La présente invention concerne des compositions, notamment à visées diététique (complément alimentaire), cosmétique ou médicale, à base de produits naturels, c'est-à-dire de produits se rencontrant dans des végétaux ou extraits de tels végétaux.
On assiste actuellement à un développement considérable dans ce domaine sur la base de composés très divers.
Les pectines de fruits sont utilisées depuis longtemps comme gélifiants ou 1o épaississants dans l'industrie alimentaire. Par ailleurs, on sait maintenant que les pectines sont capables de lier les métaux lourds et leurs radionucléides et d'en assurer l'excrétion hors du corps.
Les caroténoïdes sont aussi connus et utilisés pour leurs propriétés pigmentaires et anti-oxydantes (anti-radicaux libres) dans le domaine des crèmes et compléments cosmétiques (gélules par exemple) solaires. On peut citer l'alpha-et le bêta-carotènes et le lycopène, le plus puissant de ces composés, qui est extrait de la tomate (oléorésine de tomate).
Le brocoli et ses extraits sont connus pour leurs propriétés favorables dans la carcinogénèse et la dégénérescence des cellules.
GB-B-1 041 890 décrit des émulsions utilisables en médecine humaine, par exemple comme véhicule pour l'administration de substances médicamenteuses par le biais de l'alimentation. Ces émulsions peuvent comprendre d'une part une substance telle qu'une vitamine liposoluble, un dérivé
liposoluble d'une vitamine hydrosoluble, une caroténoïde ou leurs mélanges, et d'autre part un émulsifiant non ionique et un agent mouillant ionique, la phase aqueuse pouvant comprendre un hydrocolloïde tel que la pectine.
SU-A-1 78fi 531 décrit une boisson réalisée à partir de jus de plusieurs fruits et pouvant donc contenir entre autres choses du carotène et de la pectine.
M. Yu Tamova et al. (Database FSTA, International Food Information Service (IFIS), Frankfurt/Main, Allemagne, 97-1-02-a0017, XP 002071238) ont étudié la capacité de diverses solutions de pectine à fixer le nickel et le plomb.
Les auteurs rapportent que l'ajout d'une solution de carotène n'affecte pas de manière significative la fixation du nickel et du plomb. L'objectif est le développement de compositions diététiques pour la fixation des métaux lourds La déposante a trouvé de manière surprenante et inattendue que les pectines pouvaient être très avantageusement combinées à des caroténoïdes, en particulier le lycopène, ou à du brocoli ou des extraits de brocoli, et de préférence à ces deux types de composés, et que cette combinaison permettait aux différents composés d'assurer leurs fonctions ci-dessus (fixation et élimination des métaux lourds, action anti-oxydante et anti-radicalaire et action bénéfique sur la régénération cellulaire) dans des conditions plus favorables que lorsqu'ils sont seuls.

Sur le plan cosmétique, l'association agit de façon synergique sur les aggressions contre les cellules de l'épiderme par la pollution et les rayons solaires.
La présente invention a donc pour objet une composition, notamment à
visées diététique, médicale ou cosmétique, comprenant au moins un hétéropolysaccharide, de préférence estérifé, notamment un acide polygalacturonique ou ester, de préférence une pectine, et un composé choisi dans le groupe consistant en caroténoïdes, de préférence lycopène, et glucosinolate et/ou sulphoraphane, notamment extrait végétal, en particulier de crucifère, de préférence du brocoli, en contenant, et mélanges de ceux-ci.

