Abstract

L'invention concerne des Fabs chimériques stabilisés dérivés d'un Fab chimérique parent ayant une chaîne légère lambda. Les Fabs chimériques stabilisés comprennent une construction polypeptidique de chaîne lourde d'immunoglobuline du Fab chimérique parent, ayant une séquence CH1 et une séquence VH, ainsi qu'une construction de chaîne légère chimérique Vlambda-Ckappa. La séquence Vlambda de la construction de chaîne légère chimérique correspond à celle du Fab chimérique parent, et comprend une ou plusieurs modifications d'acides aminés stabilisantes qui augmentent la stabilité thermique du Fab chimérique stabilisé par rapport au Fab chimérique parent. Les Fabs stabilisés sont utiles en tant que polypeptides thérapeutiques, ou peuvent être utilisés pour préparer des constructions d'anticorps dans d'autres formats. Les Fabs chimériques stabilisés peuvent également être utiles généralement pour augmenter la stabilité d'anticorps ayant des chaînes légères lambda.
