Description

  

  Verfahren zur Herstellung von Verbindungen der Steroidreihe    Gegenstand der vorliegenden Erfindung ist ein  Verfahren zur Herstellung von in     12-Stellung        unsub-          stituierten    gesättigten oder ungesättigten     11-Keto-          spirostanen,    das dadurch gekennzeichnet ist, dass  man entsprechende     11,12ss-Oxyde    mit     Halogenwas-          serstoffsäuren    spaltet, in den erhaltenen     12a,

  llss-          Halogenhydrinen    die 11 ständige     Hydroxylgruppe    zur       Ketogruppe    oxydiert und das Halogenatom in     12-          Stellung    reduktiv entfernt.  



  Die Ausgangsstoffe können im Kern oder in der  Seitenkette substituiert sein, z. B. in Stellung 3,5,6 ,  17,20     undloder    21 durch freie oder funktionell ab  gewandelte     Oxy-    oder     Oxogruppen,    wie     Acyloxy-,     z. B.     Acetoxy-,        Propionyloxy-    oder     Benzoyloxygrup-          pen,    durch     Alkoxy-,    z.

   B.     Methoxy-    oder     Äthoxy-          gruppen,    durch Halogenatome, durch     acetalisierte          Oxogruppen,    ferner durch freie oder funktionell ab  gewandelte     Carboxyl-,    wie     Nitril-    oder veresterte       Carboxylgruppen,    oder durch eine     Lacton-,    z. B.       Butanolidgruppe.    Die Ausgangsstoffe können belie  bige Konfigurationen aufweisen und auch Doppel  bindungen, wie z. B. in 4,5-, 5,6- oder     20,22-Stel-          lung,    enthalten.  



  Für die Spaltung der     11,12ss-Epoxyde    eignen sich  Chlorwasserstoff, Bromwasserstoff oder auch     Jodwas-          serstoff.    Zur Spaltung kann man die genannten Ha  logenwasserstoffsäuren in     wässrigen    oder wasserfreien  Verdünnungsmitteln, z. B. in     wässrigem        Dioxan,    ver  wenden. Das Halogenatom in     12-Stellung    kann mit  Hilfe von     nascierendem    oder katalytisch angeregtem  Wasserstoff entfernt werden.

   Vorteilhaft ist die  Verwendung von mit Wasserstoff beladenem     Raney-          Nickel    oder von Zink in Gegenwart einer schwachen  Säure oder eines Alkohols, z. B. Eisessig oder     Cello-          solve.     



  Die Verfahrensprodukte können als     Heilmittel       oder als Zwischenprodukte zu deren Herstellung ver  wendet werden.  



  In den folgenden Beispielen besteht zwischen Ge  wichtsteil und     Volumteil    die gleiche Beziehung wie  zwischen Gramm und     Kubikzentimeter.    Die Tem  peraturen sind in Celsiusgraden angegeben.    <I>Beispiel 1</I>  0,2108 Gewichtsteile reines     3ss-Acetoxy-11,12ss-          oxido-23@-brom-5a,22a-spirostan    löst man in 19,1       Volumteilen        reinem        Dioxan,    vermischt mit 4,77     Vo-          lumteilen    wässriger 2,5-n. Salzsäure und lässt 1 Std.  bei Raumtemperatur stehen.

   Der Lösung werden so  dann im Verlaufe von 10 Min. unter Umschwenken  14,3     Volumteile    Wasser zugesetzt. Die ausgeschie  denen Kristalle werden nach weiteren 15 Min.     abge-          nutscht,    mit     wässrigem        40proz.        Dioxan    und Wasser  gewaschen und hierauf im Vakuum über     Phosphor-          pentoxyd    und     Kaliumhydroxyd    getrocknet.

   Durch       einmaliges        Umkristallisieren    aus Äther unter Verwen  dung von     Methylenchlorid    als Lösungsvermittler las  sen sich 0,1972     Gewichtsteile    (88,0% d.     Th.)    reines       3ss-Acetoxy    - I I     ss    -     oxy-12ai-        chlor-23        @-brom-5a,22a-          spirostan    in feinen farblosen Prismen vom F. = 240  bis 242      (Zers.)    gewinnen. [a] D = -31    2   (c = 1,000 in Chloroform).  



  Eine Lösung von 0,1313 Gewichtsteilen 3ss  Acetoxy -11     ss-        oxy-12a-chlor-23        e-brom-5a,22a-spiro-          stan    in 11,2     Volumteilen        Äthylenchlorid    und 11,2       Volumteilen    Eisessig wird bei 20  mit 1,79     Volum-          teilen    einer Lösung von Chromsäure in     99,5proz.     Essigsäure vermischt, welche pro     Volumteil    0,0024  Gewichtsteile aktiven Sauerstoff enthält.

   Nach     einer     Reaktionsdauer von     45i/2    Stunden zerstört man die  überschüssige Chromsäure durch Zugabe von 0,45       Volumteilen        wässrigem        O,Jmolarem    Methanol,     gibt     nach weiterer 11/2 Std. noch 11,2     Volumteile    Wasser      zu und konzentriert     alsdann    das Reaktionsgemisch im  Vakuum bei 40      Badtemperatur    auf ungefähr 1     Vo-          lumteil.    Das in     Äther-Benzol-Gemisch    (2:

   1) auf  genommene Rohprodukt wird nacheinander mit     0,5-          n.    Schwefelsäure, Wasser,     2proz.        Natriumhydrogen-          carbonat-Lösung    und Wasser gewaschen und die mit  Natriumsulfat getrocknete Lösung im Vakuum ein  gedampft. Der farblose, kristalline Rückstand wird  aus Äther unter Verwendung von     Methylenchlorid    als       Lösungsvermittler    umkristallisiert.

   Die erste Fraktion  wiegt 0,1007 Gewichtsteile     (77,1'/o    d.     Th.)    und       schmilzt    bei 231-236      (Zers.);    sie besteht aus rei  nem     3ss-Acetoxy-11-keto-12a-chlor-23e-brom-5a,22a-          spirostan.    Aus der Mutterlauge können durch Ein  engen und Zugeben von     Petroläther    weitere 0,0061  (4,6% d.     Th.)    vom F. = 221-231      (Zers.)    erhalten  werden.  



  Aus Äther umkristallisiert schmilzt die reine Ver  bindung bei 233-236      (Zers.);    [a] D = -70    2   (c = 0,987 in Chloroform).  



  In eine siedende Lösung von 0,058 Gewichts  teilen     3ss-Acetoxy-11-keto-12a-chlor-23.-brom-5a,          22a-spirostan    in 5,8     Volumteilen    Eisessig werden  unter starkem Rühren im Verlaufe von 4 Std. in  kleinen     Portionen    1,62 Gewichtsteile Zinkstaub ein  getragen. Man hält hierauf noch weitere 2 Std.  unter Rühren im Sieden und     filtriert    nach dem Ab  kühlen und Verdünnen des Reaktionsgemisches mit  Benzol vom Zink ab. Der Filterrückstand wird  gründlich mit heissem     Benzol    ausgewaschen und  das gesamte Filtrat im Vakuum bei 40      Badtempera-          tur    vollständig eingedampft.

   Die zurückbleibende  Substanz löst man in Äther, wäscht die Lösung nach  einander mit 0,5-n. Salzsäure, Wasser,     2proz.        Na-          triumhydrogencarbonat-Lösung    und Wasser, trock  net sie mit Natriumsulfat und dampft sie ein. Aus  wenig Methanol kristallisieren 0,039 Gewichtsteile  (83,3% d.     Th.)    reines     3ss-Acetoxy-11-keto-5a,22a-          spirostän    in flachen     Oktaedern    vom F. =     216-2210.     



  Der als Ausgangsverbindung verwendete     3ss-          Acetoxy-11,12ss-oxido-23e-brom-5.a"22a-spirostan     lässt sich wie folgt herstellen:  Zu einer auf     0     abgekühlten Lösung von 1,261  Gewichtsteilen     lla,

  23e-Dibrom-hecogenin-acetat    in  10     Volumteilen    wasserfreiem     Tetrahydrofuran    wer  den unter Rühren innerhalb von I1/2 Stunden 5     Vo-          lumteile    einer     2-molaren    Lösung von     Lithiumbor-          hydrid    in     Tetrahydrofuran        zugetropft.    Nachdem das  Reaktionsgemisch während weiteren     101/2    Stunden bei  0      gerührt    worden ist,

   wird das überschüssige Hydrid  durch vorsichtige Zugabe von 10     Volumteilen    l     Oproz.     Essigsäure zerstört und das     Tetrahydrofuran    alsdann  durch Abdampfen im Vakuum entfernt. Das aus  gefallene Reaktionsprodukt wird in     Äther-Benzol-          Gemisch    (2:1) aufgenommen, der Extrakt mit  2-n. Schwefelsäure,     2proz.        Natriumhydrogencarbonat-          lösung    und Wasser gewaschen, getrocknet und im  Vakuum eingedampft.

