Abstract
"MELHORA NA QUALIDADE DE PRODUTOS EM PROCESSOS DE CULTURA DE CéLULAS DE MAMìFEROS PARA A PRODUçãO DE PROTEìNAS". A presente invenção refere-se a métodos e processos para a produção de proteínas, particularmente glicoproteínas, por cultura de células animais ou de mamíferos, de forma ilustrativa, mas não limitada às culturas de células em batelada alimentada. Os processos compreendem a alimentação das células com a D-galactose, preferencialmente com meio de alimentação contendo a D-galactose, preferencialmente diariamente, para sustentar um nível eficiente de sialilação da D-galactose na cultura ao longo de sua duração, aumentando assim a sialilação das proteínas produzidas. Os processos podem ainda compreender pelo menos duas alterações na temperatura realizadas durante o período de cultivo, em que a temperatura é menor no final do período de cultivo do que no momento da cultura de células inicial. Os processos de cultura de células da invenção que envolvem duas ou mais alterações na temperatura sustentam uma alta viabilidade celular e podem permitir uma fase de produção de proteínas estendida. Os processos também podem compreender a adição retardada do composto polianiónico em um momento após a inoculação. A suplementação das culturas com a D-galactose, preferencialmente em um meio de alimentação, para sustentar a galactose em níveis eficientes de sialilação nas culturas até o final de uma rodada de cultura reverte um declínio na sialilação que acompanha o aumento de escala da cultura e é vantajosa para os processos de cultura em grande escala.