Les compositions bi-composants comprennent avantageusement en - de 40 à 90, de préférence de 60 à 80 d'hétéropolysaccharide, e.g. pectine, et de 10 à 60, de préférence de 20 à 40 d'extrait contenant du lycopène, (un tel extrait contient de 3 à 6 % de lycopène), ou - de 10 à 60, de préférence de 20 à 40 d'extrait contenant glucosinolate et/ou sulphorophane, e.g. extrait de brocoli (un tel extrait contient de 2000 à 5000 ppm/kg).
En cas de composition à trois composants, on a avantageusement en %:
- de 40 à 90, de préférence de 60 à 80 d'hétéropolysaccharide, e.g. pectine, et - de 1 à 20, de préférence de 5 à 10 d'extrait contenant du caroténoïde, e.g.
lycopène, (un tel extrait confient de 3 à 6 % de caroténoïde), et - de 5 à 40, de préférence de 10 à 35 d'extrait contenant glucosinolate et/ou sulphorophane, e.g. extrait de brocoli (un tel extrait contient de 2000 à 5000 ppmlkg).
La présente invention a vocation à s'appliquer à la fois au domaine thérapeutique et au domaine non thérapeutique en fonction des règlementations en vigueur, des modes d'utilisation et des doses administrées. Elle peut notamment être utilisée dans le domaine non thérapeutique à la fois en cosmétique et en diététique, l'objectif étant alors essentiellement préventif et l'usage laissé au libre choix de l'utilisateur. Elle concerne donc aussi ces produits cosmétiques d'hygiène buccodentaire comprenant une composition selon l'invention et les excipients habituels du type de produit concerné (e.g. bain de bouche, dentifrice, solution ou gel dentaire). L'incorporation des compositions selon l'invention, à titre de principe actif associé, dans des produits d'hygiène buccodentaire entre aussi dans le cadre de la présente invention.
Elle concerne encore une méthode de traitement cosmétique comprenant l'application externe ou buccale à l'homme, d'un produit cosmétique externe ou d'hygiène buccodentaire selon l'invention.

On peut aussi utiliser un extrait végétal, notamment de tomate, contenant naturellement du lycopène que l'on enrichit par un apport de lycopène ou autre caroténoïde purifié ou synthétisé.
Suivant un mode préféré, ie brocoli frais est traité par cryogénie, puis lyophilisé.

A noter que le brocoli est aussi une source de bëta-carotène et donc que l'utilisation d'un extrait approprié de brocoli va avantageusement supplémenter la composition selon l'invention en caroténoïde.
De même, l'on pourra choisir de mettre à profit les autres composés chimiques compris dans le brocoli et connus comme ayant une activité anti-carcinogène ou contre la dégénérescence cellulaire : flavonoïdes et dérivés polyphénoliques, autres dérivés de glucosinolates (e.g. glucobrassicine), S-méthylcystéine sulfoxide et ses dérivés.
De manière générale, pour le caroténoïde, notamment le lycopène, et le brocoli, on préférera utiliser des extraits naturels de végétaux, suivant le procédé

9 Procédé d'obtention Le brocoli est un légume appartenant à la famille des crucifères, qui renferme des substances très sensibles au phénomène d'oxydoréduction ainsi qu'au phénomène de lyse enzymatique. Comme ses consoeurs de la famille des crucifères, il a la particularité de contenir des glucosides soufrés (glucosinolates) qui sous l'attaque de certaines enzymes (myrosinases, catalases) transforment ce composé en isothyocyanates ou dérivés (sulphoraphanes) qui sont responsables de l'odeur très caractéristique de cette famille. Ces composés sont très volatils et s'oxydent rapidement, rendant inefficace toute préparation de plante sèche.
La déposante a pu mettre au point un procédé d'obtention approprié.
Le principe de ce procédé d'obtention consiste à
- utiliser du brocoli frais et à le congeler de façon à éclater les cellules - prébroyer ce végétal dans un appareil approprié
- effectuer un cryobroyage en utilisant toute source cryogénique, e.g. azote liquide - on obtient une poudre micronisée qui est ensuite lyophilisée.
La poudre de brocoli est ensuite récupérée et mélangée aux autres ingrédients de la préparation.
L'eau d'évaporation du piège frigorifique du lyophilisateur est ensuite récupérée, elle est concentrée par osmose inverse à une température ne dépassant pas 15°C. Le volume de solution concentrée est ramené à la proportion de 1/100 (1 litre de concentré pour 100 litres d'eau osmosée).
Cette eau renferme une partie des substances volatiles de la plante ; elle est ensuite réintroduite dans ia préparation finale selon le procédé décrit ci-dessous.
Préparation du mélange final Le mélange de pectines de diverses estérifications est effectué dans un mélangeur à bandes (type Comia Fao) ou tout autre type de mélangeur ; lorsque le mélange est homogène, on ajoute par petites fractions l'oléorésine de tomate, puis l'eau osmosée de brocoli. Cette adjonction a la particularité de créer un réseau au niveau de la fibre qui très avantageusement conduit à une encapsulation des substances actives (ce qui permet notamment d'assurer !a 5 biodisponibilité des composés additionnés {e.g. caroténoïde etlou glucosinolate et/ou sulphoraphane, etc.) et leur transport) ; on rajoute ensuite la poudre de brocoli lyophilisée, puis en final une substance type silice colloïdale (e.g.
AEROSIL) permettant de déshydrater la préparation.
Le mélange ainsi préparé est prêt pour les différentes applications.

