La présente invention concerne un procédé de préparation d'anesthési- ques locaux intéressants en clinique,de la classe des alcoylaminoacyl-anilines et elle a pour objet de préparer des toluidines de cette classe de composés, consi- dérés jusqu'ici comme sans application clinique possible. 



   Les articles de Erdtman et Löfgren (Svensk   Kemisk     Tidskrift,   49, 163, 1939) et Löfgren   (Arkiv   för Kemi,   Mineralogi   och Geologi, 22 A. (1946) N  18) ont montré que les alcoylaminoacyl-toluidines possédaient des propriétés anesthésiques locales à un degré plus ou moins élevé. Dans ces articles, ainsi que dans un ar- ticle de Goldberg (Acta   Physiol.     Seand.   18,   1,   1949) les auteurs ont toutefois montré que les toluidines, que le groupe méthyle soit en position ortho, méta ou para, ont des effets secondaires, notamment une forte action d'irritation locale et souvent un effet anesthésique médiocre, tels qu'elles ne peuvent être utilisées en clinique.

   Il semble également qu'aucun dérivé toluidinique n'ait été utilisé en clinique comme anesthésique local. Il a au contraire été montré par Löfgen et autres, par exemple dans le brevet belge n  483.439,   qu'on   ne pouvait obtenir d'anesthésiques locaux intéressants qu'en introduisant dans le noyau benzénique un groupe méthyle supplémentaire, à la condition que les deux groupes méthyle soient en position ortho, de manière à obtenir une 2.6-xylidide, toutes les au- tres   xylidides   s'étant montrées particulièrement toxiques et irritantes. 



   Cependant, il a maintenant été découvert de manière surprenante, que des alcoylaminoacyl-o-toluidines peuvent présenter également des propriétés anes- thésiques locales très intéressantes. Ces propriétés favorables ne se manifes- tent toutefois que chez certains composés particuliers de ce type, savoir les (alpha-monoalcoylamino-propionyl)-o-toluidines de formule générale

 dans laquelle R représente un groupe alcoyle de 2 à 4, de préférence 2 à 3 ato- mes de carbone, c'est-à-dire les (alpha-monoalcoylamino-propionyl)-o-toluidines dans lesquelles le groupe alcoyle est de préférence un groupe éthyle, propyle-n.   ou isopropyle et peut très bien être un groupe isobutyle (-CH2 - CE -(CE )2), mais de préférence pas un groupe butyle-n. qui possède plus de 3 atomes de car-   bone en chaîne droite après l'azote aminique.

   Les composés de ce type manifestent un effet anesthésique clinique au moins aussi bon que celui de la Lidocaine, qui est une 2.6-diméthyl-alcoylaminoacyl-aniline, et plus précisément la diéthylami- noacétyl-2.6-xylidine, et .ils sont complètement exempts d'effets secondaires à toutes les doses raisonnables. En raison de leur toxicité considérablement plus faible, ils possèdent un indice thérapeutique plus favorable que la Lidocaïne. 



   C'est un procédé de préparation de composés du type en question qui fait l'objet de   la;   présente invention. Ce procédé a pour avantage principal de permettre d'utiliser, à titre de matière première, l'o-toluidine que l'on trouve   facilement. dans¯ le   commerce sous une forme suffisamment pure pour les synthèses de produits médicinaux.,.

   La valeur clinique des composés suivant l'invention a été prouvée par certaines expériences pharmacologiques qui ont été réalisées. Le tableau ci- dessous donne les résultats d'essais effectués au moyen de différents composés répondant à la formule générale ci-dessus, dans laquelle R a la signification in- diquée dans le tableau sous l'en-tête "composé de base", la Lidocaïne constituant la norme de référence. Les composés ont été expérimentés sous forme de leur chlorhydrate et selon les procédés usuels.

   Les essais de toxicité ont été effec- tués sur la souris par injections sous-cutanées et intra-veineuses, les chiffres indiqués étant les quantités de composé, respectivement en grammes par kilogramme et en milligrammes par kilogramme de poids du corps de la souris, pour lesquelles on atteint la dose léthale chez   50 %   des animaux. L'effet irritant a été contrô- lé par injection sous-cutanée dans l'oreille du lapin et sur la face intérieure de l'avant-bras chez l'homme; cet effét est celui qui s'exerce au lieu de l'in- jection. L'effet anesthésique a été expérimenté par injection hypodermique sur la face intérieure de l'avant-bras de l'homme et on a évalué la période latente   (lat.)   et la durée.

