Description
Verfahren zur Herstellung von Oxypregnanen Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von gesättigten und un gesättigten, in 13-Stellung durch eine freie oder funktionell abgewandelte Carbinol-, Formyl- ' oder Carboxylgruppe substituierten 14-Oxy-pregnanen. Die neuen 14-Oxy-pregnane und ihre Derivate, wie z. B. das 14-Oxy-aldosteron, zeigen gegenüber den in 14-Stellung nicht hydroxylierten Verbindungen eine gesteigerte Wirksamkeit.
Die neuen 14-Oxy-pregnane werden erhalten, wenn man die entsprechenden, in 14-Stellung unsub- stituierten Verbindungen der Einwirkung von Kul turen von Pilzen der Gattungen Mucor, Helicostylum, Pleospora oder Curvularia oder von entsprechenden Enzymen unterwirft.
Die Ausgangsstoffe weisen vorzugsweise ausser dem in 3- und 20-Stellung, vor allem aber in 11- Stellung, eine freie oder funktionell abgewandelte Oxy- oder Oxogruppe auf. Sie können gesättigt sein oder Doppelbindungen enthalten, z.
B. in 1- und/oder 4-Stellung, ferner in 5-, 6-, 7-, 8-, 9 : 11-, 11 : 12- oder 16-Stellung, oder zusätzliche Substituenten be sitzen, wie freie oder abgewandelte Oxy- oder Oxo- gruppen, ferner Epoxygruppen oder Halogenatome, beispielsweise in 2-, 4-, 5-, 6-, 7-, 8-, 9-, 12-, 15-, 16-, 17- oder 21-Stellung, oder Methylgruppen, z. B.
in 17a-Stellung. Die Ausgangsstoffe können, was die Substituenten anbelangt, von beliebiger sterischer Konfiguration sein und auch als Racemate vorlie gen. Sie können auch den sogenannten Nor- und/oder Homo-Reihen, insbesondere der 19-Nor- und D- Homo-Reihe, angehören. Besonders wichtige Aus gangsstoffe sind z.
B. 18-Oxy- und 18-Oxo-proge- steron, 17a,18-Dioxy- und 17a-Oxy-18-progesteron, 18-Oxy- und 18-Oxo-cortexon, 17a,18-Dioxy- und 17a-Oxy-18-oxo-cortexon, Aldosteron, 18-Oxy-cor- ticosteron und -11-dehydro-corticosteron, 11-Epi-18- oxy- und 11-Epi-18-oxo-corticosteron, 17a-Oxy- aldosteron,
18-Oxyhydrocortison, 18-Oxy- und 18- Oxo-cortison, entsprechende 1-Dehydro- und 21- Oxo-verbindungen und bzw. oder 9a-Fluor-, 9a Chlor- oder 12a-Fluor- und 12a-Chlor-derivate, bei spielsweise 9a-Fluor-aldosteron und -1-dehydro- aldosteron, 9a-Fluor-18-oxy-corticosteron und -1 dehydro-corticosteron, Als funktionell abgewandelte Oxygruppen kommen z.
B. mit einer aliphatischen, aromatischen oder heterocyclischen Carbonsäure, beispielsweise Essigsäure, Propionsäure, Benzoesäure oder Furancarbonsäure veresterte oder eine ver- ätherte Oxygruppen, z.
B. die Tetrahydropyranyl- oxy-, Benzyloxy- oder Triphenylmethoxygruppe, in Betracht und als funktionell abgewandelte Oxogrup- pen vorteilhaft ketalisierte bzw.
acetalisierte Oxo- gruppen, abgeleitet insbesondere von einem zwei wertigen Alkohol, wie die Äthylendioxygruppe. Die Ausgangsstoffe können auch, hauptsächlich in 13- Stellung, eine freie oder funktionell abgewandelte Carboxylgruppe aufweisen, in erster Linie eine ver- esterte oder laktonisierte Carboxylgruppe.