On a évalué les propriétés préventives et curatives d'une composition selon l'invention (comprenant 70,8 % de pectine, 5,2 % d'extrait concentré de tomate et 24 % de lyophilisat de brocolis) vis-à-vis du mercure chez le rat.
Les propriétés préventives ont été évaluées en administrant simultanément HgCl2 et la composition. Les propriétés curatives ont été évaluées en administrant HgClz seul aux rats pendant 2 semaines, puis en traitant par la composition pendant les 2 semaines suivantes.

On a administré des doses de mercure non léthales et il n'a pas été
constaté de mortalité directe. II a été par contre constaté une mortalité fors d'anesthésies pratiquées sur certains rats non traités par la composition. La composition a par contre protégé les rats traités ; l'action du mercure sur l'anesthésie a été inhibée par la composition.
On a mesuré les concentrations sanguines en mercure à J15 pour les animaux des groupes 2 et 3, puis à J28 pour les animaux des 4 groupes. Les valeurs moyennes sont données ci-après - àJlS:
Groupe 2 : 308,0 Ng Hg/l Groupe 3 : 147,33 Ng Hg/l soit une réduction de 51,47 % en présence de la composition selon l'invention (significance p< 0,001 ) - àJ28:

The applicant was able to develop an appropriate procurement process.
The principle of this process of obtaining consists in - use fresh broccoli and freeze it so that the cells burst - pre-grind this plant in a suitable device - perform a cryogenic grinding using any cryogenic source, eg nitrogen liquid - a micronized powder is obtained which is then lyophilized.
The broccoli powder is then collected and mixed with the others ingredients of the preparation.
The evaporation water from the freeze dryer freeze trap is then recovered, it is concentrated by reverse osmosis at a temperature not not exceeding 15 ° C. The volume of concentrated solution is brought back to the proportion of 1/100 (1 liter of concentrate for 100 liters of RO water).
This water contains part of the volatile substances of the plant; she is then reintroduced into the final preparation according to the process described above below.
Preparation of the final mixture The mixture of pectins of various esterifications is carried out in a belt mixer (Comia Fao type) or any other type of mixer; when the mixture is homogeneous, the oleoresin of tomato, then the broccoli osmosis water. This addition has the distinction of creating a fiber network which very advantageously leads to a encapsulation of active substances (which in particular ensures! a 5 bioavailability of the added compounds (eg carotenoid and / or glucosinolate and / or sulphoraphane, etc.) and their transport); then add the powder of freeze-dried broccoli, then finally a colloidal silica-like substance (eg AEROSIL) to dehydrate the preparation.
The mixture thus prepared is ready for the various applications.

We evaluated the preventive and curative properties of a composition according to the invention (comprising 70.8% pectin, 5.2% concentrated extract of tomato and 24% broccoli lyophilisate) vis-à-vis mercury in rats.
The preventive properties were assessed by simultaneously administering HgCl2 and the composition. The healing properties have been assessed by administering HgClz alone to rats for 2 weeks, then treating with the composition for the 2 following weeks.

Non-lethal doses of mercury were administered and it was not observed direct mortality. On the other hand, there has been a marked mortality anesthesia performed on certain rats not treated with the composition. Blood mercury concentrations were measured at D15 for the animals in groups 2 and 3, then on D28 for animals in the 4 groups. The average values are given below - at JlS:
