Abstract

MEIOS DE CULTURA DE TORTA RESIDUAL DE MAMONA PARA PRODUÇçO SIMULTÂNEA DAS ENZIMAS FITASE E TANASE, E DETOXIFICAÇçO DA TORTA DE MAMONA, PELO MICROORGANISMO PAECILOMYCES VARIOTII ATRAVÉS DA FERMENTAÇAO SàLIDA, ENZIMAS OBTIDAS E SEUS USOS. O presente invento visa à produção simultânea de tanase e fitase em torta residual de mamona em fermentação em estado sólido com Paecilomyces variotii, além da diminuição da concentração de ricina após a fermentação, promovendo assim sua detoxificação. O uso de enzimas na alimentação animal é conhecido e está sendo bem. explorado. A maior dificuldade de expandir o uso de enzimas ainda é o custo de produção. O uso dos resíduos para alimentação animal representa uma alternativa viável para este setor, assim como para a produção de biocatalizadores através da fermentação sólida. Esta invenção determinou valores ótimos na constituição do meio de cultura que utiliza torta residual de mamona para produção das enzimas. fitase e tanase através da fermentação sólida pelo microorganismo Paecilomyces variotii de forma a obter-se a maior quantidade de enzima produzida e suas maiores atividades. Adicionalmente, o presente invento permite a detoxificação da torta residuar de mamona, devido à degradação da ricina, seu maior agente tóxico, e, dessa forma, tomando o resíduo um ingrediente interessante para uso em ração animal.MEDIUM RESIDUAL PIE CULTURE FOR SIMULTANEOUS PRODUCTION OF THE ENZYMES PHYTASE AND TANASE, AND DETOXIFICATION OF THE MAMONA PIE BY MICROORGANISM PAECILOMYCES VARIOTII THROUGH FEMENTATION AND SEASED SENSION. The present invention aims at the simultaneous production of tannase and phytase in castor bean residual cake in solid state fermentation with Paecilomyces variotii, as well as the reduction of ricin concentration after fermentation, thus promoting its detoxification. The use of enzymes in animal feed is known and is being well. explored. The biggest difficulty in expanding enzyme use is still the cost of production. The use of animal waste represents a viable alternative for this sector, as well as for the production of biocatalysts through solid fermentation. This invention has determined optimal values in the constitution of the culture medium utilizing residual castor bean cake for enzyme production. phytase and tanase through solid fermentation by Paecilomyces variotii microorganism in order to obtain the highest amount of enzyme produced and its largest activities. Additionally, the present invention allows the detoxification of castor bean cake due to the degradation of ricin, its major toxic agent, and thus making the residue an interesting ingredient for use in animal feed.

A partir de 48 horas de incubação a atividade enzimática diminuiu para 249 U/mL após 72 horas e manteve-se estatisticamente constante. Com objetivo de otimizar a produção das enzimas tanase e fitase na torta de mamona, foram analisados a influência da concentração do ácido tânico e volume da solução salina no meio de produção enzimática. Utilizou-se a técnica do delineamento composto central rotacional (DCCR), apresentando 8 pontos fatoriais (p.f.), 6 pontos axiais (p.a.) e 3 pontos centrais (p.c.) totalizando 17 ensaios (23 p.f. + 6 p.a. + 3 p.c. = 17). As 3 variáveis independentes foram: umidade relativa do ar (%) em Câmara Climática (Nova Ética-Modeio 420/CLDTS 300); concentração de ácido tânico, que foi expresso em porcentagem em relação ao peso total do meio (p/p); e o volume da solução salina, que foi expresso em porcentagem em relação ao peso total do meio (v/p).
