Process for the production of an anhydro-corticosterone. The present invention relates to a process for the preparation of the d "-anlydro-corticosterone acetate of the formula
EMI0001.0004
which is characterized in that a d4; 11-3-keto-etio-choladienoic acid halide is reacted with diazomethane and the product obtained is converted into the acetoxy ketone using an acetylating agent.
As has been found, the already known process product shows an extremely good pharmacological effect; it should be used as a remedy or as an intermediate product for the manufacture of remedies. For example, 0.8 mg of d "-anhydrocorticosterone acetate is capable of all controlled functions (residual nitrogen, hemoglobin, Na, K, Cl and P04 blood titers and excretion, amount of urine, body temperature) with daily subcutaneous injection in adrenalectomized dogs and weight, milk, water and feed intake) to normalize completely.
For example, the carboxylic acid halide, like the chloride, can be treated with an excess of diazomethane, the diazoketone being obtained with elimination of hydrogen halide, and acetic acid can then be allowed to act. However, the diazomethane can also be added gradually to the acid halide, the diazoketone immediately reacting with the hydrohalic acid released during the condensation to form the corresponding halogen ketone.
The halogen ketone thus obtained can be ziu. Example through treatment with. an acetic acid salt, for example with potassium acetate, into the anhydrocorticosterone acetate. Example: 10 parts by weight of the d4; 11_3_Keto-ätio-choladienoic acid chloride [producible e.g. B. from 4 4: 11-3-keto-etio-choladienoic acid according to the method of A. L. Wilds and C.
H. Shunk, J. Am. Chem. Soc. 70, 2427 (1948)], 600 parts by volume of an ethereal 1.5% diazomethane solution are added to 200 parts by volume of dry ether and 100 parts by volume of dry benzene at -15.
The solution is left to stand at -15 for 30 minutes, then at 0 for 1 hour and evaporated in vacuo.
100 parts by volume of glacial acetic acid are added to the residue obtained in this way, containing the diazoketone, and the solution is heated to 90 ° until the evolution of nitrogen ceases completely. The solution is then evaporated in vacuo and the residue is recrystallized from isopropyl ether and a little ether.
The 44: 11-21-acetoxy-pregnadiene-3,20-dione (all-anhydro-corticosterone acetate) obtained in this way melts at 179 to 181. Its mother liquors are chromatographed using aluminum oxide -ind the benzene-pentane (1:
1) - as well as the benzene Eltlate evaporated together. Another small amount of d 4: 11-21-acetoxy-pregnadiene-3,20-dione is obtained by sublimation in a high vacuum at 170 to 190 and recrystallization from isopropyl ether and a little ether.
The crude diazoketone can also be converted into 21-chloro-ketone, for example with an ethereal hydrogen chloride solution, and this into the above-described d4; 11_21-acetoxy-pregnadiene-3,20-dione with the aid of dry potassium acetate in boiling acetone.
The 44: 11-3_Keto-etio-choladienoic acid used to prepare the starting material can be obtained, for example, from its methyl ester by alkaline saponification.
