La réaction par laquelle l'acide ascorbinique se forme à partir des acides 2-cétohexoniques se produit par énolisation.
Celle-ci consiste en principe en une transposition d'un atome H sur le groupe CO voisin, par laquelle il se forme une double liaison entre les 'atomes de C considérés suivant le schéma -CO-CH2 #-CHO=CH- (l'atome H restant peut être remplacé par un autre groupe, dans ce cas, par le groupe -OH).
Comme il est connu, les acides 2-cétohexoniques, éven- tuellement leurs esters, se transforment par énolisation en milieu alcalin ou acide en acides ascorbiniques correspondants.
Par l'énolisation avec des acides, il se forme du Furol et d'autres produits de décomposition, qui réduisent le rendement en acide ascorbinique et rendent son isolement plus difficile.
L'énolisation d'esters méthyliques avec des alcalis dans des solvants anhydres offre par contre des résultats beaucoup meilleurs ; la réaction se fait en un temps plus court avec un meilleur rendement.
L'objet de l'invention consiste en un procédé pour l'é- nolisation d'acides 2-cétohexoniques dans un milieu alcalin en présence d'hydroxydes de métaux lourds amphotères, qui for- ment avec les alcalis des combinaisons,comme par exemple Pb(OH)2 ou Zn(OH), et en fait dans des solvants anhydres. Les métaux lourds amphotères sont utilisés sous la forme de leurs combi- naisons alcalines du type Ke/ONa/OH (Me métal amphotère) en suspension ou en solution dans un solvant anhydre.
De cette façon, l'énolisation s'effectue à chaud très rapidement et en

fait en l'espace de quelques minutes seulement et avec un bon rendement ; en même temps, l'acide ascorbinique est pré- cipité sous forme de sel difficilement soluble du métal ampho- tère, permettant ainsi l'isolement de cet acide à l'état pur du mélange réactionnel par un moyen approprié. Le procédé trouvé convient particulièrement pour l'énolisation d'esters difficilement cristallisables des acides 2-cétohexoniques, à partir desquels on obtient alors des acides ascorbiniques com- plètement purs. Les sels de Pb ou de Zn de l'acide ascorbinique sont décomposés par l'action de H2SO4 ou H2S dans un solvant organique. Le rendement en substance pure atteint 70-80%' de la théorie.
La réaction suit généralement le processus suivant :

L'avantage du nouveau processus pour la préparation des acides ascorbiniques réside dans la simplification du procédé et dans la possibilité d'un isolement plus facile des acides sous forme de sels de métaux lourds insolubles, qui se laissent facilement séparer et dans l'obtention de produits plus purs.
Un autre avantage important du nouveau procédé par rapport aux procédés connus jusqu'ici est l'augmentation du rendement en substance pure à 60-70%', au lieu du rendement antérieur d'envi- ron 20% de la théorie,
Exemples d'application. - 1.- 10 parties d'ester méthylique de l'acide 2-céto-l-idonique (acide gulonique) sont dissoutes dans 100 parties de méthanol anhydre et énolisées avec une suspension de 6 parties de zincate de soude Zn/ONa/OH dans 50 parties de méthanol anhydre à une température de 65 C sous réfrigération à reflux pendant 10 mi- nutes. Par la réaction, il se forme du 1-ascorbinate de zinc

