"Procédé pour la séparation dea hormones de l'hypophyse".
Il est connu qu'on récupère de l'hypophyse un très grand nombre d'hormones, et tout aussi bien du lobe antérieur que du lobe postérieur de cette glande. La séparation de ces deux parties l'une de l'autre est cependant pratiquement irréalisable ou toutefois difficilement réalisable en raison des petites dimensions de cet organe, particulièrement pour les glandes du mouton, du porc ou d'animaux encore plus petits. On prépare de ce fait pour des buts techniques une poudre sèche à partir de la glande complète de ces animaux, dont on isole ensuite, suivant des procédés différents, des hormones séparées plus ou moins épurées. Il se présente ici des difficultés particulières pour la récupération des hormones du lobe avant, dont quelques-unes sont facilement

détruites lors de l'emploi d'agents d'extraction chaude.
En outre, jusqu'à présent, lors de l'utilisation de la glande complète, il n'est pas possible, dans aucun procédé connu, d'obtenir des hormones du lobe antérieur qui sont libérées des hormones du lobe postérieur qui ont une activité sur la pression sanguine et sur l'utérus. De faibles quantités de ces hormones du lobe postérieur se trouvent mélangées aux préparations connues jusqu'à, présent des hormones thyréotropes, gonadotropes et corticotropea et dans la prolactine.
Pareilles mixtures additionnelles d'hormones du lobe postérieur de l'hypophyse influencent cependant l'utilisation thérapeutique des hormonesdu lobe antérieur, étant donné qu'elles provoquent des activités latérales indérisables et nuisibles, soit par accroissement de la pression sanguine, provocation d'attaques de vertige et contractions utérines, etc.
Il a maintenant été trouvé que l'on peut également arriver, en utilisant l'hypophyse complète comme matière de départ, à des préparations d'hormones du lobe antérieur, qui sont libérées des effets des hormones du lobe postérieur, quand on soumet les glandes à. un traitement par des solvente organiques appropriés aqueux, miscibles avec de l'eau et en présence d'une réaction alcaline. On extrait ainsi unique- ment des hormones du lobe postérieur et l'on peut en récupérer supplémentairement de cet extrait, cependant que du résidu d'extraction on peut isoler les hormones du lobe antérieur suivant une méthode connue. Le procédé est applicable aussi bien sur des hypophyses complètes fraîches de tous les ani- maux que sur la poudre sèche-acétone usuelle ou encore sur de la matière dégraissée.
Le procédé est d'autre part applicable sur des pré- parationsd'hormones du lobe antérieur,qui ont été récupérées de la glande complète suivant des méthodes connues et qui contiennent encore de ce fait des hormones du lobe postérieur.

Lors de l'utilisation de la poudre d'acétone sèche comme matière de départ, celle-ci est extraite jusqu'à trois fois à un Ph 9-10 avec vingt fois la quantité de méthanol à 80% ou d'un autre solvent organique aqueux, miscible avec de l'eau, le traitement se poursuivant comme ci-devant, lors de la mise en pratique de l'épuration, conformément à l'invention, il convient de veiller que la quantité de la base soit déterminée, de telle façon que la solution à précipiter présente de préférence un Ph entre 8 et 11 environ; cependant un Ph audessus de 11 ne gène pas inconditionnèllement mais donne cependant peu de produits purs et des rendements plus mauvais.

voulu peut également ëtre réglé par addition de bases or.. ganique appropriées, aliphatiques ou cycliques, comme le triéthanolamine, la pyridine, etc. la quantité de liquidesorganiquesà ajouter en vue de la précipitation est déterminée de telle façon qu'après addition, sa concentration dans la solution à précipiter comporte environ 60-90%, de préférence 80%. La concentration optima du liquide de précipitation et le coefficient Ph optima lors de la précipitation seront évidemment dépendants de la nature du liquide organique employé et de la composition de la matière de départ.
Ils peuvent d'ailleurs être facilement obtenus dans chaque cas par des expériences préliminaires effectuées par 1'homme de métier et par des contrôles biologiques. Il est évident que tale liquides organiques sont exclus de l'emploi qui ne dissolvent pas ou insuffisam- ment l'hormone du lobe postérieur, par exemple l'acétone.
Le procédé est avantageusement mis en pratique à basse température, de préférence à la température de chambre. Pour la séparation du précipité non dissous du liquide, on peut utiliser les méthodes de séparation connues telles que la filtration, la décantation, la centrifugation, etc.
Les hormones du lobe antérieur peuvent être obtenues par ce procédé, de façon presque quantitative et enrichies trois ou quatre fois.
Exemple 1:
10 kgr. de glandes totales broyées de porc sont amenée au Ph 9-la à l'aide d'ammoniaque concentré et additionné de 32 litres de méthanol: on agite pendant 12 heures et on sépare par filtration. Le résidu d'extraction est mis en pâte avec 2,4 litres d'eau, amené à nouveau avec de l'ammoniaque au Ph 9-10, additionné de 27,6 litres de méthanol et éven- tuellement agité pendant 12 heures, séparé par filtration et encore une fois traité avec la même quantité de méthanol. les extraits réunis sont neutralisés avec de l'acide acétique

