Abstract
VETOR DE DNA RECOMBINANTE, MÉTODO PARA PRODUÇçO DE PLANTAS TOLERANTES A ESTRESSES AMBIENTAIS E SEUS USOS. As plantas são influenciadas por um grande número de fatores ambientais bióticos e abióticos e recorrentemente estresses abióticos são mais graves, afetando todas as funções da planta, resultando em redução do crescimento e da produtividade. Nesse sentido, a identificação e a compreensão dos mecanismos de tolerância abióticos são fundamentais no desenvolvimento de novas cultivares tolerantes à seca. Dessa forma, a presente invenção proporciona um método de produção de plantas que contém em suas células uma seqúência de nucleotídeos de cana-de-açúcar e a superexpressão desse gene leva a planta em questão à maior tolerância a estresses abióticos. De uma forma mais ampla, descreve-se um polinucleotídeo que codifica a proteína de cana-de-açúcar, o qual é expresso por um promotor e um terminador que funcionam em plantas. More broadly, a polynucleotide encoding the sugarcane protein which is expressed by a plant-working promoter and terminator is described. It is found that this gene confers tolerance to different abiotic stresses.

Nos últimos anos tem-se obtido grande avanço nas tecnologias para a transformação de plantas e as pesquisas têm se voltado para a necessidade de desenvolver eficazes métodos de transformação genética das principais espécies de interesse experimental e econômico. A agrotransformação tem ganhado grande aceitação pela facilidade do método de transformação e a alta taxa de plantas transgênicas geradas a partir desse método. Por outro lado, o tabaco tem sido utilizado amplamente como ferramenta de transformação gênica pela facilidade de transformação e rápida obtenção de gerações transformadas. Sementes de tabaco selvagens e transgênicas contendo a SEQ ID NO: 1 foram germinadas por 16 dias em placas de Petri contendo meio MS e pH 5,8. As plântulas foram transferidas para vasos de 500 ml contendo Plantmax HT (Eucatex, Brasil) durante 3 semanas e mantidas em câmara de crescimento a 23 0C com fotoperíodo 16/8 h luz/escuro, sendo adubadas semanalmente com solução nutritiva (EPPQ, Brasil). Cada planta foi irrigada com 70 ml de uma solução de 175 mM de NaCl por 10 dias e, em seguida, irrigada por três dias com água pura para a recuperação.Wild and transgenic tobacco seeds containing SEQ ID NO: 1 were germinated for 16 days in Petri dishes containing MS medium and pH 5.8. Para o estresse hídrico as plântulas foram irrigadas com 70 ml de manitol 200 mM, — por"10 dias, e depois foramTrrigadas novamente com água por três dias, como descrito por . Zhang et al. Essas mesmas plantas foram usadas nas análises mostradas nos exemplos 7 e 8 e estão em concordância com a melhor performance das plantas em quanto ao parâmetros fotossintéticos. Da mesma forma, esse fenótipo observado na Figura 7 também é um reflexo dos maiores teores de água mostrados nos exemplos 9 e 10, bem como com a maior massa seca mostrada nos exemplos 11 e 12.These same plants were used in the analyzes shown in examples 7 and 8 and are in agreement with the best performance of the plants regarding photosynthetic parameters.
