Abstract

La présente invention concerne des méthodes pour la détermination et la manipulation d'épitopes immunodominants par l'intermédiaire respectivement d'une cartographie et d'une optimisation des motifs de modification de transglutaminase contenant de la glutamine. L'invention révèle que les motifs de modification de transglutaminase contenant de la glutamine donneurs d'acyle d'un peptide composent les séquences d'épitopes immunodominants reconnues par l'immunité et correspondent à celles-ci, et la réactivité de la transglutaminase des motifs est en corrélation positive avec leur immunogénicité. L'invention concerne en outre une approche pour la conception d'un immunogène, l'immunogénicité d'une protéine pouvant être manipulée par des mesures de génie protéique modifiant la réactivité de la transglutaminase comme la substitution/mutation, délétion, ou insertion d'acides aminés, l'introduction directe/la création de résidus/motifs de glutamine, ou toute combinaison de ceux-ci.