Gute Resultate werden vor allen mit folgenden Pilzarten bzw. den entsprechenden Enzymen erzielt: Pleospora gaeumanni, Mucor griseocyanus, Mucor parasiticus, Helicostylum piriforme und Curvularia pallescens. Für die Kultivierung dieser Pilze eignen sich die an sich hierfür bekannten Medien, z. B.
solche mit Zuckern, wie Glukose oder Lactose, mit Peptonen, Maisquellwasser, Sojaprodukten und der gleichen, sowie mit Mineralsalzen, oder aber syn thetische Nährlösungen. Man arbeitet insbesondere unter aeroben Bedingungen, beispielsweise in Schüt telkultur, oder submers unter Rühren und Luft zufuhr. Die genannten Pilze unterscheiden sich von andern Mikroorganismen, z. B. den Bakterien, durch gutes Wachstum unter verhältnismässig einfachen Zuchtbedingungen. Die Reaktion kann wie gesagt in den genannten Pilzkulturen selbst oder aber mit Hilfe der angereicherten oder abgetrennten Enzyme statt finden, also z.
B. in einer Aufschwemmung des ab getrennten Pilzmycels, des homogenisierten Pilz- mycels oder in Filtraten bzw. wasserhaltigen Extrak ten davon.
Die Isolierung der Verfahrensprodukte lässt sich nach bekannten Methoden vornehmen. Ihre Abtren nung kann z. B. durch Extraktion des Reaktions gemisches mit einem organischen Lösungsmittel, z. B. Methylenchlorid oder Essigester, erfolgen. Für die weitere Reinigung des so erhaltenen Extraktes eignen sich besonders Chromatographie, z.B. an Aluminium oxyd oder Silicagel, Anwendung von Verteilungs methoden, z.
B. dem Gegenstromverfahren, oder die Trennung mittels Girard-Reagentien, wie Trimethyl- ammonium- oder Pyridinium-essigsäurehydrazid. An schliessend an diese Reinigungsmethoden oder an ihrer Stelle kann schliesslich aus organischen oder wässrib organischen Lösungsmitteln umkristallisiert werden.
Die erfindungsgemäss erhältlichen 14-Oxy-steroide und ihre Derivate zeichnen sich, verglichen mit den in 14-Stellung nicht hydroxylierten therapeutisch wirksamen Verbindungen, durch eine gesteigerte Wirksamkeit aus. Die Verbindungen weisen wie gesagt in 13-Stellung eine freie oder funktionell " abgewandelte Carbinol-, Formyl- oder Carboxyl- gruppe und vorzugsweise in 11-Stellung eine freie oder funktionell abgewandelte Oxy- oder Oxogruppe auf; es kann sich also z.
B. um Ester, Äther, Thio- ester, Thioäther, Thiol- und Thionester, Acetale, Mercaptale, Ketale, Enolderivate, wie Enolester, Enoläther oder Enamine, Hydrazone, Semicarbazone und dergleichen handeln.
Von den Verfahrenspro dukten sind von besonderem Interesse: 14a,18-Dioxy- und 14a-Oxy-18-oxo-progesteron, 14a,17a,18-Trioxy- und 14a,17a-Dioxy-18- oxo-progesteron, 14a-18-Dioxy- und 14a-Oxy-18-oxo-cortexon, 14a,17a,18-Trioxy- und 14a,17a-Dioxy-18- oxo-cortexon, 14a-Aldosteron, 14a,18-Dioxy-corticosteron und -11-dehydro- corticosteron, 11-Epi-14a,
18-dioxy- und 11-Epi-14a#-oxy-18- oxo-corticosteron, 17a-Oxy-aldosteron, 14a,18-Dioxy-hydrocortison, 14a,18-Diöxy- und 14a-Oxy-18-oxo-cortison, die entsprechenden 1-Dehydro sowie 21-Oxo-verbin- dungen, ferner entsprechende funktionelle Derivate, wie Ester, Äther, Halogen-Derivate, z.
