Description
Verfahren zur Herstellung einer löslichen Halogenpyridonverbindung. Durch das sch-weiz. Patent. Nr.<B>161687</B> sind die Salze der Halogen-, im besonderen Jodpyridone, die durch zur Salzbildung be fähigte saure Gruppen substituiert sind, mit aliphatischen Aminen als besonders geeignet für die pharmazeutische Verwendung, insbe sondere für die röntgenographische Darstel lung von Organen, bekannt geworden.
Zur Salzbildung sind auch solche Amine, die eine oder zwei Ogygruppen im Molekül enthalten, wie zum Beispiel das Diäthanolamin, be nutzt worden. Solche Salze zeigen eine ver hältnismässig hohe Wasserlöslichkeit und er möglichen die Herstellung verhältnismässig konzentrierter Lösungen und damit geringe Injektionsvolumina bei der intravenösen Ver abreichung.
Bei der weiteren Bearbeitung wurde nun gefunden, dass unter den Aminsalzen der ge nannten Halogenpyridone mit einem sauren Substituenten die 11Iethylglucaminsalze sich durch besonders hohe Wasserlöslichkeit und physiologische Indifferenz auszeichnen.
So kann man zum Beispiel mit Hilfe des 1VIe- thylglucaminsalzes 65 bis 70 g der 3,5-Dijod- 4-pyridon-N-essigsäure zu<B>100</B> cm' lösen, während mittels des Diäthylaminsalzes nur 50,6 g der Säure und mittels des Diäthanol- aminsalzes nur 35g der Säure in <B>100</B> cm' Lösung gelöst werden können.
Als weiterer Vorteil der neuen Salze ist hervorzuheben, dass ihre wässrigen Lösungen neutral rea gieren, eine Lösung, die in 100 cm' 60 g des 1Vlethyl:glu oamins"alzes der 3,5-Dijad-4-gyri- don-N-essigsäure enthält, hat eine Wasser- stoffionen-Konzentration von 7,2 bis 7,5, während eine Lösung des Diäthanolamin- salzes die Wasserstoffionen-Konzentration von 5,5 zeigt.
Die neuen Salze, zum Beispiel dao 14lethyl'glucaminsalz der 3,5I)ijod,4- pyridon-N-essiggäure, haben sich als physio logisch noch indifferenter erwiesen als das bisher hauptsächlich benutzte Diäthanolamin- salz.
Die Herstellung der neuen Salze erfolgt in der für die Herstellung von Aminsalzen üblichen Weise zum Beispiel durch unmittel bares Neutralisieren der freien Säure durch die freie Base oder durch doppelte Umsetzung von deren Salzen. Die Möglichkeit, das Me- thylglucamin in Form einer reinen kristalli sierten Verbindung für die Umsetzung zu verwenden, bietet den Vorteil, dass auch die neuen Salze leicht in ausgezeichneter Rein heit erhalten \werden können.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung einer leicht löslichen Halogenpyridonverbindung. welches dadurch gekennzeichnet ist, dass man 3,5 Dijod-4-pyridon-N-essigsäure mit N-Methyl- i d-glucamin, wie es erhältlich ist durch kata lytische Reduktion von Glucose in metha- nolischer Methylaminlösung, umsetzt.
Das so erhältliche Produkt bildet feine Kristalle vom Schmelzpunkt. 162 bis 163 .
Es soll medizinische Anwendung finden. Die Umsetzung der 3,5-Dijod-4-pyridon-N- essigsäure mit dem Methy lglucamin wird zweckmässig in wässrigem Medium durchge führt, wobei Lösungen erhalten werden, die unmittelbar als solche verwendet werden können.
