Verfahren zur Herstellung neuer Pyriinido[4,5-bjchinoxaiine
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 10-substituierten 4-Oxy-5-acyl-5, 1 O-dihydro-pyrimido[4,5-b]- chinoxalinen der Formel

worin R für O, S oder NH steht und Rt einen gegebenenfalls durch eine oder mehrere Hydroxylgruppen substituierten Alkylrest bedeutet, oder von im Benzolring A substituenten oder ankondensierte aromatische Ringe tragenden Derivaten davon. Als Substituenten R1 kommt z. B. die Methyl- oder Ribitylgruppe in Frage; als Acylreste kommen in erster Linie diejenigen niederer aliphatischer Carbonsäuren, vor allem der Essigsäure, in Betracht und als Substituenten des Benzolrestes A vor allem Methylgruppen, besonders in 7- und 8-Stellung.
Es war bisher nicht gelungen, stabile 5, lO-Di- hydropyrimido[4,5-b]chinoxaline herzustellen. Wohl war es bekannt, dass Lumiflavin (2,10-Dihydro-2-oxo-4-oxy-7,8, 1 0-trimethyl- pyrimido4,5-b] chinoxalin) der Formel

mit Reduktionsmitteln ein Dihydroderivat liefert; dieses ist jedoch wegen seiner leichten Oxydierbarkeit zum Ausgangsmaterial nie gefasst worden und konnte auch nicht zu etwaigen Umsetzungen verwendet werden.
Es wurde nun gefunden, dass man stabile Verbindungen der Formel I oder deren im Benzolring A substituente oder ankondensierte aromatische Ringe tragende Derivate erhalten kann, wenn man Verbindungen der Formel

oder deren im Benzolring A substituenten oder ankondensierte aromatische Ringe tragende Derivate in Gegenwart von acylierenden Mitteln mit naszierendem oder katalytisch erregtem Wasserstoff reduziert. Als Ausgangsstoffe kommen z. B. Lumiflavin, 2-Thiono-leukolumiflavin und 2-Imino-leuko- lumiflavin in Betracht. Geeignete acylierende Mittel sind z. B. Essigsäureanhydrid, Propionsäureanhydrid, Ameisensäure. Die Reduktion kann z. B. in Gegenwart eines Platin- oder Palladiumkatalysators durchgeführt werden. Besonders vorteilhaft arbeitet man mit Zink in Gegenwart von Essigsäure und Essigsäureanhydrid.
Am Beispiel des Lumiflavins kann die Reaktion wie folgt illustriert werden:

