Verfahren zur Herstellung von Testosteronderivaten <I>Beispiel. Z</I> I1 a-Oxy-testosteron Es wird eine Nährlösung hergestellt, die 20g Lactalbuminauszug, 3 g Maisquellwasser und 50 g teehnischer Dextrose pro Liter ge löst enthält und deren pH-Wert auf 4,3-4,5 eingestellt wurde.
Drei Liter dieses sterilisier ten Mediums wurden mit Rhizopus nigrieans minus (ATCC Nr. 6227b) geimpft und 24 Stunden bei 28 C incubiert, wobei die Rühr- gesehwindigkeit und Belüftung so eingestellt wurde, dass die Sauerstoffaufnahme, gemessen nach der Methode von Cooper, Fernstrom, Miller [Ind. Eng. Chem. 36, 504 (1944)] 6,3 bis 7 Millimolen Na.S03 pro Liter -und Stunde entsprach.
Zu dieser, eine 24stündige Kultur von Rhizopus nigricans minus enthaltenden Nährlösung wurde 1,5 g Testosteron, das zwecks Bildung einer Steroidsuspension in der Nährlösung in 30 cm3 Äthanol gelöst worden war, gegeben. Nach weiterer 24stündiger In- cubation unter denselben Temperatur- und BelüftLtngsbedingungen wurden Würze und Mycel extrahiert.
Das Mycel wurde filtriert, zweimal mit einem, ungefähr dem Mycelvolu- men gleichen Volumteil Aceton gewaschen und zweimal mit einem, ungefähr dem Mycelvolu- men gleichen Vol,umteil Methylenchlorid ex trahiert. Die Aceton- und Methylenchlorid- auszüge wurden einschliesslich Lösungsmittel dem Filtrat der Würze zugegeben.
Die ver mischten Extrakte und das Filtrat der Würze wurden nacheinander zweimal mit 0,5 Vol: Teilen Methylenchlorid und dann zweimal mit 0,25 Vol.-Teilen Methylenchlorid extra- biert. Die vereinigten Methylenchloridauszüge wurden zweimal mit 0,1 Vol.-Teil 2 /oiger Uatriumbicarbonatlösung und dann zweimal mit 0,1 Vol.-Teil Wasser gewaschen.
Nach dem Trocknen des Methylenchlorids mit 3 bis 5 g wasserfreiem Natriumsulfat pro Liter Lösung und Filtrieren wurde das Lösungsmittel durch Destillation entfernt. Der Rückstand wurde in möglichst wenig Methylenchlorid aufgenommen, filtriert und das Lösungsmittel an der Luft oder am Wasserbad verdampft. Die Erfindung bezieht sich auf ein Ver fahren zur Herstellung von 1.1a.-Oxy-Testoste- ron und 11a-Oxy-10-normethyl-testosteron.
C*emäss der Erfindung werden diese Ver- bindun--en aus Testosteron bzw. 10-Normethyl- testosteron hergestellt, und zwar durch bio chemische Oxydation mittels Enzymen, die von einer Pilzart der Cxattung Rhizopus, die der Ordnung lblucorales angehört, erzeugt werden.
Vorzugsweise wird ein lebensfähiger Pilz und das Steroid unter Fermentationsbe- dingungen in Gegenwart eines für den Pilz geeigneten Nährmediums zusammengebracht, wie in den folgenden Beispielen noch ein gehend beschrieben wird. Der Rückstand wog nach dem Verdampfen des Methylenchlorids 2,5492 g.
Eine teilweise Entfärbung des rohen Rüek- standeskonnte durch Aufnehmen desselben in 35 cm3 llethy lenchlorid, Zugabe von 3 g Magnesol (synthetisches Magnesiumsilikat) und Filtrieren erreicht werden.
