Abstract

L'invention porte sur des dispositifs d'hybridation biomoléculaire comprenant un substrat avec une surface attachée de manière permanente et covalente de groupes fonctionnels et une monocouche adsorbée d'oligonucléotides simple brin non modifiés, dont la totalité sont de 10 à environ 24 bases de longueur en tant que film saturé d'oligonucléotides contraints sur la surface par l'intermédiaire d'un contact adsorptif de surface de phosphate non covalent direct de sensiblement tous les groupes phosphate de chaque oligonucléotide. Les oligonucléotides contraints sont efficaces pour s'hybrider de manière dissociable à un acide nucléique simple brin complémentaire avec un appariement de bases asymétrique, non hélicoïdal et sans dissociation des oligonucléotides à partir de la surface du dispositif. L'invention porte également sur des procédés pour hybrider les acides nucléiques cibles à l'état de solution à des acides nucléiques sondes et pour identifier une séquence nucléotidique à laquelle une protéine de liaison à un nucléotide se lie à l'aide des dispositifs d'hybridation biomoléculaire.
