Abstract
DIAGNÓSTICO DA SEPTICEMIA POR ESPECTROMETRIA DE MASSA. A presente invenção refere-se principalmente à identificação de patógenos por espectrometria de massa em culturas de sangue provenientes de infecçoes na corrente sanguínea (septicemia). A invenção proporciona um método com o qual patógenos microbianos podem ser separados na forma purificada a partir de sangue após um cultivo relativamente breve num frasco de hemocultura, sem qualquer interferência de proteínas humanas ou frações residuais de partículas sanguíneas, como eritrócitos e leucócitos, e podem ser diretamente identificados por medição por espectrometria de massa de seus perfis proteicos. O método é baseado no uso de tensoativos relativamente fortes para destruir as partículas sanguíneas, dissolvendo as membranas celulares fracas e a maior parte das estruturas internas das partículas sanguíneas, apesar do fato dos tensoativos serem considerados fortes inibidores de ionização em MALDI e outros processos de ionização necessários para medições por espectrometria de massa. Esse método permite que patógenos desconhecidos sejam obtidos em sua forma pura por centrifugação ou filtração e sejam identificados no nível taxonômico de espécie ou subespécie. Problemas com DNA provenientes de altas concentrações de leucócitos podem ser resolvidos por meios especiais. Após o cultivo suficiente, a identificação em um laboratório de espectrometria de massa leva apenas meia hora.
