Abstract

ANáLISES DE MICROARRANJO E MICRODISSECçãO A LASER DE TUMORES DE MAMA REVELAM GENES RELACIONADOS COM RECEPTOR DE ESTROGêNIO E VIAS. Cerca de 70% a 80% de cânceres de mama expressam receptor de estrogênio <244> (ER<244>), e os estrogênios exercem funções importantes no desenvolvimento e crescimento de tumores dependentes de hormónio. Juntamente com metástase linfonodal, o tamanho e grau histológico do tumor, o estado ER é considerado um dos fatores prognósticos em câncer de mama, e um indicador para tratamento hormonal. 147 genes e 112 genes com valor-P significativo e que possuem expressão diferencial significativa entre tumores ER+ e ER- foram identificados a partir de conjunto de dados de LCM e conjunto de dados de tecido volumoso, respectivamente. 61 genes foram considerados comuns em ambos os conjuntos de dados, enquanto 85 genes eram exclusivos ao conjunto de dados de LOM e 51 genes estavam presentes apenas no conjunto de dados de tumor volumoso. A análise de vias com 85 genes utilizando Gene Ontology sugeriu que os genes envolvidos em endocitose, geração de ceramida, cascata Ras/ERK/Ark, e via JATSTAT pode exercer funções relacionadas com ER. A definição de perfil de gene com células de tumor capturadas por LCM proporciona uma abordagem única para caracterizar e estudar as células tumorais epiteliais e para obter um conhecimento sobre as vias de sinalização associadas com ER.