   Aus dem farblosen Rückstand  erhält man durch Kristallisieren aus Methanol 1,020  Gewichtsteile an in     3-Stellung    teilweise verseiftem 3ss-         Acetoxy-11,c,23        t-dibrom        -12p    -     oxy    -     5a,22a-spirostan     als mikrokristallines Pulver vom F. - 190-203 .  



  Eine Lösung von 1,020 Gewichtsteilen des vor  stehend beschriebenen     3.ss-Acetoxy-lla,23;-dibrom-          12ss-oxy-5a,22a-spirostans    in 19,4     Volumteilen    was  serfreiem     Pyridin    wird zusammen mit 1,5 Gewichts  teilen frisch gefälltem und getrocknetem Silberoxyd  während 22 Stunden im Dunkeln geschüttelt. Vom  Silberoxyd wird alsdann unter Nachspülen mit Benzol  und Äther     abgenutscht    und das Filtrat im Vakuum  eingedampft.

   Den Rückstand löst man zur     Nach-          acetylierung    in 7,5     Volumteilen    wasserfreiem     Pyri-          din,    vermischt mit 3,75     Volumteilen    Essigsäure  anhydrid und lässt 16 Stunden bei Raumtemperatur  stehen. Die Mischung wird hierauf in kalte 2-n.  Schwefelsäure eingerührt, das Rohprodukt in     Äther-          Benzol-Gemisch    (2: 1) aufgenommen, die Extrakte  mit 2-n. Schwefelsäure, Wasser,     5proz.        Natriumbicar-          bonatlösung    und Wasser gewaschen, getrocknet und  eingedampft. Der Rückstand kristallisiert aus Äther.

    Man erhält als erste Fraktion 0,570 Gewichtsteile       3ss-Acetoxy-11        ss,12ss-        oxido-23-brom-5        a,22a-spiro-          stan    vom F. = 224-228      (Zers.).    Aus der Mutter  lauge lässt sich durch Einengen und Zugeben von       Petroläther    eine praktisch ebenso reine zweite Frak  tion von 0,070 Gewichtsteilen vom F. = 223-228'       (Zers.)    gewinnen.

   Das durch erneutes     Umkristalli-          sieren    aus Äther erhaltene völlig reine     Epoxyd          schmilzt        bei        231,5-233,5         (Zers.);        [a]        D        =        -32      (c = 0,925 in Chloroform).

    <I>Beispiel 2</I>  0,0552 Gewichtsteile     3ss-Acetoxy-lIss,12ss-oxido-          235-brom-5a,22a-spirostan    werden in 5,0     Volum-          teilen    reinem     Dioxan    gelöst und mit 1,25     Volum-          teilen    wässriger 2,5-n.     Bromwasserstoffsäure    ver  mischt. Nach etwa 5 Min. beginnen sich aus der  strohgelb gewordenen Lösung Kristalle abzuscheiden.

    Nach 2stündigem     Aufbewahren    im Dunkeln unter       öfterem    Umschwenken setzt man der Kristallsuspen  sion innerhalb von 20 Min. tropfenweise 3,75     Volum-          teile    Wasser zu,     nutscht    nach einer weiteren halben  Stunde ab und wäscht die Kristalle mit     wässrigem        40-          proz.        Dioxan    und Wasser aus.

   Die im Vakuum über       Phosphorpentoxyd    und     Kaliumhydroxyd    getrocknete  Substanz löst man in     Methylenchlorid,    filtriert sie  klar und     kristallisiert    durch Einengen unter Zugabe  von Äther.

   Es werden neben 0,0051 Gewichts  teilen grösstenteils kristallinem     Mutterlaugenrückstand     0,0525 Gewichtsteile     (82,9 /'o    d.     Th.)    reines     3ss-Acet-          oxy-1        lss-oxy-12a,235        -dibrom-5a,22a-spirostan    vom  F. = 237-239      (Zers.)    erhalten. [a123 = -23         2     (c = 1,000 in Chloroform).  



  Die Weiterverarbeitung kann nach Beispiel 1 er  folgen.  



  <I>Beispiel 3</I>  Eine Lösung von 0,0473 Gewichtsteilen     3ss-Acet-          oxy-llss,12ss-oxido-5a,22a-spirostan    in 3,5     Volum-          teilen        peroxydfreiem        Dioxan    vermischt man mit 0,50       Volumteilen    wässriger 4-n.     Bromwasserstoffsäure    und      lässt 1 Std. bei Raumtemperatur stehen. Innerhalb  von 30 Min. werden alsdann 4,75     Volumteile    Was  ser zugegeben, wobei das Reaktionsprodukt kristallin  ausfällt.

