Selon le procédé du brevet principal, ces dérivés peuvent être obtenus par action sur la cyano-3 phénothiazine d'un halogénure d'aminoalcool X - @ (X = atome d'halogène) ou de l'un de ses sels.
Dans une première addition a été décrit un procédé de préparation des dérivés du brevet principal consistant à chauffer un phénothiazinyl-10 carboxylate d'aminolacool de formule générale:

Selon le brevet principal les dérivés de la cyano-3 phénothiazine sont soit des antihistaminiques, soit des poten- tialisateurs de narcose et des déconnecteurs du système nerveux végétatif, soit encore des antiémétique, des spasmolytiques ou des hypotenseurs. Il a maintenant été trouvé que les déri- vés de la présente addition possèdent des propriétés pharma- codynamiques très remarquables; ils présentent en effet à la fois une activité sédative élevée, mise en évidence par exem- ple par les tests de potentialisation de narcose, et des ac- tions secondaires très faibles, en particulier une toxicité très réduite.
Les exemples suivants, donnés à titre non limitatif., montrent comment l'invention peut être mise en pratique. Les points de fusion ont été déterminés au banc Kofler.
EXEMPLE 1. - @
A une solution de tertiobutylate de sodiu, préparée à partir de 11,5 g de sodium et 900 cm3 de tertiobutanol, on ajoute 1.500 cm3 de toluène anhydre et 100,8 g de cyano-3 phénothiazine et on élimine le tertiobutanol en distillant à la pression atmosphérique jusqu'à atteindre une température de vapeur de 109 . On coule en 30 minutes sans refroidir une solu- tion de 83,4 g de chloro-1 morpholino-3 propane dans 350 cm3 de toluène et chauffe ensuite à reflux pendant 5 heures.
Après refroidissement, on lave la liqueur réaction- nelle avec 500 cm3 d'eau; on épuise la phase toluènique suc- cessivement avec 300 cm3 d'acide chlorhydrique 2 N et 250 cm3 d'acide chlorhydrique 0,5 N; on lave la phase aqueuse acide avec 250 cm3 d'éther et on l'alcalinise avec 100 cm3 de lessive de soude (d = 1,33); on extrait la base libérée avec 600 cm3 d'acétate d'éthyle, on sèche l'extrait organique sur carbonate de potassium et chasse le solvant sous une pression de 20 mm de mercure environ.

On dissout la base brute résiduelle dans 3 litres de benzène, on filtre la solution sur une colonne de 2 kg d'alu- mine pour chromatographie et on élue avec 8 litres de benzène.
On concentre à sec les fractions benzéniques réunies et on re- cristallise le résidu dans du cyclohexane. On obtient ainsi 54 g de cyano-3 (morpholino-3' propyl)-10 phénothiazine, produit cristallisé jaune fondant à 98-100 . Le picrate, pré- paré dans le n-propanol, est une poudre orangée fondant à 196- 197 .
EXEMPLE 2. -
On chauffe 138,2 g de (cyano-3' phénothiazinyl-10') éarboxylate de morpholino-3 propyle entre 180 et 210 sous une Mission de 1 mm de mercure jusqu'à la fin du dégagement de @@@ carbonique. On refroidit à 100 , on dissout la masse réac- tionnelle dans 1,25 litres d'acétate d'éthyle et on épuise solution avec 200 cm3 d'acide sulfurique 4 N et 2 fois 150 cm3 d'acide sulfurique 2N. On alcalinise la phase aqueuse acide avec 200 cm3 de lessive de soude (d = 1,33), on extrait la base libérée avec 1 litre d'acétate d'éthyle, on sèche l'extrait or- ganique avec du carbonate de potassium anhydre et on chasse le solvant sous une pression de 20 mm de mercure.
On dissout la base résiduelle dans 1 litre de benzène, on filtre la solution sur une colonne de 500 g d'alumine pour chromatographie et on élue avec 500 cm3 de benzène et ensuite avec 2,5 litres de mélange benzène-acétate d'éthyle (9 : 1). On concen- tre à sec les fractions réunies et on recristallise le résidu dans le cyclohexane. On obtient ainsi 110 g de cyano-3 (morpholino- 3' propyl)-10 phénothiazine, identique ,au composé préparé dans l'exemple 1.
Le (cyano-3' phénothiazinyl-10') carboxylate de morpholino- 3 propyle nécessaire (P.F. : 132-134 ) est préparé par condensation du chlorure de l'acide (cyano-3 phénothiazinyl), carboxylique (1 mol. ) sur le morpholino-3 propanol (1,2 mol. ) dans le toluène

à reflux pendant 4 heures.
EXEMPLE 3 . -
Dans une suspension de 67,2 g de cyano-3 phénothiazine dans 1.200 cm3 de toluène anhydre préalablement chauffée à 90 , on introduit 12,3 g d'amidure de sodium à 95% et on chauffe à reflux pendant 30 minutes. On coule ensuite 66,3 g de chloro-1 méthyl-2 morpholino-3 propane en 70 minutes et chauffe finalement à reflux pendant 19 heures.
Après refroidissement, on lave la liqueur réaction- nelle avec de l'eau; on épuise la phase toluénique avec 650 cm3 d'acide chlorhydrique normal; on alcalinise la phase aqueuse avec 200 cm3 de solution de soude 4 N et on extrait la base libérée avec du chloroforme. On lave la phase chloroformique avec de l'eau, on la sèche sur du sulfate de sodium sec et on la concen- tre à sec sous une pression de 20 mm de mercure environ.
On dissout la base résiduelle dans 2,2 litres de mélange benzène-cyclohexane (1 : 1), on filtre la solution sur une colonne de 550 g d'alumine pour chromatographie et on élue avec 5 litres du même solvant. On concentre à sec les fractions réunies et on recristallise le résidu dans du cyclohexane.
On obtient ainsi 50 g de cyano-3 (méthyl-2' morpholino-3' propyl) -10 phénothiazine, produit cristallisé jaune-crème fondant à 136- 138 . Le maléate acide, préparé dans l'acétone, fond à 219-221 .
Le chloro-1 méthyl-2 morpholino-3 propane nécessaire (P.E.15= 108-110 ) est préparé par condensation de la morpho- line (2 mol.) sur le chloro-1 méthyl-2 bromo-3 propane (1 mol. ) dans le benzène à reflux pendant 15 heures.

