  Verfahren zum Entfernen von   Eiweisssubstanzen aus solche enthaltenden Flüssigkeiten.   



   Gegenstand vorliegender Erfindung bil det ein Verfahren zur Entfernung von Ei    weisssubstanzen    aus solche enthaltenden
Flüssigkeiten, zum Beispiel solchen, die zu therapeutischer Verwendung oder zur Wei terverarbeitung auf therapeutisch verwert bare Präparate bestimmt sind, wie zum Bei spiel Seren usw. Das Verfahren der Erfin dung ist dadurch gekennzeichnet, dass die    Eiweisssubstanzen    enthaltenden   Flüssigkei-    ten mit metallsauren Verbindungen, zum Beispiel Wolframaten, Uranaten, Chromaten und insbesondere Vanadaten zum Beispiel in Form von   Losungen    oder Gemischen solcher, und organischen Säuren, wie zum Beispiel Essigsäure, behandelt werden. Die Zusatzstoffe können dabei nacheinander oder   ire    Gemisch verwendet werden.

   Als Endprodukt des Verfahrens werden mehr oder weniger oder vollständig von ihrem Eiweiss befreite Flüssigkeiten, zum Beispiel Seren, erhalten.



  Diese sind insbesondere für therapeutische Verwendung oder für die Weiterverarbeitung auf therapeutisch verwendbare Präparate be stimmt, für welche Zweeke das   Vorhanden-    sein von Eiweisssubstanzen bezw. grosseren
Mengen derselben von nachteiligen Ersehei nungen begleitet ist und daher vermieden werden muss.



   Das Verfahren gemäss der Erfindung bie tet gegenüber den bekannten, zum Beispiel mittelst starken   Säuren arbeitenden Metho-    den den Vorteil, wie Versuche ergaben,   dapa-    die   Wasserstoffionenkonzentration    der zu behandelnden Flüssigkeiten nur geringfügig verschoben zu werden braucht und dieselben so für ihre zum Beispiel therapeutische Verwendung nicht unbrauchbar gemacht oder in ihrem Wert gemindert werden.

   Gegenüber dem bekannten Verfahren, die   Eiweisssub-    stanzen durch Erhitzen zum Ausfällen zu bringen, besitzt das vorliegende Verfahren, bei welchem zum Entfernen der Eiweisssubstanzen nicht erhitzt zu werden braucht, den Vorteil, dass die wertvollen Bestandteile der Flüssigkeiten, zum Beispiel Seren, nicht zerstört oder mehr   oder weniger unbrauch-    bar gemacht werden.



   Die Vorteile des vorliegenden Verfahrens beruhen auf den sich dabei abspielenden chemischen bezw. kolloidchemischen Vorgängen. Die in den Flüssigkeiten ent  haltenen    Eiweisssubstanzen absorbieren   näm-    lich wie Versuche ergaben, die Zusatzstoffe, oder bei deren überschüssigen Verwendung zumindest einen Teil derselben und ergeben mit ihnen unlösliche und dementsprechend zur Ausscheidung gelangende Verbindungen komplexer Natur. Das Vorliegen derartiger Verbindungen ist dabei daraus ersichtlich, da¯ sie die Eigenschaften der Einzelkomponenten eingebüsst haben und je nach der Art der verwendeten   metall-    sauren Verbindung eine verschiedene Färbung aufweisen.

   So ist zum Beispiel die durch Fällung mit Vanadat erhaltene unlösliche   Eomplexverbindung ausgesprochen    gelb   gefarbt.   



   Die in den behandelten Flüssigkeiten enthaltenen, von den   Eiweisssubstanzen    nicht absorbierten Anteile der metallsauren Verbindungen können durch Dialyse, zum Beispiel mittelst   Eollodiumhäutchen,    leicht aus den Flüssigkeiten ohne Beeinträchtigung derselben wieder entfernt werden, was bei bekannten Fällmitteln meist nicht oder nur bedingt möglich ist.



   Mit Vorteil werden zur Durchfiihrung des vorliegenden Verfahrens Gemische bezw.



