Abstract

MéTODO PARA PRODUZIR FLORES MACHO ESTéREIS E FRUTOS PARTENOCáRPICOS MEDIANTE SILENCIAMENTO GêNICO, SEQüENCIAS ASSOCIADAS E VETORES QUE AS CONTêM Método para produzir flores macho estéreis e frutos partenocárpicos por meio de silenciamento gênico que consta das seguintes etapas: a) obter a seqüência codificadora de genes homólogos para Pistillata (PI) da espécie desejada; b) analisar a expressão da seqüência obtida para verificar se é expressa de acordo com o padrão descrito para genes Pistillata; c) analisar a complementação de mutantes para o gene PI com a seqüência PI obtida da espécie desejada para comprovar que a seqüência obtida cumpre a função do gene PI; d) fabricar a construção para silenciamento gênico que contenha uma zona da seqüência codificadora de PI em um vetor de expressão em plantas; e) incorporação do vetor construído em Agrobacterium tumefaciens; f) transformar as plantas de interesse com Agrobacterium tumeifaciens modificado pelo vetor de silenciamento e seleção das plantas transformadas; g) comprovar a ausência de contaminação por Agrobacterium e corroboração das plantas transgênicas por amplificação do transgene. São revelados, ainda, os genes VvPI de Vitis vinifera variedade Cabernet sauvignon e LePI de Lycopersicon esculentum; e o uso desses genes para produzir flores macho estéreis e frutos sem sementes ou partenocárpicos. Conjuntamente, são revelados vetores de silenciamento que contêm essas seqüências ou parte delas.
