Abstract

Patente de Invenção: "GENE SINTéTICO PARA EXPRESSãO DE UMA PROTEìNA RETROVIRAL ATIVA EM EUCARIOTAS". A presente invenção se caracteriza por um gene sintético ou região de um gene o qual faz uso aprimorado de um códon comparado ao gene do tipo silvestre e o qual se destina à expressão em alto nível de uma proteína retroviral em células eucariotas, a proteína retroviral expressa tendo atividade enzimática na célula eucariota. Além disso, a invenção se caracteriza por um gene sintético ou região de um gene que codifica uma enzima retroviral ou parte de uma enzima retroviral normalmente expressa em uma célula de mamífero ou outra em que pelo menos um códon não preferido no gene do tipo silvestre que codifica a enzima tenha sido substituído por um códon preferido que codifica o mesmo aminoácido. A proteína retroviral pode ser uma protease, transcriptase reversa, proteína integrase ou um precursor da poliproteína gag-pol da mesma. Em uma concretização, a proteína retroviral com atividade enzimática é uma proteína lentiviral. Em outras concretizações, a proteína enzimaticamente ativa é uma enzima pol. Em concretizações mais preferidas, a proteína enzimaticamente ativa é uma integrase lentiviral. Em uma concretização ainda mais preferida, a enzima é uma enzima do HIV. Em concretizações mais preferidas, a proteína enzimaticamente ativa é integrase de HIV. A presente invenção também inclui um método de detecção para a integrase intracelular usando-se um gene repórter sem promotor.