B. 9a- oder 12a-Halogen-, insbesondere die Fluor- oder Chlor verbindungen, beispielsweise 14a-Oxy-9a-fluor-aldo- steron und -1-dehydro-aldosteron, 14a,18-Dioxy-9a- fluor-corticosteron und -1-dehydro-corticosteron. Sofern die Verfahrensprodukte nicht die Konfigura- tion und die Substituenten von therapeutisch ver wendbaren Steroiden aufweisen, können sie als Zwi schenprodukte zu deren Herstellung, z. B. der oben genannten Verbindungen dienen.
Die erfindungsgemäss erhältlichen Umsetzungs produkte lassen sich in an sich bekannter Weise in ihre funktionellen Derivate, wie Sauerstoff-, Schwe fel- oder Stickstoffderivate, beispielsweise Ester, Äther, Enolester, Enoläther, Ketale, Thioäther und Thioketale, ferner Hydrazone, Oxime und Enamine überführen, ferner können Oxy- zu Oxogruppen dehydriert werden.
In den Estern und Enolestern können die Säurereste solche beliebiger organischer oder anorganischer Säuren sein, wie aliphatischer, alicyclischer, araliphatischer, aromatischer oder heterocyclischer Carbon-, Thioncarbon-, Thiolcarbon- oder Sulfonsäuren, vorzugsweise der Ameisensäure, Essigsäure, Chloressigsäuren, Trifluoressigsäure, Pro- pionsäure, Buttersäuren,
Valeriansäuren, Trimethyl- essigsäure, Diäthylessigsäure, Capronsäuren, Önanth- säuren, Caprylsäuren, Palmitinsäuren, Crotonsäure, Undecansäure, Undecylensäure, Oxalsäure, Bern steinsäure, Pimelinsäure, Maleinsäure, Milchsäure, Carbaminsäuren, Alkoxycarbonsäuren,
ss-Cyclopentyl- propionsäure, Hexahydrobenzoesäure, Benzoesäure, Phenylessigsäure, Cyclohexylessigsäure, Phenylpro- pionsäuren, Trimethylgallussäure, Phthalsäure, Furan-2-carbonsäure, Isonicotinsäure, Methansulfon- säure, Toluolsulfonsäure, Schwefelsäuren, Halogen wasserstoffsäuren oder Phosphorsäuren.
In den erhaltenen Verbindungen lassen sich funk tionell abgewandelte Oxy- oder Oxogruppen in freie Gruppen überführen. Auf diese Weise können ins besondere in polysubstituierten Derivaten die funk tionell abgewandelten Gruppen auch teilweise in Freiheit gesetzt werden, z. B. durch chemische oder enzymatische Hydrolyse, beispielsweise unter Ver wendung saurer oder basischer Mittel, durch Um- esterung oder Umacetalisierung. Aus den auf diese Weise oder auch direkt erhaltenen, nur partiell abge wandelten, wie veresterten bzw. verätherten Deriva ten lassen sich durch anschliessende funktionelle Ab wandlung, z.
B. Veresterung oder Verätherung, poly substituierte Derivate, insbesondere auch gemischte Ester oder Äther bzw. Ester-Äther, herstellen. Sind während der Hydrolyse, insbesondere derjenigen mit alkalischen Mitteln, 9,11- oder 12,11-Halogenhydrine in die entsprechenden 9,11- oder 11,12-Oxidover- bindungen umgewandelt worden, so lassen sich letztere durch Einwirkung von Halogenwasserstoff säuren, insbesondere Fluor- oder Chlorwasserstoff säure, wieder in die gewünschten 9,11- bzw. 12,11- Halogenhydrine verwandeln.
<I>Beispiel 1</I> Zu einer bei 28 gut entwickelten, 4 Tage alten Schüttel-Kultur von Pleospora gaeumanni in 500 cm3 70proz. wässriger Bierwürze mit 0,5 em3 Spermöl gibt man unter sterilen Bedingungen eine Lösung von 125 mg Aldosteron in 10 cm3 Aceton. Die Suspen- sion schüttelt man weitere 4 Tage bei der gleichen Temperatur. Nun wird das Mycel abgetrennt und gut mit Wasser und Essigester gewaschen. Die ver einigten klaren Lösungen werden mit Essigester aus geschüttelt.