et amenés à siccité.
Ion obtient environ 400 gr. de résidus secs qui possèdent l'activité complète utérine et de pression sanguine de la matière de départ, notamment environ 1/2 milligr. par unité. Ce produit peut être traité ensuite de manière connue pour récupérer les hormones du lobe postérieur* Des glandes extraites on peut par exemple préparer, suivant le procédé de la demande du brevet allemand Sch.114.736 IVc/12p, l'hormone thyréotrope et ensuite du résidu' extrait l'hormone oorticotrope,euivant la demande du brevet allemand Sch.116.310 IVc/12p.
Exemple 2:
50 gr. d'une poudre sèche à l'acétone de l'hypophyse complète du porc sont mis en pâte avec 200 cm3 d'une solution d'ammoniaque à 1% et broyés au moulin à boulets pendant 12 heures après addition de 600 cm3 de méthanol. On extrait encore deux fois le résidu d'une manière similaire avec la moitié de la quantité de l'agent d'extraction et à la dernière filtration on lave le résidu par après avec du méthanol acétique absolu.
Les extraits au méthanol, qui auront été neutralisés après chaque séparation du résidu d'extraction au moyen d'acide acétique et qui furent conservée dans la glacière, sont maintenant réunie et débarrassée de solvents dans le vide. Après séchage sous vide il reste un résidu de 14 gr, Dans l'expérience sur animaux il ne put être décelé aucun effet de l'hormone du lobe antérieur d'hypophyse, cependant il fut ici retrouvé l'activité complète des hormones du lobe postérieur contenues dans la matière de départ (hormone qui augmente la pression sanguine et hormone d'activité utérine). le résidu restant après triple extraction avec du méthanol à 80% fut également séché sous vide. Son poids comportait 40 gr.
Un extrait de cuisson,préparé de manière usuelle à partir d'un échantillon, ne montraitplus d'ac-

tivité aucune sur l'utérus isolé ,de même l'effet sur la pression sanguine était absent. Par contre, un extrait acétique récupéré de manière connue contenait la totalité de l'activité thyréotrope de la poudre à l'acétone sèche, notamment 15,000 unités-Cobaye. En outre, cet extrait ne présentait aucun effet toxique.
Exemple 3:
20 gr. d'hormone thyréotrope, d'une préparation obtenue suivant les demandes de brevet allemandes Sch.114.736 IVc/12p et Sch.114.759 IVc/12p à partir de l'hypophyse com- plète du porc, qui contient par milligramme la,7 d'unitésCobaye (U.C), au total soit 215.000 U.C., mais à côté encore 15.000 unités-Voegtlin (U.V) d'une hormone/croissant la pression sanguine et 15.000 unités. internationales (U.
I) d'hormone à activité utérine du lobe arrière, furent dissous dans 400 cm3 d'eau et rendus alcalins) en présence de phénolphtaline, avec 12 cm3 d'ammoniaque à 10%. La solution est introduite en agitant dans 1600 cm3 de méthanol refroidi à la glace et auquel on a ajouté préalablement 10 cm3 d'ammo- niaque à 10% et 1 gr. d'acétate de potassium. Le précipité qui se forme est séparé par centrifugation et on le dissout de nouveau dans 200 cm3 d'eau qui contient 4 cm3 d'ammoniaque à 10%. Cette solution est introduite en agitant dans 800 cm3 de méthanol refroidi à la glace, lequel était additionné de 5 cm3 d'ammoniaque à 10% et d'un gramme d'acétate de potassium.
Le précipité est à nouveau centrifugé immédiatement, dissous à nouveau dans 100 cm3 d'eau qui contient 3 cm3 d'ammoniaque à 10% et l'on introduit cette solution en agitant dans 400 cm3 de méthanol refroidi à la glace et qui contient 5 cm3 d'ammoniaque à la$ et 0,5 gr.d'acétate de potassium. Le précipité, séparé maintenant par centrifugation, est débar- rassé des hormones du lobe postérieur qui ont une activité sur la pression sanguine et sur l'utérus. Il est dissous

