Abstract
On a utilisé la transcription in vivo de l'ARN polymérase I dans des cellules transfectées de manière transitoire par de l'ADN, pour l'expression de molécules d'ARNv de la grippe codant pour la chloramphénicol-acétyltransférase (CAT) dans une orientation antisens. La surinfection à virus grippal a servi à obtenir une ARN polymérase virale ainsi que d'autres protéines destinées à la conversion par transcritpion d'ARNv à brin moins en molécules d'ARNm viral à brin plus exprimant l'activité de CAT. On a utilisé ce système pour une analyse par des échanges nucléotidiques ainsi que des délétions et insertions des deux segments terminaux de la séquence d'ARNv constituant de manière coopérative la structure du promoteur d'ARNv. On a formé plusieurs mutants présentant des taux d'expression notablement accrus par rapport aux niveaux de type sauvage, que l'on peut également encapsider et faire passer en série dans un virus de descendance. Les données obtenues pour les mutations dans divers éléments promoteurs permettent l'élaboration d'un modèle de structures consécutives de promoteur d'ARNv bicaténaire pour lier la polymérase virale et initier la synthèse de l'ARN. Des préparations du virus atténué de la grippe à des fins de vaccination comprennent un seul segment recombiné présentant une ou des mutation(s) favorisante(s) du promoteur aux fins de surexpression d'un produit génique propre ou étranger, lequel sert en même temps, étant donné sa sur-réplication, à diminuner le nombre de segments de la ribonucléoprotéine du virus auxiliaire. On a fait passer à nouveau les mêmes virus par une étape de clivage de ribozyme agissant au niveau d'un des segments viraux auxiliaires, laquelle supprime cette fonction et cette structure vitales, avec des taux élevés, de la descendance du virus. Les virus atténués obtenus ont une interaction avec leurs cellules cibles en un seul cycle d'infection avortée et ils sont incapables de produire une descendance virale.
