Abstract
ORGANIZAçãO SIMILAR A TECIDO DE CéLULAS E CONSTRUTOS SIMILARES A TECIDOS MACROSCóPICOS, GERADOS POR CULTURA DE MACROMASSA DE CéLULAS, E MéTODO DE CULTURA DE MACROMASSA" Trata-se de uma organização similar a tecido tridimensional de células pela cultura de elevada densidade de semeadura de células denominada como cultura de macromassa. Através da cultura de macromassa, as células podem ser levadas a se organizar em uma forma similar a tecido sem a ajuda de uma estrutura de suporte, e construtos similares a tecidos macroscópicos tridimensionais podem ser obtidos integralmente a partir das células. A organização similar a tecido e os construtos parecidos com tecidos macroscópicos podem ser gerados a partir de células fibroblásticas de origem mesenquimal (pelo menos), as quais podem ser tanto células diferenciadas quanto células tronco adultas multipotentes. Neste trabalho, a organização similar a tecido e os construtos similares a tecidos macroscópicos foram gerados a partir de fibroblastos dermais, células osteogênicas derivadas de células estromais de adipose, condrócitos, e de osteoblastos. O fator que causa a formação de tecido macroscópico é a cultura em larga escala a uma elevada densidade de semeadura de células por unidade de área ou espaço tridimensional, isto é, a cultura de macromassa obtida em uma larga escala. Nenhuma estrutura de suporte ou matriz estranha é utilizada para a geração dos tecidos, os tecidos são de origem completamente celular. Nenhum outro agente (exceto a elevad densidade de semeadura de células) que ajuda na formação dos tecidos, tais como produtos químicos de indução de tecido, fatores do crescimento de indução de tecido, substrato com propriedades especiais, cultura rotativa, etc., é empregado para a formação dos tecidos. Essas massas similares a tecidos podem ser potencialmente utilizadas como substituições de tecido no corpo humano. A organização similar a tecido pela cultura de macromassa de elevada densidade de semeadura de células também podem ser gerada ao nível microscópico.
