Abstract
MéTODO DE ENSAIO PARA DETECTAR A PROTEìNA DE PRìON INFECCIOSA EM AMOSTRA DE UM INDIVìDUO MAMìFERO, KIT PARA O USO EM UM MéTODO DE ENSAIO, E, USO DE UM ANTICORPO DE LIGAçãO A presente invenção porvê um método de ensaio para detectar a proteína de príon infecciosa em uma amostra a partir de um indivíduo mamífero, o referido método compreendendo: obter uma amostra contendo a proteína de príon a partir do referido indivíduo; contactar a referida amostra com um agente, que serve para digerir a proteína de príon não-infecciosa e ára digerir parcialmente a proteína de príon infectada de modo a produzir um ersíduo de polipeptídeo da proteína de príon: contactar a amostra digerida com um anticorpo capaz de se ligar a um polipeptídeo tendo a seqüência de aminoácido Vc(Gly-Gly-Gly-Trp)-Gly-Gly-Gly-Gly-R~1~-R~2~-His-R~3~-Gln- Trp-Asn-Lys-Pro-R~4~- Lys-Pro-Lys-Thr-R~5~-R~6~-Lys (His-R~7~ Ala-Gly) (Vc) edetectar conjugados do referido anticorpo e do referido resíduo de polipeptídeo da proteína de príon; em que a detecção dos referidos conjugados compreende a amplificação química, biológica ou bioquímica de espécies detectáveis e a detecção da espécie amplificada.
