Abstract

"DETERMINAçãO DE SEQUêNCIA DE UM POLINUCLEOTìDEO". A seqüência de um polinucleotídeo alvo pode ser determinada por: (i) contato do polinucleotídeo alvo com uma enzima de polimerase e um dos nucleotídeos A, T(U), G e C sob condições adequadas para que a reação de polimerase tenha prosseguimento; (ii) medição do tempo que leva para a polimerase se unir a e subseqüentemente dissociar do polinucleotídeo alvo, para dessa determinar se a polimerase incorporou o nucleotídeo ao polinucleotídeo alvo; (iii) repetição opcional das etapas (i) e (ii) com nucleotídeos adicionais, para identificar dessa forma a seqüência do polinucleotídeo alvo.
