Description
Verfahren zur Herstellung von von-Amino-caprylsäure und ihren Estern
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von c. s-Amino-caprylsäure und ihren Estern aus 7-Cyan-2,5-heptadiensäure oder ihren Estern.
Die co-Amino-caprylsäure war bisher nur schwer erhältlich, und dies war ein grosses Hindernis für ihre Verwendung als Monomer zur Herstellung von Polyamidfasern.
Das in der Literatur (Goldgebel, Br. 27, 3128, 1894) beschriebene Verfahren zur Herstellung von co-Amino-caprylsäure besteht darin, dass zunächst mit dem Oxim der 9-Ketosterinsäure eine Beckmannsche Umlagerung durchgeführt wird, worauf das erhaltene Produkt mit konzentrierter Salzsäure bei 180"C zersetzt wird. Unter diesen Bedingungen wird aber nur eine geringe Menge an Aminosäure gebildet, während der restliche Anteil vorwiegend aus Nonylamin, Dekansäure und Azelainsäure besteht.
Das erfindungsgemässe Verfahren ist nun dadurch gekennzeichnet, dass man 7-Cyan-2,5-heptadiensäure oder deren Ester mit Alikoholen mit 1 bis 4 Kohlenstoffatomen hydriert. Die erhaltenen Ester können ohne vorherige Verseifung als solche destilliert und polymerisiert werden. Für die Esterbildung kommen, wie gesagt, nur aliphatische Alkohole mit 1 bis 4 Kohlenstoffatomen in Frage, und zwar aus praktischen Erwägungen1.
Das Hydrierungsverfah, ren kann durch folgendes Schema dargestellt werden:

worin R Wasserstoff oder einen aliphatischen Rest mit 1 bis 4 C-Atomen bedeutet.
Die erfindungsgemässe Hydrierung kann entweder am gereinigten oder am rohen Ausgangsstoff vorgenommen werden.
Wenn bei der Hydrierung Raneynickel verwendet wird, wird die Reaktion mit Vorteil bei einer Tempe rabur zwischen 20 und 1 O00 C und unter einem Was serstoffdruck bis zu 200 Atrnosphären in Gegenwart von Ammoniak durchgeführt, um die Tendenz zur Bildung eines sekundären Amins herabzusetzen.
Das durch die Hydrierung erhaltene Endprodukt kann, wenn es sich dabei um einen Ester handelt, destilliert werden. Es soll aber darauf hingewiesen werden, dass es auf diese Weise schwierig ist, eine teilweise auftretende Polykondensation des Aminoesters zu vermeiden. In diesem Falle ist es günstiger, das Produkt zunächst durch Kochen mit einer Mineralsäure, wie z. B. Salzsäure oder Schwefelsäure, zu verseifen, es hierauf mit Ammoniak zu neutralisieren, zu trocknen und schliesslich die Aminosäure sorgfältig mit absolutem Alkohol zu extrahieren.
Die Aminosäure kann auch über ihr Hydrochlorid (Fp = 145-146 C) gereinigt werden.
Um ein sehr reines Produkt zu erhalten, ist es besser, das Seminitril in zwei Stufen zu hydrieren.
Palladium eignet sich sehr als Katalysator für die selektive Hydrierung der Doppelbindungen und kann, wenn es auf Trägerstoffe aufgebracht ist, auch in sehr geringen Mengen verwendet werdien. Die sehr schnelle Hydrierung findet bei niedrigen Temperaturen statt, vorzugsweise bei Temperaturen zwischen 10 und 800 C unter Wasserstoffdruck. Die darauffolgende Stufe der Hydrierung des Nitrils zur Aminogruppe wird im Gegensatz hiezu nach Abfiltrieren des Palladiumkatalysators mit einem NickelXhrom-Kata- lysator zwischen 20-600 C durchgeführt.
Die Zweistufen Hydrier, ung bietet auch noch andere Vorteile. So erübrigt sich z. B. die Destillation des rohen Ausgangsstoffes, wie er bei der Umsetzung von Chlorcyanobuten mit Acetylen, Kohlenmonoxyd und Methylalkohol anfällt. Anderseits ist es möglich, das Hydrierungsprodukt der ersten Stufe, dessen Doppelbindungen bereits aushydriert sind, das heisst das Suberonseminitril und dessen Ester, zu destillieren. Ein weiterer Vorteil dieses Verfahrens zusätzlich dazu, dass ein stabileres Produkt destilliert werden kann, liegt in der Möglichkeit, gleichzeitig eine geringere Menge Sebacindinitrii zu isolieren, welches durch die Hydrierung von Dicyanoctadien, einem im Ausgangsrohprodukt enthaltenen Nebenprodukt, erhalten wurde.
Ausserdem wird, wenn man zur zweiten Hydrierungsstufe von einem destillierten Seminitril ausgeht, die Bildung von Verunreinigungen auf ein Minimum beschränkt.
Beispiel 1
20 g Methylester von 7-Cyan-2, 5-hepttadiensäure werden in Methanol gelöst und nach Zusatz von 10 g Ammoniak und 5 g Raneynickel in einem Autoklla- ven hydriert.
Die Hydrierung findet t bereits bei 500 C statt.
Das Nickel wird abfiltriert, und der Alkohol wird im Vakuum abgedampft. Es wird so ein Rückstand erhalten, welcher noch zimmer Nickel enthält. Zu dessen Entfernung werden einige Tropfen einer Natrium hydroxydlösung zugesetzt. Nach Extraktion mit Äther wird der Methylester von co-Amino-caprylsäure bei 110 bis 120"C (15 mm Hg) abdestilliert.
Gleichzeitig findet die Bildung von Polykonden sationsprodukten statt.