Die Essigester-Lösungen wäscht man mit Wasser, trocknet sie und dampft sie im Vakuum ein. Der Rückstand wird in 80proz. Methanol gelöst und mehrmals mit Petroläther ausgeschüttelt. Die Äthanol- Lösungen dampft man hierauf im Vakuum voll ständig ein.
Das Papierchromatogramm des Rück standes (Propylenglykol-Toluol) zeigt neben wenig Aldosteron das etwas langsamer laufende 14a-Oxy- aldosteron. Der ganze Rückstand wird mittels eines präparativen Papierchromatogramms (Propylenglykol- Toluol) aufgetrennt. Die dem 14a-Oxy-aldosteron entsprechenden Zonen werden ausgeschnitten und mit 50proz. Methanol mehrmals extrahiert.
Das Methanol entfernt man dann in Vakuum, schüttelt mehrmals die zurückbleibende wässrige Lösung mit Essigester aus, wäscht die vereinigten Essigester- Lösungen mit Wasser, trocknet sie und dämpft sie im Vakuum ein, wobei als Rückstand reines 14a- Oxy-aldosteron erhalten wird.
Die Inkubation des Aldosterons kann man auch in 500 cm3 einer gut entwickelten wässrigen Kultur von Curvularia pallescens ausführen, die folgende Zusätze enthält:
5 g Rohrzucker, 5 g Difco-Trypton, 1 g Natriumnitrat, 0,5 g sek. Kaliumorthophosphat, 0,25g Magnesiumsulfat, 0,25g Kaliumchlorid, 5 mg Eisensulfat-heptahydrat, 1,25 g Calciumcarbonat und 0,5 cm3 Spermöl. Die Aufarbeitung geschieht wie oben angegeben.
Das so gewonnene 14a-Oxy-aldosteron lässt sich wie folgt verestern: 100 mg werden mit 0,2 cm3 Pyridin und 0,4 cm3 Acetanhydrid übergossen. Die Lösung lässt man 15 Stunden bei 20 stehen und dampft sie dann auf Wasserzusatz im gewöhnlichen Vakuum und zuletzt im Hochvakuum bei 35 .
Als Rückstand erhält man das 14a-Oxy-aldosteron- 18,21-diacetat. <I>Beispiel 2</I> Zu einer bei 28 gut entwickelten, 4 Tage alten Schüttel-Kultur von Pleospora gaeumanni in 500 cm3 70proz. wässriger Bierwürze mit 0,
5 cm3 Spermöl gibt man unter sterilen Bedingungen eine Lösung von 125 mg 1-Dehydro-aldosteron (erhältlich aus Aldosteron durch mikrobiologische Dehydrierung in 1-Stellung mittels des Pilzes Colo nectria decora) in 10 cm3 Aceton. Die Suspension schüttelt man weitere 4 Tage bei der gleichen Temperatur.
Das Reaktionsgemisch wird dann wie im Beispiel 1 angegeben aufgearbeitet und das gebildete 1-De- hydro-14a-oxy-aldosteron ebenfalls wie angegeben mittels eines präparativen Papierchromatogramms (Propylenglykol-Toluol) isoliert, wobei. es etwas lang samer läuft als das 1-Dehydro-aldosteron.
Wenn man das so gewonnene 1-Dehydro-14a- oxy-aldosteron mit 0,2 cm3 Pyridin und 0,4 cm3 Acetanhydrid übergiesst, die Lösung 15 Stunden bei 20 stehenlässt und dann im gewöhnlichen Vakuum und schliesslich im Hochvakuum bei 35 eindampft, so erhält man als Rückstand das 1-Dehydro-14a- oxy-aldosteron-18,21-diacetat.
<I>Beispiel 3</I> 125 mg (18 -> l lss)-Lakton der 44-3,20-Dioxo- llss-oxy-pregnen-18-säure werden wie in Beispiel 1 und 2 mit einer Kultur von Pleospora gaeumanni umgesetzt. Das Umsetzungsprodukt wird ebenfalls wie dort aufgearbeitet und das entstandene (18<B>--></B> 11ss)- Lakton der 44-3,20-Dioxo-llss,l4a-dioxy-pregnen- 18-säure wie angegeben isoliert.

