PROCESSO DE PRODUÇÃO DE LIPASES A PARTIR DE RESÍDUO AGROINDUSTRIALLIPASE PRODUCTION PROCESS FROM AGRO-INDUSTRIAL WASTE
CAMPO DA INVENÇÃOFIELD OF INVENTION
The present invention is a process for the production of lipase by filamentous fungi by solid state fermentation. Residual pies of various oilseed materials are used, with concomitant detoxification and allergenicity reduction of pies that have toxic and allergenic character and consequent valuation of biomasses.
FUNDAMENTOS DA INVENÇÃOBACKGROUND OF THE INVENTION
Although the chemical route for biodiesel production from vegetable oils is well established, the enzymatic route is still under development.
The reaction catalyzed by lipases is a promising alternative, both regarding the reduction of costs associated with the glycerin purification generated and the fact that it represents a strong reduction of the environmental impact for disposal of this residue.
On the other hand, the application in industry of lipase catalyzed enzymatic processes still presents the drawback of the high cost for enzyme production.
Lipases are enzymes that have great versatility of application and can be used in different industries, such as food, pharmaceutical, chemical, among others, as they hydrolyze ester-carboxylic bonds present in triglycerols, releasing fatty acids and glycerol, and also , act reversibly in transesterification reactions to obtain biodiesel.
A fermentação em estado sólido (FES) reproduz processos microbiológicos naturais. Thus, several value-added products have been produced by this technique, making use of raw materials from nature. The advantage of FES is the possibility of using low cost raw materials as a culture medium for the microorganism, simplicity of the fermentation medium, lower probability of contamination and the possibility of obtaining a more concentrated extracellular enzyme (Martins, 2001; Alonso, 2001 ).
The selective process of solid substrates employed for the production of enzymes by FES depends on several factors, including cost and availability of the raw material, enabling the use of a wide variety of industrial waste.
US Patent Document US 2004/0055953, cited as reference, discloses a process for preparing an enzymatic composition using fat and other nutrient rich household and industrial effluents produced by the fungus Penicillium restrictum.
In Brazil, several researches have been developed based on the use of agroindustrial residues resulting from biodiesel production processes. Processes of Iipases with different activities, obtained by FES with Burkholderia cepacia (Fernandes, 2007) on substrates such as wheat husk meal, sunflower seed meal and corn cake are described.
Brazilian patent application Pl 0703290, cited as reference, discloses a method of treating agroindustrial tailings, especially castor bean pies, with the aim of detoxification and reduction of allergicity with the production of lipase enzymes.
Castor bean cake is used as a substrate for the growth of microorganisms by FES with lipase-producing filamentous fungi. The treated waste, after sterilization, can be disposed of or used as animal feed or as fertilizer.
With the governmental incentives for the cultivation of various oilseeds, aiming at obtaining biodiesel, it is expected the increase in the formation of its sub or co-products, such as the residual pies from the oil extraction stage. Certain pies, such as the Jatropha curl, are allergenic and toxic, having restricted application as fertilizers or animal feed, for example.
Therefore an object of the present invention is the production of lipases with different activities, produced at low cost, from industrial or agroindustrial waste, in particular by employing substrates widely available in the national territory, such as jatropha pie, castor cake, soy pie, babassu pie, among others.
Jatropha curcas L. belongs to the same family as castor and cassava. The seed is relatively large and, when dry, has a resinous fracture. The fruit is being considered an agricultural option for the northeast region, as it is a native species, demanding in sunshine and with strong drought resistance and has the possibility of use for the production of biodiesel. After oil extraction, the cake still contains approximately 8% oil, which can be extracted with organic solvents, and the bran is rich in nitrogen, phosphorus and potassium.
Another object of the present invention is to provide a process for reducing the toxicity of such pies, so that the aforementioned applications become possible. Thus, in addition to adding value to a byproduct, we also obtain an input for the process of obtaining biodiesel.

The selection process of solid substrates depends on several factors. In the solid state fermentation process, as mentioned above, the substrates not only provide nutrients for the culture of microorganisms, but also serve as support for the growth of microbial cells. Material particle size and moisture content, as well as medium temperature and pH, are critical factors for cell growth and substrate activity.
The FES conditions used in the present invention were similar to those reported in the Brazilian patent application PI0703290-0, but different substrates and microorganisms were employed, thus obtaining lipase enzymes with different enzymatic activities.
Os esporos foram obtidos em meio de cultura contendo a seguinte composição, tendo por base 100 mL de meio de cultura: amido solúvel - 2,0 g; MgSO4JH2O - 0,025 g; KH2P04 - 0,05 g; CaCO3 - 0,5 g; extrato de levedura - 0,1 g; óleo de oliva - 1,0 g; goma guar - 1,0 g, e incubados a 30°C por 7 dias. Para aplicação de 106 a 108 esporos por grama de substrato, os esporos foram suspensos em solução tampão de fosfato de sódio (100 mM, pH 7).The spores were obtained in culture medium containing the following composition, based on 100 ml culture medium: soluble starch - 2.0 g; MgSO 4 JH 2 O - 0.025 g; KH2 PO4 - 0.05 g; CaCO3 - 0.5 g; yeast extract - 0.1 g; olive oil - 1.0 g; guar gum - 1.0 g, and incubated at 30 ° C for 7 days. For application of 106 to 108 spores per gram of substrate, the spores were suspended in sodium phosphate buffer solution (100 mM, pH 7).
The spores were applied directly to the substrate to conduct the fermentation, or first applied to a small amount of substrate (between 5% and 15% of the mass used in the fermentation) for pre-inoculation, and then to the pre-inoculum. This pre-inoculum with the grown cells was inoculated into a larger amount of substrate for fermentation.
The humidity was adjusted between 30% and 80% m / m and the temperature maintained between 25 ° C and 35 ° C.
In some cases, in order to increase lipase production, the pies were supplemented with other agroindustrial tailings, such as molasses (rejects from sugar cane production) and glycerin (rejects from biodiesel production from vegetable oils and animal tallow). concentrations between 0% and 10%.
Table 1 presents some comparative results of lipase production by FES, using castor bean cake (PI 0703290) and using babassu cake as raw material.

Os resultados são apresentados na Tabela 2.The use of agroindustrial waste as a supplement during fermentation is exemplified by the cultivation of Rhizopus sp. in babassu pie. The results are presented in Table 2.

Jatropha cake was an excellent raw material for the production of lipases, since higher production values (139 U / g) were obtained compared to those reported in PI0703290 (87 U / g), using if the same microorganism. Protease production, undesirable to reduce lipase stability, was well below that of the enzymes of interest, and N-acetylglycosamine concentration portrays an indirect measure of filamentous fungus growth in the system, showing that it is associated with enzyme production. .
The photographs of a stereoscopic microscope, shown in Figure 2, prove that the P. simplicissimum strain was able to grow completely in the pie, with consequent enzyme production and detoxification of the same pie. The microscopic aspect shows the development of P. simplicissimum on jatropha cake after 24 h (a) and 96 h (b) fermentation.
The proposed process also promotes the reduction of allergenicity of allergenic pies, so that they have potential for application as fertilizers and animal feed. As an example, Table 3 shows the concentrations of phorbol esters, a class of toxic compound present in jatropha cake during fermentation.
