zubereitungsformen, wie Tabletten, Dragees, Kapseln und Suppositorien, gegebenenfalls in Kombination mit andern Wirksubstanzen, einarbeiten. Die Einzeldosis beträgt hiebei 2 bis 20 mg, vorzugsweise 5 bis 10 mg, und die Tagesdosis 5 bis 80 mg, vorzugsweise 10 bis 40 mg.

Wasser 5 min lang bei etwa 20 C intensiv geschüttelt, wobei alles in Lösung geht. Man versetzt mit 700 ml Äther, trennt die Phasen und zieht die wässerige Phase nochmals mit Äther aus. Die vereinigten organischen Extrakte werden mit Wasser gewaschen und mit Magnesiumsulfat getrocknet.
Auf Zugabe von ätherischer Salzsäure kristallisiert die oben genannte Verbindung aus, die nach Umkristallisieren aus Tetrahydrofuran/Äther bei 185 bis 187 C (Zers. ) schmilzt.

4-benzodiazepin-2-on,diazepin-2-on 10 g 7-Chlor-5- (2-chlor-phenyl)-l, 3-dihydro-3-hydroxymethylen-l-methyl-2H-l, 4-benzodiazepin'-2- on Natrium-Salz werden in 11 Wasser gelöst. Unter Rühren gibt man hiezu tropfenweise so lange Essigsäu- re, bis ein PH = 5 erreicht ist. Der entstandene blassgelbe Niederschlag wird abgesaugt, gut mit Wasser und anschliessend mit Petroläther gewaschen und an der Luft getrocknet.

Beispiel 3 : 3-Acetoxymethylen-7-brom-5- (2-chlor-phenyl)-1, 3-dihydro-l-methyl-2H-l, 4-benzo- diazepin-2-on

Beispiel 4 : 3-Äthoxymethylen-7-brom-5- (2-chlor-phenyl)-1, 3-dihydro-l-methyl-2H-1, 4-benzo- diazepin-2-on

Schmelze wird zwischen Äther und Wasser verteilt. Die ätherische Phase wird mit Wasser gewaschen, getrocknet und im Vakuum eingedampft. Nach Kristallisation und Umkristallisation mit Äther schmelzen die erhaltenen Kristalle bei 175 bis 1760C.

5, 5 g 7-Brom-5- (2-chlor-phenyl)-1, 3-dihydro-3- (dimethylaminomethylen)-1-methyl-2H-l, 4-benzo- diazepin-2-on werden mit 4, 9 g Oxalsäure-dihydrat, wie im Beispiel 1 beschrieben, behandelt.
Die erhaltene, getrocknete ätherische Lösung der 3-Hydroxymethylen-Verbindung wird im Vakuum eingedampft und mit 2, 15 ml Orthoameisensäure-triäthylester, 0, 8 ml absolutem Äthanol und 0,06 g Ammoniumnitrat versetzt.
Man rührt über Nacht bei Raumtemperatur, fügt nochmals die gleichen Mengen der oben genannten Reagenzien hinzu und erhitzt 10 min zum Sieden. Anschliessend entfernt man im Vakuum alle flüchtigen Anteile und reinigt den Rückstand über eine Kieselgel-Säule mit Chloroform : Methanol = 30 : 1 als Elutionsmittel. Die Fraktionen mit der gewünschten Substanz werden vereinigt und im Vakuum vom Lösungsmittel befreit. Der Rückstand wird aus Äther kristallisiert und noch einmal umkristallisiert.
Schmelzpunkt : 122 bis 124 C.

1. Muscle relaxing and sedating effects in mice:
This was done according to the method of Young and Lewis (Seience Vol. 105 [1947], p. 368) on female NMRI mice of our own breeding with a body weight between 20 and 26 g by means of slowly rotating wire cylinders inclined by 300 to the vertical (length: 43 cm; diameter: 22 cm; mesh size of the wire mesh: 0.6 cm). 2. Anti-convulsive effects in mice:
The anticonvulsant effect was used as a protective effect against the maximum electric shock convulsions in male NMRI mice of our own breeding with a body weight between 20 and 26 g based on the method of Swinyard, Brown and Goodman (J. Pharmacol. Exp. Therap. Vol. 106 [ 1952], p. 319).