   Nach weiteren 30 Min.     nutscht    man ab,  wäscht das farblose Rohprodukt mit 7,5     Volumteilen          wässrigem        40proz.        Dioxan    und<B>12,5</B>     Volumteilen    Was  ser aus und trocknet es im Vakuum über     Phosphor-          pentoxyd    und     Kaliumhydroxyd.    Durch     Umkristalli-          sieren    aus Äther und     Äther-Petroläther-Gemisch    wer  den 0,0399 Gewichtsteile vom F. = 224-226   (72,0% d.

       Th.)        3ss-Acetoxy-1        lss-oxy-12a-brom-5a,          22a-spirostan    gewonnen. Die nochmals aus Äther um  kristallisierte reine Substanz schmilzt bei 224,5 bis  <I>226 ; [a] D = -27 </I>   4  (c = 1,080     in    Chloro  form).  



  Die Weiterverarbeitung kann nach Beispiel 1 er  folgen.  



  Die in diesem Beispiel als Ausgangsmaterial  verwendete Verbindung lässt sich wie folgt gewinnen:  Zu einer siedenden Lösung von 0,276 Gewichts  teilen des     3ss-Acetoxy-11f,12ss-oxido-23e-brom-5a,          22a-spirostans    in 27,6     Volumteilen        Cellosolve    gibt  man unter dauerndem Rühren im Verlaufe von 4 Std.  in fünf Portionen 8,20 Gewichtsteile Zinkstaub und  hält anschliessend noch weitere 4 Std. unter Rühren  im Sieden. Das abgekühlte Reaktionsgemisch wird  filtriert, der Filterrückstand sorgfältig mit heissem  Benzol ausgewaschen und das Filtrat im Vakuum bei  40"     Badtemperatur    eingedampft.

   Die vollständige  Entfernung des     Cellosolves    gelingt am einfachsten  durch wiederholtes Aufnehmen des Rückstandes in  Benzol. Das so erhaltene Rohprodukt wird in 4,12       Volumteilen    wasserfreiem     Pyridin    gelöst, mit 2,62       Volumteilen        Essigsäureanhydrid    vermischt und 16 Std.    bei Raumtemperatur stehengelassen.

   Man dampft  hierauf bei einem Druck von etwa 1 mm     Hg    ein, löst  die zurückgebliebene kristalline Substanz in     Äther-          Benzol-Gemisch   <B>(3:</B> 1), schüttelt die Lösung zunächst  anhaltend mit Wasser, dann in rascher Folge mit     0,1-          n.    Essigsäure, Wasser,     2proz.        Natriumhydrogencar-          bonat-Lösung    und Wasser und destilliert nach dem  Trocknen mit Natriumsulfat das Lösungsmittel im  Vakuum ab.

   Aus Methanol kristallisieren 0,1954 Ge  wichtsteile (82,6% d.     Th.)        3ss-Acetoxy-11ss,12,l-oxido-          5a,22a-spirostan    in     glänzenden    Plättchen vom F.  = 186,5-194,5 . Durch erneutes     Umlösen    aus Me  thanol steigt der Schmelzpunkt auf 188-194,5 .

  A solution of 1.020 parts by weight of the above-described 3.ss-acetoxy-lla, 23; -dibromo-12ss-oxy-5a, 22a-spirostans in 19.4 parts by volume of what is serfree pyridine is freshly precipitated together with 1.5 parts by weight dried silver oxide for 22 hours in the dark.   <I> Example 3 </I> A solution of 0.0473 parts by weight of 3ss-acetoxy-11ss, 12ss-oxido-5a, 22a-spirostane in 3.5 parts by volume of peroxide-free dioxane is mixed with 0.50 parts by volume of aqueous 4-n.   The compound used as the starting material in this example can be obtained as follows: To a boiling solution of 0.276 parts by weight of 3ss-acetoxy-11f, 12ss-oxido-23e-bromo-5a, 22a-spirostane in 27.6 parts by volume of Cellosolve is added 8.20 parts by weight of zinc dust in five portions over the course of 4 hours with constant stirring and then simmered for a further 4 hours with stirring. PATENTANSPRUCH Verfahren zur Herstellung von in 12-Stellung un- substituierten gesättigten oder ungesättigten 11-Keto- spirostanen, dadurch gekennzeichnet, dass man ent sprechende 11,12ss-Oxyde mit Halogenwasserstoff säuren spaltet, in den erhaltenen 12a-11ss-Halogen- hydrinen die 11 ständige Hydroxylgruppe zur Keto- gruppe oxydiert und das Halogenatom in 12-Stellung reduktiv entfernt. UNTERANSPRÜCHE 1.