In den erhaltenen Acylverbindungen können wasserstofftragende Ringstickstoffatome des Pyrimidinringes in üblicher Weise durch Alkylreste, wie Methylreste, substituiert werden, vorzugsweise durch Behandlung mit reaktionsfähigen Estern der entsprechenden Alkohole, z. B. mit Alkylhalogeniden, Dialkylsulfaten oder Arylsulfonsäurealkylestem.
Die erfindungsgemäss hergestellten, neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. Insbesondere wirken sie hemmend auf das Wachstum von Tumorzellen. Weiter besitzen sie bakteriostatische Eigenschaften, speziell gegen gram-positive Bakterien, wie Streptococcen. Die neuen Verbindungen sollen als Heilmittel oder als Zwischenprodukte zur Herstellung solcher verwendet werden.
Die erfindungsgemäss erhaltenen Verbindungen können je nach den Substituenten in freier Form oder als Salze mit Metallen oder Säuren vorliegen. Als Metalle sind in erster Linie Alkali- oder Erdalkalimetalle und als Säuren organische, besonders aber anorganische Säuren, wie Halogenwasserstoff-, Schwefel- oder Phosphorsäuren oder Salpetersäure zu nennen.
Die als Ausgangsstoffe verwendeten Verbindungen sind zum Teil bekannt, wie z. B. Lumiflavin, 2-Thionolumiflavin oder 2-Lumiflavimin.
In den nachfolgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
In eine siedende Suspension von 0,5 g
7,8, O-Trimethyl-2-oxo-4-hydroxy-2, 10 dihydro-pyrimido [4, 5-b] chinoxalin in 20 cm3 Acetylierungsgemisch (Essigsäureanhydrid : Essigsäure: Schwefelsäure = 50:50: 1) wird langsam Zinkstaub bis zur bleibenden Entfärbung eingetragen. Da das Metall schnell desaktiviert wird, muss man einen Überschuss in kleinen Portionen zugeben. Das Lumiflavin geht dabei allmählich ganz in Lösung, es bildet sich zunächst ein tiefrotes Semichinon, dann das farblose
7,8,1 0-Trimethyl-5-acetyl-2, 4-dihydroxy-
5,1 0-dihydro-pyrimido [4, 5-b] chinoxalin.
Wenn die überstehende Lösung nur noch hellgelb ist, filtriert man schnell siedendheiss durch eine Glasfritte, wäscht mit heisser Essigsäure gut nach und dampft das Filtrat am Vakuum ein. Den sirupösen, hellgelben Rückstand nimmt man mit 50 cm: in Natriumacetatlösung auf. Nach kurzem Schütteln fällt ein mikrokristalliner Niederschlag aus. Man lässt einige Stunden bei 00 absitzen und filtriert.
Der cremefarbene Rückstand wird in in wässrigem Ammoniak gelöst, die Lösung mit Tierkohle geschüttelt und nach Filtration mit 2nEssigsäure neutralisiert. Das 7,8,10-Trimethyl-5-acetyl-2,4-dihydroxy-
5,1 O-dihydro-pyrimido [4,5-b]chinoxalin fällt in farblosen, zu Sternen geordneten Stäbchen aus.
Ausbeute 0,45 g = 77 6/o der Theorie. Die Substanz hat keinen charakteristischen Schmelzpunkt. Bei etwa 2800 tritt langsame dehydrierende Acetolyse, Abspaltung von Essigsäure unter Bildung von
7,8, 10-Trimethyl-2-oxo-4-hydroxy-2, 10 dihydro-pyrimido[4,5-bjchinoxalin ein. Totalzersetzung findet bei 320 statt.
Beispiel 2
5 g rohes, rotbraunes
7,8,1 O-Trimethyl-2-thiono-4-hydroxy-2, 10 dihydro-pyrimido[4, 5-b]chinoxalin werden in 50 cm3 eines Gemisches von gleichen Teilen Essigsäure und Essigsäureanhydrid zum Sieden erhitzt und langsam Zinkstaub eingetragen, bis die Farbe der Suspension von tiefviolett nach hellbraun umgeschlagen ist. Dabei geht das Ausgangsprodukt grösstenteils unter Entfärbung in Lösung. Nach 5 Minuten weiteren Siedens tritt eine voluminöse, hellbräunliche Fällung auf. Man filtriert beim Abkühlen und löst den Rückstand in ln Natronlauge. Neben unverbrauchtem Zinkstaub bleibt nur wenig Zinksulfid ungelöst. Das Filtrat wird nach Entfärbung mit Kohle bei Zimmertemperatur mit in Salzsäure versetzt und die entstandene Fällung nach mehrstündigem Absitzen bei 00 abfiltriert.
Nach je einmaligem Umfällen aus in Natronlauge und 2n wässrigem Ammoniak erhält man
7,8,1 O-Trimethyl-5-acetyl-2-thio-4-hydroxy-
5,1 10-dihydro-pyrimido [4, 5-bichinoxalin als cremefarbene, amorphe Substanz. Sie geht bei etwa 2200 unter teilweiser Zersetzung und Abspaltung von Essigsäure in
7,8, 10-Trimethyl-2-thiono-4-hydroxy-2, 10 dihydro-pyrimido[4,5-b]chinoxalin über, welches seinerseits bei 2630 unter Zersetzung zu schmelzen beginnt.
Bei Behandlung des
7,8,1 O-Trimethyl-5-acetyl-2-thio-4-hydroxy-
5,1 0-dihydro-pyrimido{4,5-b]chinoxalins mit Raney-Nickel W-6 in überschüssigem wässrigem Ammoniak wird es in das
7,8,1 O-Trimethyl-5-acetyl-4-hydroxy-5, 10 dihydro-pyrimido[4, 5-b]chinoxalin übergeführt.
Zwecks Methylierung wird das
7,8,1 0-Trimethyl-5-acetyl-2-thio-4-hydroxy-
5,10-dihydro-pyrimido[4,5-b]chinoxalin in ln Natronlauge gelöst und viermal langsam unter Schütteln mit Dimethylsulfat bis zur neutralen Reaktion auf Kongopapier versetzt, wobei jeweils das verbrauchte Alkali wieder ersetzt wird. Zuletzt geht beim Zusatz von Alkali nur noch wenig Substanz in Lösung. Die alkalische Suspension wird daraufhin filtriert, der Rückstand gut mit 1 n Natronlauge und Wasser gewaschen und aus Äthanol zweimal kristallisiert.
Das so erhaltene
3,7,8,1 0-Tetramethyl-5-acetyl-2-methylmercapto-
4-oxo-3 ,4, 5,1 O-tetrahydro-pyrimido- [4, 5-b] chinoxalin bildet feine, an den Enden abgeschrägte, farblose Stäbchen, F. = 2682720 unter geringfügiger Zersetzung.
Das alkalische Filtrat gibt mit 2n Essigsäure einen körnigen, teilweise kristallinen, cremefarbenen Niederschlag, der sich aus Alkohol nicht kristallisieren lässt.
Dieser stellt das
7,8,1 O-Trimethyl-5-acetyl-2-methylmercapto-4- oxy-5, 10-dihydro-pyrimido[4,5-bjchinoxalin dar.
Beispiel 3
1 g Riboflavin (Vitamin BS Ph. H. V. purum) wird in 30 cms eines Gemisches aus gleichen Teilen Eisessig und Acetanhydrid suspendiert. In das siedende Reaktionsgemisch trägt man in kleinen Portionen Zinkstaub ein, bis nach einigen Minuten unter starker Aufhellung und Verschwinden der Fluoreszenz fast aller Farbstoff in Lösung gegangen ist. Es wird etwa 0,5 g Zink benötigt. Danach erhitzt man während weiterer 5 Minuten unter Zugabe von Tierkohle und filtriert das siedendheisse Gemisch schnell durch eine vorgewärmte Glassinternutsche. Das hellgelbe Filtrat wird am Vakuum möglichst zur Trockne eingedampft, mit wenig Alkohol aufgenommen und das Acetylleukoflavin durch Zugabe von Äther gefällt.
Die Roh Ausbeute an cremefarbenem, amorphem Acetylleuko-riboflavin beträgt 1,5 g = 86 0/o Das Produkt enthält etwa 1 äquivalent freie Essigsäure. Im Gegensatz zum freien Riboflavin ist es in Wasser, Alkohol, Aceton, Chloroform und Eisessig gut löslich. Es reagiert mit Diazomethan unter Freisetzung von 2 Äquivalenten Stickstoff. Zur Analyse wird mehrmals aus Alkohol/j2ither umgefällt. Es schmilzt unscharf bei 155 unter Zersetzung. Nach Trocknen während 12 Stunden bei 20 /0, ûl mm enthält es noch 1/2 Mol Essigsäure, welche durch weiteres Trocknen während 12 Stunden bei 110 l0, 01 mm entfernt werden kann.

