Abstract
Cette invention concerne des cellules capables de compenser au moins partiellement la fonction E2A d'un adénovirus chez lequel cette fonction fait défaut. Ces cellules comprennent un acide nucléique qui code l'E2A d'un adénovirus, ou une partie fonctionnelle, un dérivé et/ou un analogue de celle-ci, et qui est intégré dans le génome de la cellule. De préférence, la cellule possède un acide nucléique d'E2A qui est dérivé d'un adénovirus sensible à la température tel qu'un adénovirus ts125. Cette invention concerne également des procédés de production d'une particule d'adénovirus contenant un vecteur d'adénovirus qui présente un effacement fonction de l'E2A. Ces procédés consistent à préparer une cellule, comme décrit précédemment, dans laquelle le vecteur d'adénovirus a été effacé de manière fonctionnelle, à cultiver cette cellule et à récolter les particules de virus. L'effacement fonctionnel peut comprendre un effacement d'une partie au moins de l'acide nucléique codant l'E2A. D'après ce procédé, l'acide nucléique codant l'E2A d'adénovirus dans le génome de la cellule ne présente de préférence pas de chevauchement de séquence avec le vecteur, ce qui entraîne une réplication de l'adénovirus compétent et/ou la formation d'un vecteur d'adénovirus possédant la fonction E2A. dans ce procédé, le vecteur d'adénovirus présente également et de préférence un effacement fonctionnel de l'acide nucléique codant la région E1. Cette invention concerne également des vecteurs d'adénovirus présentant un effacement fonctionnel de l'E2A d'adénovirus, de préférence un effacement d'une partie au moins de l'acide nucléique codant l'E2A. Cette invention concerne aussi des préparations à base de vecteur d'adénovirus contenant des particules d'adénovirus, le vecteur d'adénovirus présentant un effacement fonctionnel de l'E2A. Ce vecteur d'adénovirus présente de préférence un effacement de l'acide nucléique codant les protéines de la région E1, et peut être exempt de vecteurs d'adénovirus comprenant une fonction E2A. Cette invention concerne enfin des procédés permettant de fournir aux cellules d'un individu un acide nucléique présentant un intérêt et ceci sans courir le risque d'une administration simultanée d'un vecteur d'adénovirus compétent et de réplication, lesquels procédés consistent à administrer à un individu l'une des préparations susmentionnées.
