  Inhibiteur   de   fonctions de thrombocytes.

 <Desc/Clms Page number 2>

 <Desc/Clms Page number 3>

  Inhibitor of thrombocyte functions.

 <Desc / Clms Page number 2>

   The present invention relates to an inhibitor of thrombocyte functions, such as adhesion, agglomeration, contraction and release of biogenic amines from amine accumulating cells.

   During the last years, the physiological and pathological functions of blood platelets or thrombocytes during the phenomena of hemostasis and thrombosis have been elucidated. They are based on the capacities of thrombocytes to adhere to the vascular dothelium, to agglomerate and to modify their structure as well as to release coagulation factors and other biologically active substances (MARKWARDT, F .: "On the influence on thrombogenesis by drug blockade of function of thrombocytes", Dtsch. Ges. Wesen 22, 1967).

   These phenomena initiate both the formation of the hemostatic plug and that of the white intravascular thrombus. The effect of the enticoagulant substances, used until now in the prophylaxis of thrombosis, is based on the intervention in the subsequent coagulation phenomena which act on the formation of fibrin. They are not able to pocket the initial phase of thrombus formation which is triggered by thrombocytes.

   During his research into the drug influence exerted on the function of platelets, MARKWARDT suggested that the opium alkaloid, papaverine, long known as a spasmolytic agent, can inhibit several functions of platelets (MARKWARDT , F .: BARTHELt W., GLUSA, E and HOFFMANN, A .: "Research on

 <Desc / Clms Page number 3>

 the influence of papaverine on the functions of platelets eanguinea ", Experentia (Bas * 1) 22, 578, 1966. Same authors:" Research on the influence of papaverine on the reactions of blood platelets ", Naunyn- Schmiedeberge Arch, exp.

  Path. Pharmak, 257, 420, 1967. This pharmaceutical product, however, is not suitable for application as an anti-thrombosis factor because the high doses of papaverine, necessary for the inhibition of the functions of the thrombocytes, as well as others. Pharmacodynamic properties of papaverine (such as vaaodilatanta spasmolytic effects which lead to hypotonia) preclude its therapeutic use in the prophylaxis of thrombosis.

   On the other hand, the substance obtainable according to the present invention produces inhibitory effects on the functions of thrombocytes, which far exceed those of papaverine and do not exhibit harmful pharmacodynamic effects, specific to papaverine. and others derived from papaverine.

   With this substance, we succeed in producing in vitro and ir. vivo the desired state of reversible drug thrombasthenia. This is why this substance, in accordance with the present invention, is particularly suitable for use in the prophylaxis of thrombosis because it already stops the initial phase of the formation of the thrombus.

   The product can be administered in the form of tablets, dragees, capsules or ampoules, while still being able. contain, for each form of application, the appropriate excipients.