Suitable acylating agents are e.g. B. acetic anhydride, propionic anhydride, formic acid. The reduction can e.g. B. be carried out in the presence of a platinum or palladium catalyst. It is particularly advantageous to work with zinc in the presence of acetic acid and acetic anhydride.

Example 2
5 g raw, red-brown
7,8,1 O-trimethyl-2-thiono-4-hydroxy-2, 10 dihydro-pyrimido [4, 5-b] quinoxaline are heated to the boil in 50 cm3 of a mixture of equal parts of acetic acid and acetic anhydride and zinc dust is slowly introduced until the color of the suspension has turned from deep purple to light brown. Most of the starting product goes into solution with discoloration. After 5 minutes of further boiling, a voluminous, light brownish precipitate occurs. It is filtered while cooling and the residue is dissolved in 1N sodium hydroxide solution. In addition to unused zinc dust, only a little zinc sulfide remains undissolved. After decolorization with charcoal, hydrochloric acid is added to the filtrate at room temperature, and the resulting precipitate is filtered off after sitting at 00 for several hours.

Example 3
1 g of riboflavin (Vitamin BS Ph. H. V. purum) is suspended in 30 cm of a mixture of equal parts of glacial acetic acid and acetic anhydride. Zinc dust is introduced into the boiling reaction mixture in small portions until, after a few minutes, almost all of the dye has gone into solution with strong lightening and disappearance of the fluorescence. The raw yield of cream-colored, amorphous acetylleuco-riboflavin is 1.5 g = 86%. The product contains about 1 equivalent of free acetic acid. In contrast to free riboflavin, it is easily soluble in water, alcohol, acetone, chloroform and glacial acetic acid. It reacts with diazomethane, releasing 2 equivalents of nitrogen. For analysis, alcohol / j2ither is reprecipitated several times. It melts indistinctly at 155 with decomposition. After drying for 12 hours at 20/0, ûl mm, it still contains 1/2 mol of acetic acid, which can be removed by further drying for 12 hours at 110 l0.01 mm.
