Description
Verfahren zur Herstellung von 3-N-[(5-Nitro-2-furfuryliden)-amino]-oxazolidonen-(2) 3-N- [ (5-Nitro-2-furfuryliden) -a.minol- oxazolidone-(2) der Formel

in welcher R Wasserstoff, Methyl oder Äthyl, vorzugsweise Wasserstoff, und R1, R2, R3 und R4 Wasserstoff oder einen organischen Rest, wie z. B. Alkyl, Oxyalkyl oder Halogenalkyl bedeuten und gleich oder verschieden sein können, sind bisher noch nicht beschrieben worden.
Diese neuen Verbindungen besitzen eine hohe in vivo-Wirksamkeit gegen Mikro beninfektionen, z. B. mit Salmonella typhosa und Vibxio comma. Gegenüber den bisher bekannten antibakteriell wirksamen Nitro- ('ttran-Derivat.ett zeichnen sie sich durch ihre ausserordentlich geite Verträglichkeit und das Fehlen toxischer Nebenerscheinungen, insbe sondere bei oraler Applikation, aus.
Das erfindungsgemässe Verfahren zur Ilerstellung der neuen Nitrofuranderivate ist dadurch ;ekennzeichnet, dass man entspre- eliende 5-Nitro-2-fnrale mit. gegebenenfalls funktionell abgewandelter Oxogr@ippe mit 3-Amino-oxazolidonen-(2) umsetzt.
Die zur Du,rehführung des erfindungs- geiltässen Verfahrens benötigten 3 - Amino- oxazolidone-(2) können durch Umsetzung von f-Oxy alkyl-hydrazinen mit Alkylcarbona ten, z. B. Diäthylcarbonat, in Gegenwart von Al kalimetallkatalysatoren, z. B. Natriummethy- la.t, hergestellt werden.
Die bei dieser Um setzung erhaltenen Lösungen können ohne Isolierung der 3-Amino-oxazolidöne-(2) di rekt mit den 5-Nitro-2-furalen oder deren funktionellen Derivaten, wie z. B. den Diace- taten, umgesetzt werden.
<I>Beispiel 1</I> 3-N- [ (5-Nitro-2-fu.rfuryliden) -aminol- oxazolidon-(2)

Eine Lösung von 1,8g Natrium in 25 em3 wasserfreiem Methanol wird zu einer Vii- schung aus 46 g f)-Oxäthyl-hydrazin und 90 cm3 Diäthylearbonat gegeben. Die erhal tene Lösung wird 3 Stunden am Rückfluss erhitzt und dann auf Raumtemperatur abge kühlt.
Jetzt können zwei Wege zur Herstel lung des gewünschten Produktes ringesehla- gen werden a) Die obige Reaktionsmischung wird mit 375 cm3 Alkohol und 25 cm3 konzentrierter Salzsäure versetzt. Diese Lösung wird dann unter Rühren in eine Lösung von 75 g 5-Ni- tro-2-fural in 750 cm3 Alkohol eingetragen. Die erhaltene Mischung wird 1 Stunde ge rührt. Die abgeschiedenen gelben Kristalle werden abgesaugt und nacheinander mit Wasser und Alkohol gewaschen.
Nach dem Trocknen werden 107 g 3-N-[ (5-Nitro-2-fur- furyliden)-amino]-oxazolidon-(2) erhalten, die nach dem Umkristallisieren aus N,N-Di- methylformamid bei 256-257 C schmelzen. Ausbeute 791/o der Theorie.
b) Die obige Reaktionsmischung wird nach Zusatz von 40 eins Alkohol, 240 eins Wasser, 38 g Schwefelsäure und 127 g 5-Ni- tro-2-fural-diaeetat 1 Stunde unter Rückfluss erhitzt, auf Raumtemperatur abgekühlt und wie in a) weiterverarbeitet. Nach dem Troek- nen werden 103 g 3-N- [ (5 Nitro-2-furfuryl- iden)-amino]-oxazolidon-(2) erhalten, die nach dem Umkristallisieren aus N,N-Di- methylformamid bei 256-257 C schmelzen.
Ausbeute 76 % der Theorie.
In gleicher Weise kann für diese Um setzung .eine durch Reduktion von .3-Nitro- oxa.zolidon-(2) erhaltene Lösung von 3-Amino- oxazolidon-(2) verwendet. werden.
<I>Beispiel 2</I> 3-N- [ (5-Nitro-2-fLirfuryliden) -amino]- 5-m.ethyl-oxazolidon- (2).
Unter Verwendung von 45,0 g 2-Oxypro- pyl-hydrazin (das durch Kondensation von Propylenchlorhydrin mit Hydrazinhydrat er halten wurde), 74 eins Diäthylca,rbonat und einer Lösung von 1,4g Natrium in 16 eins wasserfreiem Methanol werden gemäss Bei spiel 1a 85 g 3-N-[ (5-Nitro-2-furfuryliden)- a.mino] -5 methyl-oxazolidon- (2) vom F.
255 bis 956 C erhalten, was einer Ausbeute von 70,4% der Theorie entspricht.
<I>Beispiel 3</I> 3-N- [(5-Nitro-2-f-Lirfuryliden) -amino]- 4-methyl-oxazolidon- (2).

Zu einer Lösung von 7,8 g Methyl-5-nitro- 2-furylketon in 75 cm3 30 vol.%iger sieden- der Essigsäure werden 5 g 3-Amino-oxazo- lidon-(2) gegeben. Die Lösung wird 7 Minu ten am Rückfluss erhitzt und dann abgekühlt. Die erhaltenen Kristalle werden abgesaugt und mit Wasser und dann mit Alkohol ge waschen und getrocknet.
Es werden 8,9 g <B>3-N-</B> [ (5 - Nitro-furyl-2'-ätliyliden)-amino]- oxazolidon-(2-) vom F. 125-12811C erhalten. Ausbeute 76% der Theorie. Nach dein Um- kristallisieren aus absolutem Alkohol steigt der Schmelzpunkt auf 132,5-133 C.
<I>Beispiel 11</I> <B>3-N-</B> [ (5 -Nitro- 2-furfuryliden) - amino] - 4,5-diniethyl-oxazolidon-(2). Unter Benutzung von 50,4 g 3-Hydrazino- bu.tanol-(2) (das durch Kondensation von 2,3-Epoxybutan mit.
Hydrazinhydrat herge stellt wurde), 63 g Diäthylearbonat und einer Lösung von 3,5 g Natrium in 35 em3 wasser freiem Methanol werden gemäss der in Bei spiel 1a beschriebenen Methode 80 g 3-N-[(5 Nitro - 2 -i furfury liden) -amino]-4,5-dimethyl- oxazolidon-(2) vom F. 143-144 C erhalten.

UNTERANSPRÜCHE 1. Verfahren nach Pa.tentansprueh, da durch gekennzeichnet, da-ss man als Ausgangs material ein 3-Amino-oxazolidon-(2) verwen- det, das in mindestens einer der Stellungen 4 und 5 mindestens .einen gegebenenfalls substi tuierten Alkylr est enthält. 2.Verfahren nach Patentanspruch und Unteransprueh 1, dadurch gekennzeichnet, dass der genannte Alkylrest durch eine Hv- droxygruppe substituiert ist. 3. Verfahren nach Patentanspruch und Unteranspruch 1, dadurch gekennzeichnet, dass der genannte Alkylrest durch ein Ha logen substituiert ist..
