bezüglich der Iminogruppe der D-Form entspricht, einer eine Umlagerung der D-Form in die L-Form bewirkenden Behandlung unterwirft. Die die Umlagerung bewirkende Behandlung erfolgt zweckmässigerweise durch Erhitzen.
Schon seit langer Zeit ist Digenea simplex Ag. als Wurmmittel bekannt, doch war es erst kürzlich möglich, seine Wirksubstanz zu bestimmen. Shinzo Murakami und Tsunematsu Takemoto gelang es endlich, die Wirksubstanz, die sie Kaininsäure (J. Pharm. Soc. Japan, 73, 1055 [1953]) nannten, zu extrahieren und ihre Konstitution aufzuklären.
Es handelt sich um das 2-Carboxy-3-carboxymethyl-4- isopropenyl-pyrrolidin (J. Pharm. Soc. Japan 75, 840 [1955]) der Formel:

Wie aus dieser Formel ersichtlich ist, hat die Kaininsäure theoretisch acht Isomere. Es wurden in der Tat in Digenea simplex Ag. zwei Isomere fest gestellt, die Kaminsäure resp. Allokaininsäure benannt wurden.
Eine stereochemische Untersuchung dieser Substanzen ergab, dass die Carboxylgruppe am Kohlenstoffatom in 2-Stellung L-Konfiguration auf weist, wie dies im allgemeinen bei natürlichen Amino- säuren der Fall ist. Die Substituenten an den Kohlen stoffatomen in 2- und 3-Stellung sind zueinander in trans-Stellung angeordnet.
Im übrigen wurde fest gestellt, dass die Substituenten an den Kohlenstoff atomen in 3- und 4-Stellung bei Kaininsäure cis- und bei Allokaininsäure trans-Konfiguration aufweisen.
Es wurde ferner gefunden, dass ein Isomer von Kainin- säure und Allokaininsäure existiert, bei welchem nur die Konfiguration am C2-Atom umgekehrt ist, so dass die Carboxylgruppe am C2-Atom D-Konfiguration aufweist (Proceedings of the Japan Academy 32, 41 [1956]).
Zu Unterscheidungszwecken werden die früher gefundene Kaininsäure und Allokaininsäure a-Kaininsäure resp. a-Allokaininsäure genannt.
Wie bereits in J. Pharm. Soc. Japan 73, 1026;<B>1055</B> (1953) und 75, 1253 (1955), berichtet wurde, besitzen die a-Kaininsäure und die a-Allokaininsäure eine starke anthelminthische Wirksamkeit, während die ss-Kainin- säure und die ss-Allokaininsäure diese Wirksamkeit nicht aufweisen.
Wenn es also möglich ist, ss-Kaininsäure und ss-Allokaininsäure durch Umkehrung ihrer Konfigu ration am C2-Atom in die entsprechenden a-Verbin- dungen überzuführen, so gelangt man zu wertvollen anthelminthisch wirksamen Substanzen. Dasselbe gilt für ss-Kaininsäure- und ss-Allokaininsäurederivate, die sich in die entsprechenden Stammverbindungen über führen lassen.
Zu diesem Zweck ist es nötig, die Konfiguration am C2-Atom umzukehren, ohne jedoch die Konfigu ration an den C-Atomen 3 und 4 zu verändern. Es wurde nun gefunden, dass Verbindungen der ss-Gruppe leicht in die entsprechenden a-Verbindungen über geführt werden können, indem man ihre molekulare Energie erhöht, z. B. durch Erhitzen.
Die zur Durchführung des vorliegenden Verfahrens verwendeten Ausgangsmaterialien können durch die folgenden Formeln dargestellt werden:

Die Ausgangsmaterialien können z.
B. wie. folgt hergestellt werden N-Acetyl-kaininsäureanhydrid (J. Pharm. Soc. Japan 75, 74 [1955]) ergibt ss-N-Acetyl-kainins4ure, wenn man eine wässrige Lösung des ersteren kocht und bei vermindertem Druck eindampft.
Durch Hydrolyse von ss-N-Acetyl-kaininsäure mittels Alkali erhält man ss-Kaininsäure. Wenn N-Acetyl-kaininsäureanhydrid mit Methanol umgesetzt wird, so erhält man ss-N- Acetyl-kaininsäure-2-monomethylester. Wenn man von diesen Verbindungen ausgeht, so erhält man durch übliche Veresterung die entsprechenden Diester,
durch Hydrierung die entsprechenden Dihydrover- bindungen und durch Oxydation die ss-2-Carboxy- 3 -carboxymethyl -4- acetyl - pyrrolidin - Verbindungen.
Bei Behandlung von ss-Kaininsäure oder ihren Mono- oder Diestern mit einer Mineralsäure erhält man ihr Lakton infolge Laktonbildung zwischen der Doppel bindung der Isopropenylgruppe und dem Carboxyl der Carboxymethylgruppe. Die ss-Allokaininsäure und ihre Derivate können durch analoge Behandlungen aus N-Acetyl-allokaininsäureanhydrid erhalten wer den.
Es wird angenommen, dass die Verbindungen der ss-Reihe durch Erhöhung ihrer molekularen Energie in die entsprechenden a-Verbindungen übergeführt werden, wobei sie infolge Umkehrung der Anordnung der Carboxylgruppe C2 Atom eine beständigere Kon figuration annehmen.
Die geeignetste und zweckmässigste Methode zur Erhöhung der molekularen Energie ist das Erhitzen. Um die molekulare Energie durch Erhitzen zu erhöhen, wird das Material vorzugsweise soweit als möglich in einem solchen Zustand gehalten, dass seine Moleküle frei beweglich sind. Aus diesem Grunde wird das Material, wenn es fest ist, in der flüssigen Phase erhitzt. Falls sich das Material beim Schmelzen zersetzt oder gar- nicht schmilzt, wie die ss-Kaininsäure, so wird es als Lösung in Wasser, in einem wässrigen Lösungs mittel oder in einem organischen Lösungsmittel, wie z. B. Alkohole, erhitzt.
Die Umlagerung verläuft vor zugsweise in einem neutralen oder leicht sauren oder alkalischen Medium. Falls sich das Material ver dampfen lässt, so kann es in der Dampfphase um gelagert werden. Zu diesem Zweck wird es destilliert oder in einem geschlossenen Gefäss erhitzt. Falls die Iminogruppe des Materials z. B. mit einer Acylgruppe substituiert ist, oder falls die Carboxylgruppe ver- estert ist, so können diese Substituenten je nach den Reaktionsbedingungen im Verlaufe der Reaktion mehr oder weniger angegriffen werden.
<I>Beispiel 1</I> Einer Lösung aus 1 g ss-N-Acetyl-kaininsäure in 10 cm3 Wasser werden 6 g Bariumhydroxyd zugesetzt, und das Gemisch wird während etwa 6 Stunden in einer geschlossenen Röhre bei 200' C erhitzt. Das Reaktionsgemisch wird mit 60 cm3 Wasser ausge waschen, und das entstandene Bariumsalz wird filtriert und mit 20 cm3 Wasser gewaschen. Das Bariumsalz wird in 30 cm3 Wasser suspendiert und durch die tropfenweise Zugabe von verdünnter Schwefelsäure (unter Vermeidung eines Überschusses der Säure) zersetzt.
Das Bariumsulfat wird abfiltriert, und das Filtrat wird bei vermindertem Druck konzentriert, wobei man farblose Nadeln vom Smp. 245' C (Zers.) erhält. Die Ausbeute beträgt 0,7 g (84 l0). Nach Umkristallisieren aus Wasser schmilzt das Produkt bei<B>251'</B> C (Zers.) und zeigt keine Schmelzpunkt- depression, wenn es mit a-Kaininsäure vermischt wird.