Nach dem Verdampfen des Lösungsmittels wurde der Rückstand in einem L berschuss Aceton auf genommen, mit 0,2 g Celit Nr.545 (Diato- meenerde) vermischt, filtriert und zur Trok- kene verdampft. Der Rückstand wurde in 150 cm3 Benzol gelöst und über 75 g Tonerde (mit Salzsäure gewaschen und bei l20 C ge trocknet), mit 150 cm3-Mengen Lösungsmittel, wie in der Tabelle angegeben, fraktioniert. Zur Papierchromatographie wurde das Pro pylenglykol-Toluolsystein benutzt..
Die vom Lösungsmittel befreiten Frak tionen 23 bis 30 ergaben 378 mg festes Ehiat, das in Aceton gelöst, wurde. Diesa,Lösung7:wurde auf 10 cm3 eingeengt, eine kleine Menge eines flockigen Niederschlages abfiltriert und zur Trockene verdampft. Der Rüekstand wurde in 15 em3 :.1thy lacetat gelöst, am Wasserbad auf 5 cm3 eingeengt und bis zur Kristallisa tion gekühlt.
Diese Kristalle wurden durch Filtration abgetrennt und dreimal mit je 1 ein3 Äthy laeetat gewaschen.
Diese Kristalle wogen<B>319,9</B> mg und wur den in Methylenchlorid aufgenommen, die Lösung mit Magnesol entfärbt, die Sus pension filtriert: und das Filtrat eingedampft. Der Rüekstand wurde nochmals in 2 cm3 Äthylaeetat gelöst und daraus auskristalli siert, wobei 7.98,7 mg Kristalle mit einem Schmelzpunkt von 179-l82 C erhalten wur den.
Diese Kristalle wurden in 7 em3 Äthyl- acetat gelöst, die Lösung auf 4 em3 eingeengt, wobei nach der Kristallisation 169,7 mg 11.a- Oxy-testosteron mit einem Erweichungspunkt von 17811 C und einem Schmelzpunkt von 181. bis 181,5 C erbalten zenirdc,.n. Die. optische Drehung betrug

Das erhaltene oxydierte Steroid wird aus der Fermentationsmischung auf die in Beispiel 1. gezeigte Weise isoliert und das lla-Oxy-10-normethyl-testosterori dureh Umkristallisieren aus Äthylacetat ge reinigt.
Die Verwendung von Rhizopus arrhizus in den beiden angeführten Beispielen liefert die selben Ergebnisse.
Das l.1a-Oxy-testosteron und das 11a-Oxy- 10-normethyl-testosteron sind gegen Wärme und verdünnte Säuren beständig. Diese Ver bindungen besitzen selbst pharmakologische Wirksamkeit und sind als Zwischenprodukte für die Synthese anderer, pharmakologisch nützlicher Steroide geeignet.

The mixed extracts and the filtrate of the wort were successively extracted twice with 0.5 part by volume of methylene chloride and then twice with 0.25 part by volume of methylene chloride. The combined methylene chloride extracts were washed twice with 0.1 part by volume of 2% sodium bicarbonate solution and then twice with 0.1 part by volume of water.
After drying the methylene chloride with 3 to 5 g of anhydrous sodium sulfate per liter of solution and filtering, the solvent was removed by distillation. The residue was taken up in as little methylene chloride as possible, filtered and the solvent was evaporated in the air or on a water bath. The invention relates to a process for the production of 1.1a.-oxy-testosterone and 11a-oxy-10-normethyl-testosterone.
According to the invention, these compounds are produced from testosterone or 10-normethyl testosterone, specifically by bio-chemical oxidation by means of enzymes which are produced by a species of fungus of the Rhizopus species, which belongs to the order lblucorales.
A viable fungus and the steroid are preferably brought together under fermentation conditions in the presence of a nutrient medium suitable for the fungus, as will be described in detail in the following examples. The residue weighed 2.5492 g after evaporation of the methylene chloride.
Partial decolorization of the raw residue could be achieved by taking it up in 35 cm3 of ethylene chloride, adding 3 g of magnesol (synthetic magnesium silicate) and filtering.
