Process for the preparation of new acyl hydrazones
It has been found that new (N-alkyl piperidone) acyl hydrazones of the formula I
EMI1.1
where Ri is alkyl and R2 is hydrogen or lower alkyl groups and R3 is a mono- or polysubstituted aromatic or a heterocyclic radical which can also be substituted, can be obtained by N-alkylpiperidones of the formula II
EMI1.2
wherein Ri has the above meaning with a hydrazide of an aromatic or heterocyclic carboxylic acid of the formula III
EMI1.3
where Ri, and Ro have the same meaning as in formula I,
condensed.
The process can, for example, be carried out in such a way that the suspension of the hydrazide of the formula III in abs. Ethanol with a solution of N-alkyl-4-piperidone, such as. B. 1-methyl-4-piperidone heated.
Fighting tuberculosis using chemical compounds has long been tried. Mention should be made of inorganic and organic gold compounds, which have been abandoned today, as well as sulfones, thiosemicarbazones, mercaptotriazinones, thiohydric compounds, various antibiotics, e.g. B. streptomycin, p-aminosalicylic acid and their derivatives.
A new class of tuberculostatics was found in the series of hydrazides of 4-pyridine-carboxylic acid. For example, isonicotinic acid hydrazide is a recognized tuberculostatic agent which has the greatest activity of all compounds used clinically to date, which is used in both human and veterinary medicine.
It has now been shown that when isonicotinic acid hydrazide is used as well as with other tuberculostatics, the tubercle bacilli easily get used to the drug and become resistant if it is administered over a long period of time.
It is therefore common practice to change medication and it is therefore an advantage if a larger number of well-tolerated tuberculostatics are available.
As is known from the literature, undesirable side effects such as dizziness, especially in older people, limb twitching, constipation, drowsiness, excessive tendon reflexes and the like, are associated with isonicotinic acid hydrazide. a. m., observed.
The new compounds prepared according to the invention are distinguished by various advantages over the already known tuberculostatics.
They are almost completely free of undesirable side effects with lower toxicity, and large doses, which are essential for combating tuberculosis, are well tolerated. Due to their very good solubility in water, which is due to the piperidyl residue, they are particularly suitable for infusion solutions and thus differ from the already known tuberculostatics. But they can also be administered orally or rectally.
As we have shown pharmacological findings, the inhibitory values of the new substances produced in accordance with the process in various strains of tubercle bacilli are of the same order of magnitude as isonicotinic acid hydrazide. On individual human tubercle bacilli strains, total inhibition is achieved at a dilution of 1: 160,000,000, and partial inhibition even at a dilution of 1: 25,000,000.
The new acyl hydrazones produced by the present process are solid, crystallized bases at room temperature which form stable salts with organic and inorganic acids. They should be used therapeutically as tuberculostatics.
In the following examples, all temperatures are given in degrees Celsius. The melting points are corrected.