  Lösungen von metallsauren Verbindungen und organischen Säuren verwendet, deren   Wasserstoffionenkonzentration    3,   3    bis 5,   4      beträgt-.    Dabei hat es sich insbesondere bei Behandlung von Seren herausgestellt, da¯ es besonders zweckmässig ist, auf einen, durch einen Raumteil zweckmäBig destillierten Wassers verdünnten Raumteil Flüssigkeit 1, 1 Raumteil 0, 1 n-Lösung einer metallsauren Verbindung, vorzugsweise Natriumvanadat, und 8, 9 Raumteile 0, 1 n-organischer Säure, vorzugsweise Essigsäure, zu verwenden.



   Das vorliegende Verfahren gestattet bei der Behandlung von Seren usw. gleichzeitig, die Beschaffenheit der behandelten Flüssigkeit festzustellen und zu kontrollieren. Dies beruht darauf, da¯ eine gewisse Relation zwischen Beschaffenheit der Flüssigkeit, zum Beispiel des Serums einerseits und der für die Entfernung von EiweiBstoffen   nol ;

  -    wendigen Art und Menge der Zusatzstoffe anderseits besteht, insofern, als die   Ausschei-    dung der   EiweiBsubstanzen,    wie   Versuehe    ergaben, in bestimmten Fällen nur bei Zugabe ganz bestimmter Mengen   metallsaurer    Verbindungen und organischer Säuren erfolgt.   Diese Erscheinung läBt sich    sinngemäB dazu verwenden, festzustellen, ob eine normalerweise bei Zugabe bestimmter, zum Beispiel der oben zuletzt genannten Zusatzmengen,   EiweiBsubstanzen    ausseheidende Flüssigkeit, zum Beispiel Serum, diese   Aus-      scheidung    zeigt oder nicht, das   beisst    ob zum Beispiel das Serum als normal anzusprechen ist oder Abweichungen aufweist.

   Diese Abweichungen können ihrerseits wieder   charak-    teristisch sein und zum Beispiel darin be  stehen, daB    bei Zugabe anderer, auch wieder ganz bestimmter Mengen metallsaurer Verbindungen und organischer Säuren die Ei  weiBsubstanzen    zur Ausscheidung gelangen.



  Diese Abweichungen können so, nachdem ihr Vorhandensein erst generell festgestellt ist. auch spezifisch näher erfasst werden.



   Nach Zugabe der metallsauren Verbindungen und organischen Säuren empfiehlt es sich, die   behandelten Flüssigkeiten lan-      germe    Zeit, zum Beispiel etwa 24 Stunden, zweckmässig unter Aussehaltung von Erschütterungen oder sonstiger Bewegungen des Behandlungsgutes, der Einwirkung der Zusatzstoffe zu überlassen, um eine   mög-       lichst weitgehende Entfernung der Eiweiss- @    substanzen und eine entsprechende Klärung der von demselben befreiten Flüssigkeiten zu erzielen. Hierauf kann durch Dekantieren, Filtrieren, Zentrifugieren oder dergleichen leicht eine Scheidung der   fliissigen    von den festen Anteilen vorgenommen werden.



   Ausf hrungsbeispiele:
1. 5   cm.3 eines Menschenserums, entha@-    tend 0, 42 gr EiweiB, wurden zunächst mit 5 cm3 destillierten Wassers und dann mit 50   em3 einer 2,    5   cm3 n/10    Trinatriumvanadat und 7, 5 cm3 n/10 EssigsÏure enthaltenden Mischung versetzt. Diese Mischung besass eine Wasserstoffionenkonzentration von   pn    = 5, 14. Das Ganze wurde dann 24 Stunden   lang unter möglichster Aussehaltung    von   Ersehütterungen    stehen gelassen, wobei ein reichlicher Niederschlag von Eiweissvanadinsäure ausfiel, der dann durch Filtration entfernt wurde.

   Seine Untersuchung   ergab, daB    er etwa 95% der ursprünglich vorhandenen Eiweisssubstanz enthielt, das Serum durch die Behandlung also fast vollständig von seinem   EiweiBgehalt    befreit worden war.