De plus, les essais cliniques effectués à l'aide du composé 3 qui, d'après le tableau ci-dessus, apparaît comme possédant des propriétés particuliè- rement favorablesont montré que ce composé était nettement supérieur à la Lido-   caine   en tant qu'anesthésique de conduction. 



   Il ressort de ces essais que tous les composés selon l'invention (2 à 6) sont au moins aussi bons que la   Lidocalne   pour ce qui concerne l'effet anes- thésique latent et, compte tenu des limites d'erreur, ont un effet aussi prolongé

 que le composé de référence. Le plus remarquable est que les composés 2 à 4, dans lesquels R est un groupe alcoyle contenant 2 ou 3 atomes de carbone, non seulement égalent la Lidocaine, mais sont même considérablement moins toxiques que cette dernière et sont égaux à celle-ci pour ce qui concerne l'irritation, en tout cas chez l'homme, ce qui est en fait ce qui compte en application clinique. 



  Il est remarquable que le composé   propylique-n.   ait un effet un peu moins irri- tant que la Lidocaine. Il est également très surprenant que les composés butyli- ques 5 et 6 se rapprochent de la Lidocaine pour ce qui concerne l'effet irritant chez l'homme, ainsi que la toxicité. L'oreille du lapin est très sensible et l'ir- ritation relativement forte obtenue chez le lapin avec le composé butylique-n. 



  (6) peut être significative, car elle semble indiquer que la chaîne de groupes méthyléniques ne doit pas être trop longue. Le composé isobutylique étant nette- ment meilleur que le composé butylique n. et se rapprochant beaucoup des composés éthylique et propylique, il semble qu'on puisse conclure que les composés répon- dant à la formule générale dans laquelle le groupe alcoyle R ne contient pas plus de 3 atomes de carbone en chaîne droite à partir de l'azote aminique, c'est-à- dire les composés isobutylique et butylique secondaire et tertiaire, forment un groupe de composés anesthésiques plus intéressants que ceux possédant des chaînes carbonées plus longues. Dans ce groupe, le sous-groupe dans lequel le groupe al- coyle R contient 2 ou 3 atomes de carbone est le plus intéressant. 



   Les exemples suivants illustrent la présente invention mais ne sont pas limitatifs.

  EXEMPLE 1 - (alpha-éthylaminopropionyl)-.2.-toluidide 
On dissout une mole d'o-toluidine dans 800 ml d'acide acétique gla- cial. On refroidit le mélange à 10 C puis on ajoute 1,1 mole de bromure d'alpha- bromopropionyle. On agite énergiquement le mélange pendant environ une minute et on introduit, en une fois, une solution d'acétate de sodium (330 g de CH3COONa,   3 H 0 dans 1380 ml d'eau) ou une autre substance ou solution tampon ou alcalinisante. On secoue le mélange réactionnel pendant une demi-heure. On sépare par   filtration le précipité formé, on le lave à l'eau et on le sèche. Le produit est suffisamment pur pour l'opération qui suit. Rendement 70 à 80 % du rendement théorique. Point de fusion 133-134 C. 



   On mélange une mole de la bromotoluidide ainsi obtenue avec une so- lution de 2,7 moles de monoéthylamine dans 550 ml de benzène anhydre. On chauffe alors le mélange réactionnel à 8000 pendant huit heures dans un autoclave. La réaction terminée, on étend le mélange au moyen d'éther absolu et on sépare par filtration le bromure d'éthylamine précipité. On épuise alors la solution dans le mélange d'éther et de benzène au moyen d'acide chlorhydrique 4 N jusqu'à ce que la solution ne s'opacifie plus par l'ammoniaque. On lave le mélange des ex- traits en le secouant une fois avec de l'éther. On rejette l'extrait éthéré et - on alcalinise la phase aqueuse par addition d'une solution aqueuse concentrée d'ammoniaque. On reprend la base mise en liberté, au moyen d'éther.

