Description
Verfahren zur Reinigung von Extrakten segualhormonartig wirkender Stoffe. Die aus den verschiedenen Ausgangs materialien (Sexualorganen, Harn, Faeces, pflanzlichen Organismen) gewonnenen Roh extrakte sexualhormonartiger Stoffe ergeben nach den bisher bekannt gewordenen Reini gungsmethoden unbefriedigende Ausbeuten an reinem Hormon.
Es wurde gefunden, dass durch mehrmali ges Ausziehen einer wässerig methylalkoho- lischen Lösung der Extrakte mit Petroläther in der Weise, dass die methylalkoholische Lö sung immer weiter mit Wasser verdünnt wird, so dass das Sexualhormon begleitende Stoffe in Form von chemischen Molekular- und Addi tionsverbindungen des Petroläthers in die Petrolätherschicht übergehen, ein Produkt er halten wird, welches beim kastrierten Tier die Geschlechtsorgane zur Entwicklung und zum Wachstum bringt.
Man verfährt zum Beispiel wie folgt Man löst rohes Hormonöl in Methylalko hol und gibt so viel Wasser hinzu, dass die Lösung etwa 70'/o Methylalkohol enthält. Diese Lösung wird erschöpfend mit Petrol- äther extrahiert, alsdann die methylalkoho- lische Lösung durch weiteren Zusatz von Wasser bis auf etwa 60 % Methylalkohol- gehalt gebracht,
worauf erneut mit Petrol- äther extrahiert wird. Beim dritten Male bringt man vor der Extraktion mit Petroläther die Verdünnung der methylalkoholischen Lö sung auf etwa 50'/o Methylalkohol.
Bei der Behandlung mit dem Petroläther bilden sich zwischen den Begleitstoffen des Hormons und dem Petroläther Additions- und Molekularverbindungen, die in den Petrol- äther übergehen. Das Hormon dagegen ver bleibt in der wässerig - methylalkoholischen Lösung, nach deren Eindunsten man das Hormon in einer Ausbeute von 95-1000/0 mit höherer Reinheit wieder erhält.

The raw extracts of sex hormone-like substances obtained from the various starting materials (sexual organs, urine, faeces, plant organisms) result in unsatisfactory yields of pure hormone according to the previously known cleaning methods.
It has been found that by repeatedly drawing out an aqueous methyl alcoholic solution of the extracts with petroleum ether in such a way that the methyl alcoholic solution is diluted more and more with water, so that the substances accompanying the sex hormone in the form of chemical molecular and addition compounds the petroleum ether pass into the petroleum ether layer, a product he will keep, which brings the genital organs to development and growth in the castrated animal.
One proceeds, for example, as follows: Dissolve crude hormone oil in methyl alcohol and add so much water that the solution contains about 70% methyl alcohol. This solution is exhaustively extracted with petroleum ether, then the methyl alcohol solution is brought to about 60% methyl alcohol by further addition of water,
whereupon it is extracted again with petroleum ether. The third time, before the extraction with petroleum ether, the methyl alcoholic solution is diluted to about 50% methyl alcohol.
During the treatment with petroleum ether, addition and molecular compounds are formed between the accompanying substances of the hormone and the petroleum ether, which are transferred to the petroleum ether. The hormone, on the other hand, remains in the aqueous - methyl alcoholic solution, after the evaporation of which the hormone is obtained again in a yield of 95-1000 / 0 with higher purity.