avec un rendement de 80-85% de la théorie.
On obtient une pré- cipitation complète du sel de zinc par refroidissement du mé- lange réactionnel et addition de 50 parties d'éther. Le préci- pité légèrement coloré en jaune de 1-ascorbinate de zinc est filtré par aspiration, mis en suspension à l'état humide dans 100 parties de méthanol anhydre et précipité avec une quantité équivalente d'HSO ou de H2S. Le ZnSO ou ZnS séparé est fil- tré et le filtrat concentré dans le vide à 20 C. On obtient 6,2-6,5 parties d'acide 1-ascorbinique, c'est à dire un rende- ment de 74-77% de la théorie. Le produit recristallisé du mé- thanol a un point de fusion de 181-185 C.
2.- 10 parties d'ester méthylique de l'acide 2-céto-d-gluconique sont dissoutes dans 200 parties de méthanol anhydre, énolisées avec une suspension de Zn/ONa/OH et traitées ensuite suivant l'exemple 1. Le rendement de l'acide isoascorbinique formé atteint 75-78% de la théorie ; produit pur recristallisé du méthanol fond à 173 C.
3. - 10 parties d'ester méthylique de l'acide 2-céto-l-idonique sont dissoutes dans 100 parties de méthanol anhydre et énoli- sées avec une suspension de 13 parties de Pb/ONa/OH dans 50 par- ties de méthanol anhydre à une température de 65 C sous ré- frigération à reflux pendant 15-20 minutes. Par l'énolisation, il se forme, comme il fut établi par titration, de l'acide 1- ascorbfue avec un rendement de 70% de la théorie. Le sel de plomb jaune qui se sépare est filtré après refroidissement du mélange réactionnel, mis en suspension encore humide dans du méthanol anhydre,et précipité avec H2S ou par addition d'une quantité équivalente de HSO. Le PbS séparé, éventuellement PbSO4, est filtré et le filtrat renfermant l'acide 1-ascorbi- nique concentré dans le vide.
Le rendement en substance pure atteint 60-65% de la théorie et le point de fusion l80-l83 C.

The reaction by which ascorbinic acid is formed from 2-ketohexonic acids occurs by enolization.
This consists in principle of a transposition of an H atom onto the neighboring CO group, by which a double bond is formed between the 'atoms of C considered according to the scheme -CO-CH2 # -CHO = CH- (l the remaining H atom can be replaced by another group, in this case by the group -OH).
As is known, 2-ketohexonic acids, optionally their esters, are converted by enolization in an alkaline or acidic medium into the corresponding ascorbinic acids.
On enolization with acids, Furol and other decomposition products are formed, which reduce the yield of ascorbinic acid and make its isolation more difficult.
The enolization of methyl esters with alkalis in anhydrous solvents, on the other hand, offers much better results; the reaction takes place in a shorter time with a better yield.
The object of the invention consists of a process for the enolization of 2-ketohexonic acids in an alkaline medium in the presence of amphoteric heavy metal hydroxides, which together with the alkalis form combinations, such as for example Pb (OH) 2 or Zn (OH), and indeed in anhydrous solvents. Amphoteric heavy metals are used in the form of their alkaline combinations of the type Ke / ONa / OH (Amphoteric metal Me) in suspension or in solution in an anhydrous solvent.

The process found is particularly suitable for the enolization of difficult-to-crystallize esters of 2-ketohexonic acids, from which completely pure ascorbinic acids are obtained. Pb or Zn salts of ascorbinic acid are decomposed by the action of H2SO4 or H2S in an organic solvent. The yield of pure substance is 70-80% of theory.

The advantage of the new process for the preparation of ascorbinic acids lies in the simplification of the process and in the possibility of easier isolation of the acids in the form of insoluble heavy metal salts, which are easily separated and in the obtaining of more pure products.
Another important advantage of the new process over the hitherto known processes is the increase in the yield of the substance pure to 60-70%, instead of the previous yield of about 20% of theory.
Examples of application. - 1.- 10 parts of 2-keto-l-idonic acid methyl ester (gulonic acid) are dissolved in 100 parts of anhydrous methanol and enolized with a suspension of 6 parts of sodium zincate Zn / ONa / OH in 50 parts of anhydrous methanol at a temperature of 65 ° C. under reflux refrigeration for 10 minutes. with a yield of 80-85% of theory.
Complete precipitation of the zinc salt is obtained by cooling the reaction mixture and adding 50 parts of ether. The slightly yellow colored precipitate of zinc 1-ascorbinate is filtered off with suction, suspended in the wet state in 100 parts of anhydrous methanol and precipitated with an equivalent amount of HSO or H2S. The separated ZnSO or ZnS is filtered off and the filtrate concentrated in vacuo at 20 ° C. 6.2-6.5 parts of 1-ascorbinic acid are obtained, i.e. a yield of 74-77. % of theory. The recrystallized product from methanol has a melting point of 181-185 C.
2.- 10 parts of methyl ester of 2-keto-d-gluconic acid are dissolved in 200 parts of anhydrous methanol, enolized with a suspension of Zn / ONa / OH and then treated according to Example 1. The separated PbS, optionally PbSO4, is filtered off and the filtrate containing 1-ascorbinic acid concentrated in vacuo.
The yield of pure substance is 60-65% of theory and the melting point 180-183 C.
