   Das in der AT-PS Nr. 216666 angegebene Verfahren zur Herstellung von therapeutisch wirksamen Präparaten aus Humanplazenta wurde, wie folgt, erweitert :
Durch mehrfaches Einfrieren und Auftauen eines filtrierten Extraktes aus humanen, gesunden Gesamtplazenten, durch Kryosedimentation, konnte ein Kjeldall-negativer, ein wesentlich eiweissfreier niedermolekularer Extrakt gewonnen werden, der in Spuren das humane Aminosäurespektrum, Progesteron und in geringster Menge Östriol enthielt. (Prof. Dr. W. Stöckl, Institut für Biochemie der Tierärztlichen Hochschule Wien.) Weiters wurden nach   UV-Aufnahmen   (Prof. Dr. G. Kellner, Histolog. Univ. Institut, Wien) Nukleinsäuren und nach neuesten Untersuchungen auch energiereiches Phosphate-Adenosintriphosphat als mögliche therapeutische Faktoren zur Diskussion gestellt.

In eigenen Versuchen konnte eine Beeinflussung viraler Infektionen (Mäuseversuch mit Semliki-Forest-Viren, Prof. Dr. Ch. Kunz, Virologische Lehrkanzel der Univ. Wien), sowie virusbedingter Mäuse-Mamma-Tumoren (Bundesstaatliche Impfstoffgewinnungsanstalt Wien) beobachtet werden. Die Manipulierbarkeit des Auftretens der Pockeneffloreszenzen und die verstärkte Borkenbildung nach zusätzlicher Therapie mit dem Extrakt am Modell des   Kaninchen-Pocken-Versuches   lässt nach Prof. Dr. H. Stickl, Leiter der Bayerischen Landesimpfanstalt, München, eine therapeutisch verstärkte Interferon-Induktion vermuten. (Lit. : Ackermann, G. : Induktion der körpereigenen Abwehr beim Krebs ? Der therapeutische Einsatz kennt kaum Kontraindikationen, er wird weniger durch den Charakter der Erkrankung, als vielmehr durch den Zustand des kranken Organismus und seiner Abwehrlage, im Einklang mit dem System der Grundregulation nach Pischinger bestimmt. 



   Das Präparat kann mit allen andern Heilmethoden ohne Schwierigkeiten kombiniert werden. 



  Das erfindungsgemäss hergestellte Präparat kann sowohl als flüssiger Extrakt (im Gefrierschrank   bei-12 C   aufzubewahren) oder als Lyophilisat in Ampullen parenteral, oral, lokal, vaginal und rektal verabreicht werden. Während der native, flüssige Frischextrakt bei Gefrierhaltung bis zu sechs Wochen unbedingt wirksam bleibt, kann der sterile, lyophilisierte Trockenextrakt in Ampullenform jahrelang verwendet werden. Bisher kontrollierte Verwendbarkeit : 10 Jahre, er benötigt keine Gefrierhaltung und ist gegen jede Aussentemperatur beständig. 



   Nähere Beschreibung des erfindungsgemässen Verfahrens :
Humane Gesamtplazenten werden innerhalb von ungefähr 1 bis 2 h nach der Gewinnung der gesunden, reifen Nachgeburt, abhängig von der herrschenden Aussentemperatur und Jahreszeit, in sterile Tücher und sterile Gefässe verpackt, in den Kühlschrank gebracht und dort bei einer

Das homogene Gemisch von Plazentabrei und Lösungsmittel wird durch entsprechende Filtration (von Papierfiltern bis zu Seitzfiltern je nach Bedarf reichend) von korpuskulären Gewebeteilchen befreit und zusätzlich werden durch Anwendung von technischen Saugwirkungen (etwa Wasserstrahlpumpe usw. ) plazentare, gewebliche Inhaltsstoffe gewonnen, die in den flüssigen Extrakt gelangen.

   Dieser wird nun tiefgefroren (maximal-12 G) und nach einem oder nach zwei Tagen der Kryosedimentation unterworfen. Das Verfahren des Einfrierens und des allmählichen Wieder-Auftauens des Rohextraktes führt allem Anschein nach zur Entstehung oder zur Induktion biologisch aktiver Substanzen, die teilweise heute unter verschiedenen Namen bekannt sind. Beim Auftauen des Extraktes wird der zuerst flüssig werdende,   rötliche-sanguinolente,   hochmolekulare Anteil in ein anderes Gefäss verbracht, so dass letztlich - nach oft tagelang dauernder   Tätigkeit - ein   fast   wasserklarer   Extrakt als niedermolekularer Plazentaextrakt resultiert.

 anfänglich verflüssigte Teil wird wieder eingefroren und nach etwa 24 h neuerlich teilweise wieder aufgetaut, der flüssige Anteil wieder eingefroren usf. bis letztlich der beschriebene Extrakt entsteht. 



   Die ganze Präparation und die sich immer wiederholende Umschichtung des teilweise aufgetauten Extraktes in ein anderes, steriles Gefäss geschieht unter ständiger Ultraviolettbestrahlung. 



   Die Gewinnung von haltbaren Präparaten erfolgt durch Lyophilisation nach den üblichen Methoden und Aufbewahrung in sterilen Ampullen, deren Haltbarkeit bis zu 10 Jahre beträgt. 



   Die Keimfreimachung und die bakteriologischen Kontrollen geschehen nach den für biologische Präparate üblichen Regeln. 



    PATENTANSPRÜCHE :    
1. Verfahren zur Herstellung therapeutisch wirksamer Präparate aus niedermolekularem Humanplazentaextrakt, dadurch gekennzeichnet, dass man reife, frische und gesunde Gesamtplazenten nach einer etwa 20 min dauernden Aufbewahrung bei Temperaturen unter   0 C   zunächst durch heisses Wasser von maximal 570C grob reinigt, die obersten Begrenzungsschichten entfernt und das Innere der Plazenten unter möglichst wenig   Druck-und Zuganwendung   fein zerkleinert und den Brei mit sterilem Aqua dest., und/oder physiologischer Ringerlösung und/oder keimfreiem, 2wertige Metallionen enthaltendem Mineralwasser homogenisiert, die entstehende Suspension filtriert und einfriert,

   worauf der hochmolekulare Extrakt zur Gewinnung eines niedermolekularen Extraktes mit properdin- ähnlicher unter interferoninduzierender Wirkung einer Kryosedimentation unterzogen wird und worauf man langsam auftaut und die zuerst sich verflüssigenden hochmolekularen und   korpuskulären   Teile steril absaugt und wieder einfriert, von restlichen hochmolekularen Teilen befreit und man so eine niedermolekulare, Kjeldahl-negative Lösung erhält, die unter anderem auch in Spuren alle im menschlichen Blut vorkommenden freien Aminosäuren enthält.
