Abstract

La présente invention concerne un vecteur innovant pour la transformation de végétaux. Le vecteur est un cosmide qui a une longueur totale inférieure ou égale à 15 kb et qui possède les propriétés suivantes : (1) le vecteur contient une origine de réplication du plasmide IncP mais il ne contient pas d'origine de réplication d'autres plasmides ; (2) le vecteur contient le gène trfA1 du plasmide IncP ; (3) le vecteur contient oriT du plasmide IncP ; (4) le vecteur contient le gène incC1 du plasmide IncP ; (5) le vecteur contient un site cos d'un phage lambda et le site cos se trouve à l'extérieur de l'ADN-T ; (6) le vecteur contient un gène de résistance à un médicament qui peut être exprimé chez Escherichia coli et chez une bactérie appartenant au genre Agrobacterium ; (7) le vecteur contient une séquence de bordure droite de l'ADN-T d'une bactérie appartenant au genre Agrobacterium ; (8) le vecteur contient une séquence de bordure gauche de l'ADN-T d'une bactérie appartenant au genre Agrobacterium ; (9) le vecteur contient un gène marqueur sélectif de la transformation des végétaux, le gène marqueur sélectif se trouvant entre les séquences mentionnées en (7) et (8) et pouvant être exprimé dans un végétal ; et (10) le vecteur contient un site de reconnaissance d'enzyme de restriction pour le clonage d'un gène étranger, le site de reconnaissance d'enzyme de restriction se trouvant entre les séquences mentionnées en (7) et (8).