   Après sécha-   ge sur SO4Na2, on évapore la solution éthérée et on distille le résidu sous pres= sion réduite, de manière à obtenir une huile incolore de point d'ébullition de   145-147 C sous 0,9 mm. L'huile se solidifie en cristaux incolores dont le point de solidification est de   36-37 Co   Rendement 80   %.   



    EXEMPLE   2 - (alpha-éthylaminopropionyl)-o-toluidide - 
On dissout une mole - d'o-toluidine dans cinq fois son volume d'un solvant organique soluble dans l'eau (acétone, dioxane, etc..). On refroidit la solution à 10 C et on ajoute 1,2 mole de bromure d'alpha-bromo-propionyle, puis on introduit une solution d'acétate de sodium comme dans l'exemple 1, ou d'un au- tre agent tampon ou alcalinisant, et on traite le mélange réactionnel de la même manière que dans l'exemple 1.

   Rendement   75-80%.   Au lieu d'ajouter une substance ou une solution tampon,.on peut utiliser pour la synthèse une quantité double d'o-

 EXEMPLE 3 - (alpha-propyl-n-amino-propionyl)-o-toluidid.e 
On mélange une mole d'alpha-bromopropionyl-o-toluidide avec une solu- tion de 3 moles de propyl-n..-amine dans 500 ml de benzène anhydre et on chauffe le mélange réactionnel à l'autoclave à 80 C pendant huit heures. Après refroidis- sement, on traite le mélange réactionnel comme dans l'exemple 1. La base obtenue est une huile incolore bouillant à 159-162 C sous 0,1 mm. Rendement 55 %.

  EXEMPLE 4 - (alpha-butyl-n.-aminopropionyl)-o-toluidide - 
On chauffe une mole   d'alpha-bromopropionyl-o-toluidide   et 2,7 moles de   butyl-n-amine   dans 1 litre de benzène anhydre au reflux pendant huit heures. 



  On refroidit le mélange réactionnel et on l'étend au moyen d'un litre d'éther ab- solu. Après filtration, on précipite la base à l'état de chlorhydrate par addi- tion d'une solution d'acide chlorhydrique dans l'éther. On fait recristalliser le chlorhydrate ainsi obtenu.au sein d'alcool absolu, ce qui donne des cristaux incolores fondant à 175-177 C. 



  Rendement 82 %.

  EXEMPLE 1 - (alpha-isopropylamino-propionyl)-o-toluidide On fait réagir 1'alpha-bromopropionyl--toluidide de la même manière que dans l'exemple 3, la seule différence étant qu'on utilise l'isopropylamine au lieu de la propylamine. Après refroidissement, on étend le mélange réaction- nel au moyen'd'éther absolu et on sépare le précipité par filtration (bromhydrate d'isopropylamine). On traite alors la solution dans l'éther-benzène au moyen d'acide chlorhydrique dans l'éther et on fait recristalliser le chlorhydrate de la base ainsi obtenue au sein d'eau en ajoutant quelques millilitres d'acide chlor- hydrique: on obtient des cristaux incolores fondant à 236-237 C. Rendement 75%. 



  EXEMPLE 6 -   (alpha-isobutylaminopropionyl)-o-toluidide -   
On chauffe une mole   d'alpha-bromopropionyl-o-toluidide   à   80pendant   huit heures avec 2,7 moles d'isobutylamine dans 1 litre de benzène anhydre. Après refroidissement, on effectue la précipitation au moyen d'un excès d'acide chlor- hydrique en solution éthérée. On sépare le précipité de chlorhydrate par filtra- tion sous vide et on le lave au moyen de petites quantités d'eau froide. La sub- stance ainsi obtenue peut être purifiée par recristallisation au sein d'eau. On obtient des cristaux incolores fondant à   221-223 C.   Rendement 75 %.

  De.la même manière, on peut obtenir l'(alpha-butyl-sec.-amino-propio- nyl)-¯o-toluidide. On peut obtenir la base libre, à partir du chlorhydrate, sous forme d'une huile incolore. Point d'ébullition 148-1500C sous 0,7 mm de mercure. 



