Beschreibung
Die Erfindung betrifft neue Di- und Trisaccarid-Derivate des D-Glucosamin, deren besondere Merkmale in da1 3-Subsütution, der 6-Substitution oder der 3,6-Disubstitution mit α-L-Fucosylresten besteht
Die a-1-3* und die a-1 -6-Fucosylierung von N- Acetylglucosamin sind weit verbreitete Charakteristika natürlich vorkommender Glykoproteine (E. Staudacher etal. (1991) Eur. J. Biochem. 199.745-751). Die chemische Synthese von Teilstrukturen, die diese Merkmale enthalten, bildet ein Weg, die Beziehung zwischen Struktur und biologischem Effekt, zum Beispiel Immunogenität, zu erforschen. Aus eigenen Arbeiten geht hervor, daß Insekten-Glykoproteine beide Bindungen der Fucose an denselben N-Acetylglucosaminrest, entweder alternativ oder kumulativ, aufweisen können (E. Staudacher et al. (1992) Glycoconj. J., im Druck) und daß durch diese Substitutionsmuster immunogene Gruppierungen entstehen (Ch. Prenner et al. (1992) Biochem. J., im Druck).
Es ist daher von Interesse, D-Glucosamin-Derivate mit den genannten Strukturmerkmalen in einer reaktiven Form darzustellen, um sie mit geeigneten Makromolekülen, zum Beispiel Proteinen, zu Immunogenen und anderen anwendbaren Verbindungen konjugieren zu können. Die Errungenschaft der neuen D-Glucosamin-Derivate liegt darin, daß sie einen Zugang zur biologischen Signifikanz eines neu entdeckten natürlich vorkommenden Strukturcharakteristikums ermöglichen.
Die ist deswegen von Bedeutung, weil Insektenzellen für die rekombinante Expression von heterologen Proteinen und Glykoproteinen verwendet werden; das Auftreten insektenspezifischer Strukturmerkmale kann bei der Administration im Wirbeltierorganismus zu Effekten führen, die einer Anwendung entgegenstehen; der Entwicklung von Nachweis- und Analysendiagnostika kommt dabei besondere Bedeutung zu.
Die besonderen Vorteile der neuen D-Glucosamin-Derivate liegen darin, daß sie 1. Teilstrukturen neu entdeckter natürlich vorkommender immunogener Kohlenhydrat-Seitenketten biologisch aktiver Glykoproteine darstellen, 2. den Zugang zur Aufklärung von Struktur-Wirkungsbeziehungen eröffnen, 3. aufgrund ihrer reaktiven Gruppe zur Herstellung von Konjugaten für die Anwendung als Immunogene, Antigene, Inhibitoren, an Chromatographie-Matrices immobilisierbare Affinitätsreaktanten und als Enzymsubstrate geeignet sind.
Beispiel 1 3,4-0-Isopropyliden-2-0-(4-methoxybenzyl)-a-L-fucopyranosyl-(l -> 6)-0-allyl 2-acetamido-2-desoxy-3,4- 0-(l,U,3,-tetraisopropyldisiloxan-13-diyl)-ß-ß-glucopyranosid
Eine Suspension von S-Ethyl 3,4-0-isopropyliden-2-0-(4-methoxybenzyl)-ß-L-fucopyranosid (350 mg), 0-Allyl2-acetamido-2-desoxy-3,4-0-(1,133,-tetraisopropyldisiloxan-13*diyl)-ß-D-glucopyranosid(330mg)und Molekularsieb4A(3g)m 12mlDiethylether/Mchlormethan5:l wurde während6hmitIodonium-di-symCollidinium-perchlorat (520mg) versetzt und 15 h bei Raumtemperatur gerührt. Beispiel 2 3,4-0-Isopropyliden-2-0-(4-methoxybenzyl)-a-ii-fucopyranosyl-(l-6)-0-allyl 2-acetamido-2-desoxy-ß-D-glucopyranosid
EineLösungvon3,4-0-Isopropyliden-2O-(4-meÜtoxybenzyl)-a-L-fucopyranosyl-(l->6)-0-allyl2-acetamido-2-desoxy-3,4-0-(1,l,33.-tetraisopropyldisiloxan-l,3-diyl)-ß-E)-glucopyranosid (87 mg) in 4 ml THF wurde mit 0,1 ml einer 1,1 molaren Lösung von Tetrabutylammoniumfluorid in THF versetzt Nach 4 h Rühren bei Raumtemperatur wurde eingeengt und über Kieselgel mitEthylacetat/Methanol 5:1 chromatographisch gereinigt. Beispiel 4 a-L-Fucopyranosyl-(l -> 6)-0-allyl 2-acetamido-2-desoxy-ß-D-glucopyranosid
Eine Lösung von 16 mg 2,3,4-Tri-0-acetyl-a-L-fucopyranosyl(l -> 6)-0-allyl 2-acetamido-3,4-di-0-acetyl-2-desoxy-ß-D-glucopyranosid in 5 ml absolutem Methanol wurde mit 0,5 ml einer 0,1 M methanolischen Natriummethoxid-Lösung versetzt und 3 h bei Raumtemperatur gerührt. Nach Zusatz von DOWEX-50 (H^-Form) bis zu neutralem pH wurde abfiltriert, eingedampft, in Wasser aufgenommen und lyophilisiert. Man erhält so 10 mg Produkt als amorphes Pulver.

