Abstract

La présente invention concerne une méthode améliorée permettant l'isolement différentiel des cellules souches mammaires de souris et de cellules souches unipotentes (CFU). La méthode comporte la déplétion des cellules non épithéliales de tissus mammaires de souris fraîchement dissociés par incubation avec une composition d'anticorps contenant des anticorps spécifiques des CD45, Ter119, CD31 et éventuellement de la CD140a. Après la formation de conjugués entre les cellules mammaires non épithéliales et les anticorps spécifiques des CD45, Ter119, CD31 et, éventuellement de la CD140a, les conjugués des cellules sont retirés et les cellules épithéliales restantes sont ensuite incubées avec une composition d'anticorps contenant des anticorps spécifiques des CD24 et CD49f. Après la formation de conjugués entre les cellules épithéliales et les anticorps spécifiques des CD24 et CD49f, les cellules souches mammaires de souris et les cellules CFU dont le luminal est restreint peuvent être isolées de façon différentielle. L'invention concerne également les trousses permettant de mener à bien cette méthode et les préparations de cellules préparées en suivant cette méthode.