<I>Bespiele:</I> 67,5 g 3,5-Dijod-4-pyridon-N-essigsäure werden mit 32,5g N-14lethyl-d-glucamin, wie es durch katalytische Reduktion von Glucose in methanolischer @ethy laminlösung erhält lich ist, vermischt, die Mischung wird mit v enig Wasser angerührt, dann mit Wasser auf zunächst etwa 90 cm' aufgefüllt und durch Schütteln in Lösung gebracht;
die so erhaltene Lösung wird dann mit Wasser auf 100 ein aufgefüllt. 100 cm' dieser Lösung enthalten somit 100g Methylglucaminsalz- 6I,5 g 3.5-Dijod-4-pyridon-N-essigsäure.
Zur Darstellung des Salzes kann man auch so verfahren, dass man zu einer kochen den Lösung von 250 g 3,5-Dijod-4-pyridon- N-essigsäure in 1250 cm' Methanol eine Lö sung von 132 g N-Methyl-d-glucamin, das wie oben beschrieben erhalten wurde. in 1250 cm' Methanol zusetzt, wobei aus der heissen Lö sung das so gebildete Salz sich in feinen Kri stallen vom Schmelzpunkt 162 bis 163 ab scheidet. Dieses Salz kann direkt in der er forderlichen Menge Wasser gelöst werden.

No. <B> 161687 </B> are the salts of the halogen, in particular iodopyridones, which are substituted by acidic groups capable of salt formation, with aliphatic amines as being particularly suitable for pharmaceutical use, in particular for X-ray imaging of organs.
Amines which contain one or two Ogy groups in the molecule, such as diethanolamine, have also been used for salt formation. Such salts have a relatively high solubility in water and allow the production of relatively concentrated solutions and thus low injection volumes for intravenous administration.
In further processing it has now been found that among the amine salts of the halopyridones mentioned with an acidic substituent, the methylglucamine salts are distinguished by particularly high water solubility and physiological indifference.
For example, 65 to 70 g of 3,5-diiodo-4-pyridone-N-acetic acid can be dissolved to <B> 100 </B> cm 'with the help of the ethylglucamine salt, while with the diethylamine salt only 50.6 g of the acid and, by means of the diethanolamine salt, only 35 g of the acid can be dissolved in <B> 100 </B> cm 'solution.
Another advantage of the new salts should be emphasized that their aqueous solutions react neutrally, a solution that contains 60 g of the 1Vlethyl: glu oamine "salt of 3,5-Dijad-4-gyridone-N-acetic acid in 100 cm contains, has a hydrogen ion concentration of 7.2 to 7.5, while a solution of the diethanolamine salt shows the hydrogen ion concentration of 5.5.
The new salts, for example the 14lethyl'glucamine salt of 3,5I) iodine, 4-pyridone-N-acetic acid, have proven to be physiologically even more indifferent than the diethanolamine salt mainly used up to now.
The new salts are prepared in the manner customary for the preparation of amine salts, for example by neutralizing the free acid directly with the free base or by double conversion of the salts thereof. The possibility of using the methylglucamine in the form of a pure crystallized compound for the reaction offers the advantage that the new salts can easily be obtained in excellent purity.
The present patent relates to a process for the preparation of a readily soluble halopyridone compound. which is characterized in that 3.5 diiodo-4-pyridone-N-acetic acid is reacted with N-methyl-i d-glucamine, as can be obtained by catalytic reduction of glucose in methanolic methylamine solution.
The product thus obtainable forms fine crystals with a melting point. 162 to 163.
It should find medical application. The reaction of 3,5-diiodo-4-pyridone-N-acetic acid with methyl glucamine is expediently carried out in an aqueous medium, solutions being obtained which can be used directly as such.
<I> Examples: </I> 67.5 g of 3,5-diiodo-4-pyridone-N-acetic acid are mixed with 32.5 g of N-14lethyl-d-glucamine, as is achieved by catalytic reduction of glucose in methanolic @ethy lamin solution is available, mixed, the mixture is stirred with a little water, then filled with water to initially about 90 cm 'and brought into solution by shaking;

This salt can be dissolved directly in the required amount of water.