Beispiel 5 O-Allyl 2-acetamido-2-desoxy-3,6-di-0-(3,4-0-isopropyliden-2-4-methoxybenzyl-a-L-fucopyranosyl)-ß-ß-35 glucopyranosid 50mg3,4-O-Isopropyliden-2-O-(4-methoxybenzyl)-a-L-fiicopyranosyl-(l->6)-O-allyl2-acetamido-2-desoxy-ß-D-glucopyranosid, 37 mg S-Ethyl 3,4-0-isopropyliden-2-0-(4-methoxybenzyl)-ß-L-fucopyranosid wanden in 9 ml Diethylether/Dichlormethan 1:1 gelöst und mit 2 g Molekularsieb 4A versetzt Während 3 h werden 60 mg Iodonium-di-collidinium-perchlorat portionsweise zugesetzt und weitere 3 h bei Raumtemperatur gerührt. Nach 40 Verdünnen mit 50 ml Dichlormethan wird über Celite abfiltriert, mit 5%iger Natriumthiosulfat-Lösung ausgeschüttelt und nach Trocknen über Natriumsulfat eingedampft. Reinigung an Kieselgel mit Ethylacetat gibt 23 mg Produkt als Sirup.

Beispiel 6 50 O-Allyl 2-acetamido-4-0-acetyl-3,6-di-0-(2,3,4-tri-0-acetyl-a-L-fucopyranosyl)-2-desoxy-ß-D-glucopyranosid 17mgO-Allyl2-acetamido-2-desoxy-3,6-di-0-(3,4-0-isopropyliden-2-[4-methoxybenzyl]-a-L-fucopyranosyl)-ß-ß-glucopyranosid wurde in 4 ml Dichlormethan/Wasser 18:1 gelöst, mit 16 mg 2,3-dicyano-5,6-dichloro-benzochinon versetzt und 24 h bei Raumtemperatur gerührt. Die Lösung wurde eingedampft, getrocknet, in 2 ml Pyridin aufgenommen und mit 0,3 ml Acetanhydrid versetzt. Nach 6 h Rühren bei Raumtemperatur wurde eingeengt 55 und über Kieselgel mit Hexan/Ethanol 3:1 gereinigt Man erhält so 12 mg Produkt als farblose Kristalle.

Eine Lösung von 7 mg O-Allyl 2-acetamido4-0-acetyl-3,6-di-0-(23,4-tri-0-acetyl-a-L-fucopyranosyl)-2-desoxy-ß-ß-glucopyranosid in 1,5 ml Methanol wurde mit 0,2 ml einer 0,1 M methanolischen Natrium-methanolat Lösung versetzt und 5 h bei Raumtemperatur gerührt Die Lösung wurde mit DOWEX 50 (H*-Form) neutralisiert, abfiltriert und eingedampft. Der Rückstand wird in 2 ml Wasser aufgenommen und lyophilisiert. Man erhält so 4,5 mg Produkt als amorphes Pulver.

Beispiel 8 a-L-FucopyranosyHl -> 6)-0-3-(2-aminoethylthio)propyl 2-acetamido-2-desoxy-ß-I)-glucopyranosid Eine Lösung von 5 mg a-L-Fucopyranosyl-(l -> 6)-0-allyl 2-acetamido-2-desoxy-ß-I)-glucopyranosid und 6,5 mg cysteamin-Hydrochlorid in 2 ml Wasser wird in einer Quarzküvette 5 h bei254nm bestrahlt. Die Lösung wird über Sephadex G-25 entsalzt und gefriergetrocknet Man erhält so 4,5 mg Produkt als amorphes Pulver.