According to the main patent, the derivatives of 3-cyanophenothiazine are either antihistamines, or potentiators of narcosis and disconnectors of the vegetative nervous system, or even antiemetics, spasmolytics or hypotensives. It has now been found that the derivatives of the present addition possess very remarkable pharmacodynamic properties; they present in fact both a high sedative activity, demonstrated for example by the tests of potentiation of narcosis, and very weak secondary actions, in particular a very reduced toxicity.
The following examples, given without limitation, show how the invention can be put into practice. The melting points were determined on the Kofler bench.
EXAMPLE 1. - @
To a solution of sodium tert-butoxide, prepared from 11.5 g of sodium and 900 cm3 of tert-butanol, 1,500 cm3 of anhydrous toluene and 100.8 g of cyano-3-phenothiazine are added and the tert-butanol is removed by distillation with the atmospheric pressure until a vapor temperature of 109 is reached. A solution of 83.4 g of 1-chloro-3-morpholino-propane in 350 cm 3 of toluene is poured over 30 minutes without cooling and then heated under reflux for 5 hours.
After cooling, the reaction liquor is washed with 500 cm3 of water; the toluene phase is exhausted successively with 300 cm3 of 2N hydrochloric acid and 250 cm3 of 0.5N hydrochloric acid; the acidic aqueous phase is washed with 250 cm3 of ether and basified with 100 cm3 of sodium hydroxide solution (d = 1.33); the base liberated is extracted with 600 cm3 of ethyl acetate, the organic extract is dried over potassium carbonate and the solvent is removed under a pressure of about 20 mm of mercury.

The residual crude base is dissolved in 3 liters of benzene, the solution is filtered through a column of 2 kg of aluminum for chromatography and eluted with 8 liters of benzene.
The combined benzene fractions are concentrated to dryness and the residue recrystallized from cyclohexane. 54 g of cyano-3 (morpholino-3 ′ propyl) -10 phenothiazine are thus obtained, a yellow crystalline product melting at 98-100. Picrate, prepared in n-propanol, is an orange powder melting at 196-197.
EXAMPLE 2. -
138.2 g of morpholino-3-propyl (3'-cyano-phenothiazinyl-10 ') -arboxylate are heated to between 180 and 210 under a concentration of 1 mm of mercury until the evolution of carbon dioxide has ceased. The mixture is cooled to 100, the reaction mass is dissolved in 1.25 liters of ethyl acetate and the solution is exhausted with 200 cm3 of 4N sulfuric acid and 2 times 150 cm3 of 2N sulfuric acid. The acidic aqueous phase is basified with 200 cm3 of sodium hydroxide solution (d = 1.33), the liberated base is extracted with 1 liter of ethyl acetate, the organic extract is dried with anhydrous potassium carbonate. and the solvent is removed under a pressure of 20 mm of mercury.
The residual base is dissolved in 1 liter of benzene, the solution is filtered through a column of 500 g of alumina for chromatography and eluted with 500 cm3 of benzene and then with 2.5 liters of benzene-ethyl acetate mixture ( 9: 1). The combined fractions are concentrated to dryness and the residue recrystallized from cyclohexane. 110 g of cyano-3 (morpholino-3 'propyl) -10 phenothiazine, identical to the compound prepared in Example 1, are thus obtained.
The required morpholinopropyl (3 'cyano-phenothiazinyl-10') carboxylate (mp: 132-134) is prepared by condensation of (3-cyano-phenothiazinyl) carboxylic acid (1 mol.) Chloride on the morpholino-3 propanol (1.2 mol.) in toluene

at reflux for 4 hours.
EXAMPLE 3. -
12.3 g of 95% sodium amide are introduced into a suspension of 67.2 g of 3-cyanophenothiazine in 1,200 cm 3 of anhydrous toluene heated to 90% and the mixture is refluxed for 30 minutes. 66.3 g of 1-chloro-2-methyl-3-morpholino-propane are then poured in over 70 minutes and finally heated under reflux for 19 hours.
After cooling, the reaction liquor is washed with water; the toluene phase is exhausted with 650 cm3 of normal hydrochloric acid; the aqueous phase is basified with 200 cm3 of 4N sodium hydroxide solution and the liberated base is extracted with chloroform. The chloroform phase is washed with water, dried over dry sodium sulfate and concentrated to dryness under a pressure of about 20 mm Hg.
The residual base is dissolved in 2.2 liters of benzene-cyclohexane (1: 1), the solution is filtered through a column of 550 g of alumina for chromatography and eluted with 5 liters of the same solvent. The combined fractions are concentrated to dryness and the residue recrystallized from cyclohexane.
50 g of cyano-3 (2 'methyl morpholino-3' propyl) -10 phenothiazine are thus obtained, a yellow-cream crystalline product, melting at 136-138. The acid maleate, prepared in acetone, melts at 219-221.
The 1-chloro-2-methyl-3-morpholino-propane (PE15 = 108-110) is prepared by condensation of morpholine (2 mol.) With 1-chloro-2-methyl-3-bromo-propane (1 mol. ) in benzene at reflux for 15 hours.