  EXEMPLE 7 -(alpha-propyl-.-aminopropionyl)-±-toluididê - On mélange ufia môle..,'o:itely.lphôàphazô-o-tdlR.id,.dê,defn.v.e' ïCHC6H4¯Nlg-Çi ;   C6H@ avec 2 moles de chlorhydrate d'acide alpha-propyl-n-aminopropionique et on chauffe à 100-150 C pendant une heure. Après refroidissement, on étend le mélan-   ge à l'aide d'eau et on alcalinise par addition d'hydroxyde de sodium, puis on reprend au moyen d'éther la base mise en liberté. On procède à un épuisement suffisant de la solution au moyen d'acide chlorhydrique 4 N, puis on traite comme dans l'exemple 1. Rendement 85 %.

  EXEMPLE 8 - (alpha7isopropylamino-propionyl)-.2.-toluidide 
On mélange intimement une mole d'o-toluidine et 1 mole de chlorhydra- te d'acide alpha-isopropyl-amino-propionique. Il se forme une masse cristallisée avec dégagement de chaleur. On traite alors le mélange réactionnel au moyen de 0,25 mole de P205 et on chauffe au bain-marie pendant quinze minutes. On étend

 ensuite à l'aide d'un litre d'eau et on alcalinise par addition d'hydroxyde de sodium 4 N. On épuise la toluidide ainsi mise en liberté au moyen d'éther et on sèche la solution éthérée sur sulfate de sodium anhydre. L'addition d'acide ohlor- hydrique gazeux précipite le chlorhydrate correspondant, qu'on purifie par recris- tallisation. Rendement 70   %.   



    EXEMPLE   9 -   (alpha-isopropylamino-propionyl)-o-toluidide   
On pulvérise une mole du sel de sodium de l'acide alpha-(N-carbobenzo-

 xy-isopropylamino)-propionique et on le met en suspensiont-dans du benzène anhydre, puis on ajoute 1 mole de chlorure de benzoyle et on agite le mélange pendant deux heures et demie à la température ambiante. On ajoute alors 1 mole   d'o-toluidine   et 1 mole de triéthylamine. On continue à agiter pendant deux heures à la tempé- rature ambiante, puis pendant le même temps à 100 C. Après refroidissement, on épuise la phase benzénique à l'eau et on la sèche sur sulfate de sodium. On ajoute alors du catalyseur au palladium à la solution benzénique et on effectue l'hydrogénation à la température ambiante sous une pression de trois atmosphères. 



  On filtre la solution ainsi obtenue et on précipite le chlorhydrate par addition d'acide chlorhydrique au sein d'éther. On sépare par filtration le précipité for- mé, qu'on peut faire recristalliser au sein d'eau contenant un peu   d'acidlor-   hydrique. Rendement 70 %.

  EXEMPLE' 10 - (alpha-éthylaminopropionyl)-o-toluidide On ajoute lentement une solution d'une mole de chlorure   d'alpha-,(± -

Par refroidissement, l'apha-(p-toluène-sulfonyloxy)   propionyl-o-toluidide   cristallise de la solution filtrée. Rendement 80   %.   



   On mélange une mole du composé toluène-sulfonyle ainsi obtenu avec une solution de 2,7 moles de monoéthylamine dans 500 ml de benzène exempt d'eau et on chauffe à l'autoclave à 80 C pendant huit heures. L'isolement est effec- tué selon l'exemple 1.

On dissout une mole de l'éther éthylique de l'acide alpha-propyl-n.- amino-propionique et 1 mole   d'o-toluidide   dans 1 litre de benzène anhydre. On ajoute, par portions et en agitant, une mole d'amide sodique. Après cette addi- tion, on laisse le mélange réactionnel pendant une heure à la température ambian- te en agitant de manière continue, puis on chauffe au reflux. On traite le mé- lange réactionnel refroidi selon l'exemple 1. Rendement 55 %.

On dissout une mole d'acide alpha-n-butylaminopropionique et 1,2 mole d'isocyanate d'o-toluyle dans 250 ml de pyridine sèche. On chauffe le mélange au reflux jusqu'à cessation du dégagement d'anhydride carbonique. On chasse la pyri- dine par distillation et on chauffe le réside avec 500 ml d'acide chlorhydrique 4 N, au   bain-marie,   pendant trente minutes, puis on refroidit et on filtre. On lave le précipité ainsi obtenu à l'aide d'une petite quantité d'eau froide, puis on le fait reoristalliser au sein d'éthanol, ce qui donne des cristaux incolores fondant à 175-177 C. Rendement 67 %.