dans de l'eau additionnée d'acide chlorhydrique jusqu'à réaction acide congo et précipité avec dix fois le volume d'acétone chlorhydrique faible, Le précipité est séparé par aspiration, lavé avec de l'acétone et de l'éther et séché sous vide.
On obtient ainsi 7 gr. d'une poudre d'un blanc pur et facilement soluble dans l'eau. Selon les ré- sultats de l'essai biologique,elle est débarrassée deshor- mones du lobe postérieur qui ont une activité sur la pres- sion sanguine et sur l'utérus elle contient par milligramme 30,6 d'U.O., au total soit 213.000 U.C.
Les solutions de méthanol réunies sont neutralisées avec de l'acide acétique, le solvent est distillé sous vide à une température de bain de 35 et le résidu est séché sous vide. on obtient 22,7 gr. de substancessèches qui con- tiennent par milligramme 0,5 d'U.V. d'hormone accroissant à pression sanguine, soit au total 11.300 U.V. et en outre 11.300 U.I. de l'hormone à activité utérine.
Exemple 4.
5 kgr.d'hypophyses complètes broyées du porc ont été rendue alcalins à l'égard de phénolphtaline avec de l'ammo niaque concentré et additionnés de 13 litres d'éthanol, on a agité pendant 12 heures et séparé par aspiration. Le ré- sidu d'extraction est mis en pâte avec 2,6 litres d'eau, ensuite additionné de 13 litres d'éthanol, agité pendant 12 heures, séparé par aspiration et le résidu est lavé avec de l'éthanol acétique et de l'acétone neutre et ensuite séché. Ces résidus pèsent 890 gr., la triple extraction de ce résidu avec de l'acide acétique à 1% conduit à un extrait qui contient la quantité totale des hormones thyréo- tropes contenues dans la matière de départ, notamment 400.000 U.C.
Les extraits à l'éthanol ammoniacal réunis sont évaporés sans délai et sans neutralisation préalable à 30 dans le vide. On obtient 400 gr. d'un sirop qui con-

tient par milligramme 1,8 d'U.I.d'oxytocine, soit donc la quantité totale des hormones du lobe postérieur contenues dans la matière de départ* Le traitement des fractions des lobes antérieur et postérieur s'effectue selon l'exemple 1.
Exemple 5:
5 kgr. des mêmes hypophyses broyées comme dans l'exemple 4 furent comme dans ce cas extraits deux fois, chaque fois pendant 12 heures, avec une quantité d'acétone correspondant à celle d'éthanol, les mêmes quantités d'ammoniac et d'eau.
Dans le résidu d'extraction il ne pouvait plus être décelé d'hormone du lobe postérieur, mais par contre la quantité totale de l'hormone thyréotrope. Les extraits à l'acétone ammoniacal réunis contiennent, après l'évapopation sous vide à basse température, la majeure partie des hormones du lobe postérieur d'hypophyse présentes dans la matière de départ et qui présentent des qualités d'expitation utérine et d'accroissement de pression sanguine.
Exemple 6;
5 kgr. d'hypophyses complètes du porc sont additionnés après broyage de 6 litres d'alcool propylique et 160 cm3 d'ammoniaque concentré. (Comme les hypophyses fraîches contiennent 80% d'eau, on obtient ainsi une solution aqueuse à environ 60% d'alcool propylique ammoniacal). La mixture est agitée pendant 12 heures, centrifugée, et le résidu est ensuite amené au Ph 9-10 avec 4 litres d'eau et 50 cm3 d'ammoniac concentré et on ajoute ensuite 6 litres d'alcool propylique. On agite encore une fois pendant 12 heures, on centrifuge et le résidu est d'abord lavé avec de l'alcool propylique acétique et ensuite avec de l'acétone et séché.
Un extrait à l'acide acétique préparé de manière connue contient l'ensemble des hormones du lobe antérieur. Les extraits à l'alcool propylique ammoniacal réunis sont éva- porés à basse température sous vide, et on obtient 850 gr. d'un sirop dont on peut récupérer de manière connue les hormones du lobe postérieur,

5 kgr. of complete pituitary gland of the pig are added after grinding 6 liters of propyl alcohol and 160 cm3 of concentrated ammonia. (As the fresh pituitaries contain 80% water, this gives an aqueous solution of about 60% ammoniacal propyl alcohol). The mixture is stirred for 12 hours, centrifuged, and the residue is then brought to Ph 9-10 with 4 liters of water and 50 cm3 of concentrated ammonia and then 6 liters of propyl alcohol are added. Stir again for 12 hours, centrifuge and the residue is first washed with propyl acetic alcohol and then with acetone and dried.
An acetic acid extract prepared in a known manner contains all the hormones of the anterior lobe. The combined ammoniacal propyl alcohol extracts are evaporated at low temperature under vacuum, and 850 g are obtained. a syrup from which the hormones of the posterior lobe can be recovered in a known manner,