Konjugation mit Rinder-Serumalbumin (BSA)
Eine Lösung von 2 mg a-L-Fucopyranosyl-(l -> 6)-0-3-(2-aminoethylthio)propyl 2-acetamido-2-desoxy-ß-ß-glucopyranosid in 1 ml 0,1 M wäßrige NaHCO^-Lösung wird zu ein»- Lösung von 2 μΐ Thiophosgen in 1 ml Chloroform zugesetzt und 4 h bei Raumtemperatur kräftig gerührt. Die organische Phase wird abpipettiert, und die wäßrige Phase wird zweimal mit je 2 ml Chloroform extrahiert Nach Durchleiten von Stickstoff wird die wäßrige Phase zu einer Lösung von 4,0 mg BSA in 0,1 M NaHCOß/O.S M NaCl (2 ml) zugegeben und 48 h bei Raumtemperatur gerührt Die Lösung wird über Sephadex G-25 mit 0,01 M NäHCOj gereinigt Die Ninhydrin-positiven Fraktionen werden gepoolt und gegen Wasser dialysiert. Man erhält so 33 mg BSA-Konjugat als weißes amorphes Pulver mit einer Ligandbelegung von 3 - 4 mol Oligosaccharid pro mol BSA. -4-
Claims (10)
- AT 396 474 B PATENTANSPRÜCHE 1. Neue D-Glucosaminderivate der allgemeinen Formel I und Πwobei R für Alkenyl, Aminoalkyl und R* für Wasserstoff und Acyl steht.
- 2. Neue D-Glucosaminderivate der Formel ΠΙ
- 3. Verfahren zur Herstellung der neuen D-Glucosaminderivate der allgemeinen Formel I und Π, dadurch gekennzeichnet, daß man an sich bekannte D-Glucosaminderivate der allgemeinen Formel IVwobei Rjj für Alkyl, Alkenyl, Aryl, R* für Acyl, R2 und für eine bifunktionelle Schutzgruppe Alkylsilyl, Alkyliden, Aryliden und R^ für Wasserstoff oder unabhängig davon R^ und R^ für eine bifunktionelle Schutzgruppe 20 Alkylsilyl, Alkyliden, Aryliden steht, in einem inerten Lösungsmittel wie Halogenkohlenwasserstoffen, Ether, Aromatischen Kohlenwasserstoffen, N,Ν-Dimethy lformamid unter Katalyse mit Schwermetallsalzen wie Silbersalzen, Quecksilbersalzen, Kupfersalzen oder thiophilen Reagenzien wie Iodoniumdicollidiniumperchlorat, Methyltriflat, Dimethyl(methylthio)sulfoniumtriflat mit einem an sich bekannten L-Fucosederivat der allgemeinen Formel V 2530 wobei Rc für Halogen, Thioalkyl, Trichloracetimidat, R2 für Arylmethyl, R^ und R^ für Alkyl, Aryl, Acyl, Alkyliden, Aryliden steht, zur Reaktion bringt, chromatographisch reinigt, und die Schutzgruppen nach bekannten Verfahren entfernt
- 4. Verfahren zur Herstellung da* neuen D-Glucosaminderivate der allgemeinen Formel ΠΙ, dadurch gekennzeichnet, daß man D-Glucosaminderivate der allgemeinen Formeln VI und VII
- 5. Verwendung der neuen D-Glucosaminderivate der Formel I, Π und m als Liganden zur Herstellung von Glykokonjugaten nach bekannten Verfahren, insbesondere mit Enzymen und Antikörpern, mit Avidin und Biotin, sowie Fluorochromen.-5- III AT396474B wobei R& für Wasserstoff, Alkyl, Alkenyl, Aminoalkyl und R* für Wasserstoff oder Acyl steht

- 7. Verwendung der neuen D-Glucosaminderivate der Formel I, Π und III als Antigene für den Nachweis und die Bestimmung der Antikörper nach Anspruch 6.
- 8. Verwendung der neuen D-Glucosaminderivate der Formel I, Π und m als Inhibitoren von Antigen-Antikörper-Reaktionen im Rahmen von Testsystemen für Allergien.
- 9. Verwendung der neuen D-Glucosaminderivate der Formel I, II und ΙΠ als Ausgangsmaterial zur Herstellung von Affinitätschromatographiemedien für die Reinigung von Enzymen sowie poly- und monoklonalen Antikörpern durch Kopplung der endständigen Aminogruppe nach bekannten Verfahren an controlled-pore-glass, Silikagel, Agarose, Dextran, Polyacrylamid oder andere makromolekulare Träger.
- 10. Verwendung der neuen D-Glucosaminderivate der Formel I, II und III als Ausgangsmaterial für Substrate zur Bestimmung von spezifischen Enzymen, insbesondere Fucosidasen undFucosyltransferasen. -7-
