L'iso- lement de la thréonine à partir de ce mélange est assez dif- ficile et coûteux.

Selon la présente invention, la demanderesse a trouvé un procédé par lequel on obtient du bis(acétaldéhyde)bis (thréoninato)cuivre que l'on peut transformer en thréonine pure par élimination de l'acétaldéhyde et du cuivre. Un mode de réalisation préféré pour cette invention comporte les éta-

1 afin de produire le bis(acétaldé- hyde)bis(thréoninato)cuivre, et de récupération du produit sous forme pure, on conduit la réaction en incorporant les réactifs dans le rapport molaire précité à un solvant tel que l'eau, le méthanol, l'éthanol ou leur mélange, qui con- tient un catalyseur alcalin tel que l'hydroxyde de sodium, l'hydroxyde de potassium, le carbonate de sodium ou le car- bonate de potassium, et en chauffant le mélange durant quel- ques heures à une température de 50 à 80 C. Ou bien, on peut ajouter la quantité requise d'acétaldéhyde au milieu réaction- nel en deux ou plusieurs fois pendant que la réaction se pour- suit .
Le bis(acétaldéhyde)bis(thrêoninato)cuivre résultant est relativement insoluble dans l'eau, alors que le composé correspondant d'allothréonine est soluble dans l'eau.Lorsqu'or effectue cette réaction dans l'eau, on peut isoler ledit com- posé de coordination de la thréonine que l'on a ainsi purifié en opérant une cristallisation qui sépare le composé du mi- lieu réactionnel.
La réaction dans un solvant organique tel que le métha- nol et l'éthanol aboutit à former une solution, dans ce sol- vant, du bis(acétaldéhyde)bis(thréoninato)cuivre et de son isomère de type allo. Lorsqu'on concentre la solution à sic- cité et qu'on extrait le résidu à l'eau, on récupère par

filtration le bis(acétaldéhyde)bis(thréoninato)ouivre purifié, le dérivé correspondant d'allothréonine restant dissous dans le filtrat. On peut préparer la thréonine libre purifiée en éliminant le cuivre et l'acétaldéhyde à partir de l'intermé- diaire purifié. Il est commode d'effectuer cette opération en faisant barboter de l'hydrogène sulfuré gazeux dans une suspension aqueuse de cet intermédiaire.
Puis on filtre le mélange ainsi traité pour en éliminer le sulfure de cuivre, et l'on concentre le filtrat. Lorsqu'on ajoute à ce concentré une grande quantité de méthanol absolu, la thréonine purifiée cristallise. On peut également effectuer cette opération en utilisant une résine échangeuse de cations, seule ou en con- jonction avec ledit hydrogène sulfuré. Ainsi, on prépare la thréonine libre en faisant passer une solution de l'intermé- diaire dissous dans de l'ammoniaque aqueuse à travers une colonne de résine échangeuse de cations, comme par exemple P'Amberlite IR-120", que l'on a traitée au préalable avec de l'ammoniaque aqueuse, en concentrant la solution après ce passage et en ajoutant ensuite du méthanol absolu.
EXEMPLE 1
On incorpore une suspension de 105 grammes (0,5 mole) de bls(glycinato)cuivre dans 850 ce de méthanol à une solution de 10 grammes d'hydroxyde de potassium dans 180 cc de méthanol. on ajoute à ce mélange 140 ce (2,5 moles) d'acétaldéhyde et l'on chauffe ensuite le mélange résultant à 60 C durant une heure tout en agitant.
Les particules de bis(glycinato) cuivre disparaissent au fur et à mesure de la progression de la réac- tion. on filtre le mélange pour en éliminer les matières inso- lubles On ajoute au filtrat 11 grammes d'acide acétique et l'on concentre à siccité sous pression réduite. on extrait le résidu avec 150 cc d'eau, on lave à l'eau et l'on sèche, on obtient 92,7 grammes de bis(acétaldéhyde)bis(thréoninato)cuivre qui ne contient presque pas de diastéréo-isomère du type allo.

Trouvé: C: 36;87 Ó;H; 6,06 Ó; N: 7,48 Ó CU: 16,39 % on met ce produit en suspension dans 500 ce d'eau con- tenant 1,2 gramme d'acide acétique, on fait barboter dans cette suspension de l'hydrogène sulfuré gazeux jusqu'à ce que disparaisse la couleur bleue de la suspension. on filtre la solution résultante pour en éliminer le précipité et l'on concentre le filtrat au 1/5e du volume d'origine. on ajoute à ce concentré 400 cc de méthanol et l'on recueille la DO- thréonine cristallisée. On obtient 48,4 grammes de DL-thrée- nine (qui se décompose à 237 C) après avoir recristallisé le produit dans du méthanol dilué.
Le produit s'avère être de la IL-thréonine à 100 Ó d'après un essai biologique et il n'ap- parait aucun point d'allothréonine lors d'une chromatogra- phie sur papier.
EXEMPLE 2
On chauffe à 52 pendant une heure, tout en agitant, une suspension de 58 grandes (0,25 mole) de bis(glyoinato) cuivre dans 600 ce d'eau, avec 150 ce de solution aqueuse d'hydroxyde de potassium à 4 % et 67 grammes (1,5 mole) d'acétaldéhyde. On filtre le mélange réactionnel pour en éli- miner les matières insolubles. On ajoute au filtrat 6,5 gram- mes d'acide acétique et 44 grammes (1 mole) d'acétaldéhyde tout en agitant, et on laisse ensuite le mélange reposer pour permettre la cristallisation. on obtient par filtration, la- vage à l'eau et séchage 30 grammes de bis(acétaldédyde)bis (thréoninato)cuivre. Le produit s'avère ne contenir pratique- ment pas de dérivé d'allothréonine.
