Abstract
A novel plasmid vector for expression in Bacillus and used for cloning the structural gene which codes for the human growth hormone (hGH) is obtd. by substituting the 750 bp XbaI-ECORI fragment of plasmid pSM143 ATCC 53038 by a synthetic 73 bp fragment having a specified nucleotide sequence. The cloning vector is constructed from pSM143 (ATCC53038) described in IT 19960A/85. The 73 bp fragment was constructed so as to obtain a promoter sequence and a ribosome recognition sequence, both sequences recognised by the RNA polymerase and Bacillus ribosomes and ensuring efficient expression of the structural gene of the hGH placed under control of the sequences.