Das farblose ölige Produkt, das bei 210' C/3 mm überdestilliert, ver festigt sich bei Raumtemperatur. Die durch Analyse ermittelten Werte entsprechen denjenigen des a-N- Acetyl-kaininsäure-methylester-laktons.

Das Produkt ist mit dem a-Kaininsäure- lakton, Smp. <B>276'</B> C, identisch, welches durch Lakton- bildung mittels konz. Schwefelsäure aus a-Kainin- säure, die aus Digenea simplex Ag. extrahiert wurde, erhältlich ist.
<I>Beispiel 9</I> Eine Lösung aus 2,3 g ss-Kaininsäure-lakton in 100 cm3 2%iger Natriummethoxydlösung wird wäh rend etwa 2 Stunden unter Rückfluss erhitzt. Hierauf wird dem Gemisch unter Kühlen mit Eiswasser trockene Salzsäure zugeführt, um Natriumchlorid auszufällen.
Nachdem das Gemisch mit HCl-Gas gesättigt ist, wird es über Nacht stehen gelassen. Das Reaktionsgemisch wird bei einer 'Temperatur unter halb von 40' C verdampft, und der Rückstand, der eine grosse Menge Natriumchlorid enthält, wird in 100 cm3 Wasser gelöst. Die Lösung wird mit Natrium- carbonat gesättigt und, nachdem das ausgeschiedene Natriumchlorid abfiltriert wurde, mit 14 Portionen zu 50 cm3 Essigsäure-äthylester extrahiert.
Die vereinigten Extrakte werden über wasserfreiem Natriumsulfat getrocknet, und das verwendete Lösungsmittel wird verdampft. Hierauf wird der ölige Rückstand (ss-Iso- kaininsäure-dimethylester) nochmals im Vakuum de stilliert, wobei man ein farbloses öliges Destillat erhält.
Es handelt sich bei diesem Produkt um a-Isokainin- säure-dimethylester, Sp. 149' C, [a]D = 30,2 <B>L0,5'</B> (C = 0,993%, CH'30H). Dieses Produkt entspricht der planaren Strukturformel von a-Kaininsäure-di- methylester. Die Ausbeute beträgt 1,6 g.

Das Reaktionsgemisch wird mit Aktivkohle behandelt und bei vermindertem Druck auf etwa 1 cm3 eingeengt. Nach Zugabe von Methanol wird der Rückstand an einem kühlen Ort stehen gelassen. Die ausgeschiedenen Kristalle von ss-Allo-2-carboxy-3-carboxymethyl-4-acetyl-pyrrolidin werden aus Wasser umkristallisiert. Smp. <B>213'</B> C (Zers.); [a]" -<B>-23,5'</B> (1,0%, H20).
Eine Lösung aus 500 mg dieses Produktes in 20 cm3 Wasser wird während 8 Stunden in einer geschlossenen Röhre bei etwa 160-180' C erhitzt. Das Reaktions gemisch wird mit Aktivkohle behandelt und bei ver mindertem Druck auf etwa 1 cm3 eingeengt. Der Rückstand wird an einem kühlen Ort stehen gelassen, worauf sich Kristalle auszuscheiden beginnen, die aus Wasser umkristallisiert werden. Man erhält a-Allo- 2-carboxy-3-carboxymethyl-4-acetyl-pyrrolidin vom Smp. 205 C (Zers.); [a]D = -19,2 (1,0%, H20). Die Ausbeute beträgt 250 mg.

The same applies to ß-kainic acid and ß-allokainic acid derivatives, which can be converted into the corresponding parent compounds.
For this purpose it is necessary to reverse the configuration at the C2 atom, but without changing the configuration at the C atoms 3 and 4. It is assumed that the compounds of the ss series are converted into the corresponding a compounds by increasing their molecular energy, whereby they adopt a more stable configuration as a result of the reversal of the arrangement of the carboxyl group C2 atom.
The most suitable and practical method for increasing the molecular energy is heating. In order to increase the molecular energy by heating, the material is preferably kept as far as possible in such a state that its molecules can move freely.