   2. 5 cm3 eines Pferdeserums, enthaltend 0, 49   gr Eiweisssubstanz,    wurden mit 5 cm3 destillierten Wassers vermischt und dann mit 50   em 3 einer    1, 35   cm"n/10      Trinatriumvana-    dat und 8, 65 cm3 n/10 EssigsÏure enthaltenden Mischung versetzt und durchgeschüttelt.



  Die   Wasserstoffionenkonzentration    der VanadatessigsÏurel¯sung betrug pH =4,14. Das erhaltene Gemisch wurde dann 24 Stunden   lang unter möglichster Ausschaltung    von Er  schütterungen stehen gelassen,    wobei sich ein 'L'eil der Eiweisssubstanz   abschied.    Hierauf wurde der   Niedersehlag    abzentrifugiert und eine Dialyse mittelst KollodiumhÏutchen durchgeführt. Im Filtrat wurden noch etwa 90 % der ursprünglichen   Eiweisssubstanz    festgestellt.



   Vorstehende Beispiele illustrieren gleichzeitig, dass je nach den gewählten   Fällmittel-    zusatzmengen und deren MengenverhÏltnissen untereinander der Umfang der Eiweissausfällung ein   versehiedener    ist. In dieser Entscheidung besitzt man damit auch eine M¯glichkeit, anhand des Auftretens der Eiweissfällung und ihres Umfanges die Be  schaffenheit    eines Serums festzustellen und damit auch das Vorliegen etwaiger Krank  heitsfälle    erkennen zu können.



   Stellt man sich nÏmlich 20   Fällmittel-    gemische mit immer mehr steigendem Vanadat-und immer mehr fallendem   Essigsäure-    gehalt aus 0, 6 bis   3,    45 cm3 0, 1   n-Losung      Trinatriumvanadat    und 9, 40 bis 6, 55 cm3 0, 1   n-Losung    Essigsäure von entsprechend abgestuftem   PH-Gehalt    her und versetzt je 5   em3 eines    mit der gleichen Menge destil  lierten    Wassers verdünnten Serums mit diesen Fällmittelgemischen 1 bis 20, so ergibt sich bei einem normalen Serum folgendes Bild : Die relativ wenig   Vanadat    und viel Essigsäure   enthaltenden-Zusatze 1 bis    5 bewirken überhaupt keine Fällung.

   Bei dem nächstfolgenden Zusatz 6 (vergl. das mit dem entsprechenden FÏllmittelgemisch arbeitende obige Beispiel   2)    tritt eine   gering-    fügige Eiweissfällung auf. Diese steigert sich dann bei den einen immer grösseren Vanadatgehalt aufweisenden Zusätzen 7 bis 12, um bei dem Zusatz   13    (vergl. das   mitdem    entsprechenden Fallmittelgemisch arbeitende obige Beispiel 1) ihr Maximum zu erreichen, um von dem nächstfolgenden noch vanadatreicheren Zusatz 14 bis zum Zusatz 20 allmählich wieder etwas zu fallen.



   Bei   Erebsserum    dagegen findet eine Verschiebung statt, in der Weise, dass schon bei Zusatz 5 oder 4 oder vielleicht schon bei Zusatz 3 und in ganz schweren   Fällen aucli    schon bei dem vanadatärmsten Zusatz   1    eine   Eiweissausfällung    eintritt.



   Mit derartigen Versuchsreihen, welche naturgemäss in erster Linie dazu anzustellen sind, die geeignetsten VerhÏltnisse f r die   Enteiweissung    der Seren je nach deren spezifischer Beschaffenheit zu ermitteln, ist damit also gleichzeitig auch eine Möglichkeit gegeben, in Abhängigkeit von der Art der Enteiweissung die Beschaffenheit des jeweils behandelten Serums festzustellen und diag  nostisch    auszuwerten, wie sie durch andere   Fällmittelmethoden,    zum Beispiel die be  kannte    Verwendung von Phosphorwolframsäure als   Eiweissfällungsmittel    in keiner Weise gegeben ist.