The fungi mentioned differ from other microorganisms, e.g. B. the bacteria, through good growth under relatively simple cultivation conditions. As I said, the reaction can take place in the mentioned fungal cultures themselves or with the help of the enriched or separated enzymes, so z.

Chromatography is particularly suitable for the further purification of the extract obtained in this way, e.g. on alumina or silica gel, application of distribution methods, z.

The 14-oxy-steroids obtainable according to the invention and their derivatives are distinguished by an increased effectiveness compared with the therapeutically active compounds which are not hydroxylated in the 14-position. As mentioned, the compounds have a free or functionally modified carbinol, formyl or carboxyl group in the 13-position and preferably a free or functionally modified oxy or oxo group in the 11-position;

The suspension is shaken for a further 4 days at the same temperature. The mycelium is then separated off and washed well with water and ethyl acetate. The ethyl acetate solutions are washed with water, dried and evaporated in vacuo. The residue is in 80 per cent. Dissolved methanol and extracted several times with petroleum ether. The ethanol solutions are then completely evaporated in a vacuum.
The paper chromatogram of the residue (propylene glycol-toluene) shows, in addition to a little aldosterone, the somewhat slower 14a-oxyaldosterone. The entire residue is separated by means of a preparative paper chromatogram (propylene glycol-toluene). The zones corresponding to 14a-oxy-aldosterone are cut out and treated with 50%. Methanol extracted several times.
The methanol is then removed in vacuo, the remaining aqueous solution is shaken out several times with ethyl acetate, the combined ethyl acetate solutions are washed with water, dried and evaporated in vacuo, pure 14a-oxyaldosterone being obtained as residue.
The incubation of aldosterone can also be carried out in 500 cm3 of a well-developed aqueous culture of Curvularia pallescens containing the following additives:
5 g cane sugar, 5 g Difco tryptone, 1 g sodium nitrate, 0.5 g sec. Potassium orthophosphate, 0.25 g magnesium sulfate, 0.25 g potassium chloride, 5 mg iron sulfate heptahydrate, 1.25 g calcium carbonate and 0.5 cm3 sperm oil. The work-up is carried out as indicated above.
The 14a-oxy-aldosterone obtained in this way can be esterified as follows: 0.2 cm3 of pyridine and 0.4 cm3 of acetic anhydride are poured over 100 mg. The solution is left to stand for 15 hours at 20 and then evaporated with the addition of water in a normal vacuum and finally in a high vacuum at 35.
The residue obtained is 14a-oxy-aldosterone-18,21-diacetate. <I> Example 2 </I> For a shaking culture of Pleospora gaeumanni well developed at 28 and 4 days old in 500 cm3 70 percent. watery wort with 0,
5 cm3 of sperm oil is given under sterile conditions a solution of 125 mg of 1-dehydro-aldosterone (obtainable from aldosterone by microbiological dehydration in 1-position using the fungus Colo nectria decora) in 10 cm3 of acetone. The suspension is shaken for a further 4 days at the same temperature.
The reaction mixture is then worked up as indicated in Example 1 and the 1-dehydro-14a-oxy-aldosterone formed is also isolated as indicated by means of a preparative paper chromatogram (propylene glycol-toluene), with. it runs a little slower than the 1-dehydro-aldosterone.
If the 1-dehydro-14aoxy-aldosterone obtained in this way is poured over with 0.2 cm3 of pyridine and 0.4 cm3 of acetic anhydride, the solution is left to stand for 15 hours at 20 and then evaporated in a normal vacuum and finally in a high vacuum at 35, this gives 1-dehydro-14a-oxy-aldosterone-18,21-diacetate is the residue.
<I> Example 3 </I> 125 mg (18 -> lss) lactone of 44-3,20-dioxolss-oxy-pregnen-18-acid are obtained as in Examples 1 and 2 with a culture of Pleospora gaeumanni implemented.