Beispiel 2
14 g Methylester von 7-Cyan-2,5-heptadiensäure werden 30 Minuten lang bei 30" C in einer Methanol lösung unter einem Wasserstoffdruck von 30 Atmosphären in Gegenwart von 1 g Palladiumkatalysator auf Trägerstoffen hydriert.
Der Katalysator wird hierauf abfiltriert, und es werden 5 g eines NickelChrom-Katalysators und 0,3 g NaOH zugesetzt. Die Hydrierung wird dann 30 Minuten lang bei 300 C unter einem Wasserstoffdruck von 30 Atmosphären fortgesetzt.
Hierauf wird der Katalysator abfiltriert. Die Lösung erscheint klar und farblos. Nun wird der Alkohol abdestilliert und die Lösung drei Stunden lang mit 360/obiger Salzsäure am Rückfluss gekocht. Sie wird dann während des Erhitzens mit Natriumkarbonat neutralisiert, und hierauf werden ungefähr 0,5 g eines basischen Öls mit Äther extrahiert.
Die Lösung wird hierauf mit Schwefelsäure angesäuert, mit Ammoniak neutralisiert und getrocknet.
Der Rückstand wird wiederholt mit absolutem Alko hoi extrahiert, und das Salz wird abfiltriert. Der Alkoholextrakt wird getrocknet, wobei man wAmino- caprylsäure erhält.
Das Hydrochlorid der Säure schmilzt bei 145 bis 146 C.
Beispiel 3
50 g des Rohproduktes aus der Reaktion von Chlorcyanbuten mit Acetylen, CO und Methylalkohol werden in Methylalkohol mit 2 g Palladiumkatalysator hydriert.
Das Produkt wird bei 150=160"C (18 mm Hg) destilliert. Man erhält so 45 g Methylester von Suberonseminitril; als Nebenprodukt sind ferner 3 g Sebacindinitril im Destillationsrückstand enthalten.

Die Weiterverarbeitung zum gewünschten End produkt erfolgt wie im vorhergehenden Beispiel beschrieben. Dabei werden 33 g co-Amino-caprylsäure erhalten.
Beispiel 4
10 g 7-Cyan-2, 5-heptadiensäure werden mit Na triumbicarbonat neutralisiert und in wässriger Äthanollösung zunächst mit Palladium und dann mit Nikkel-Chrom hydriert.
Die Lösung wird abfiltriert, mit Schwefelsäure verseift, mit Ammoniak neutralisiert und getrocknet.
Der trockene Rückstand wird wie vorher beschrieben mit Alkohol extrahiert. Es werden so 8 g w Amino-caprylsäure erhalten.

The present invention relates to a process for the preparation of c. s-Amino-caprylic acid and its esters from 7-cyano-2,5-heptadienoic acid or its esters.
The co-aminocaprylic acid has hitherto been difficult to obtain and this has been a major obstacle to its use as a monomer for the production of polyamide fibers.
The process described in the literature (Goldgebel, Br. 27, 3128, 1894) for the preparation of co-amino-caprylic acid consists in first performing a Beckmann rearrangement with the oxime of 9-ketosteric acid, whereupon the product obtained is treated with concentrated hydrochloric acid is decomposed at 180 "C. Under these conditions, however, only a small amount of amino acid is formed, while the remainder consists mainly of nonylamine, decanoic acid and azelaic acid.
The process according to the invention is now characterized in that 7-cyano-2,5-heptadienoic acid or its esters are hydrogenated with alcohols having 1 to 4 carbon atoms. The esters obtained can be distilled and polymerized as such without prior saponification. As already mentioned, only aliphatic alcohols with 1 to 4 carbon atoms are suitable for ester formation, for practical reasons1.

The hydrogenation according to the invention can be carried out either on the purified or on the crude starting material.
If Raney nickel is used in the hydrogenation, the reaction is advantageously carried out at a Tempe rabur between 20 and 10 000 C and under a hydrogen pressure of up to 200 atmospheres in the presence of ammonia in order to reduce the tendency to form a secondary amine.
The end product obtained by the hydrogenation, if it is an ester, can be distilled. However, it should be pointed out that it is difficult in this way to avoid a partial polycondensation of the amino ester. In this case it is more advantageous to first boil the product with a mineral acid, such as. The amino acid can also be purified via its hydrochloride (mp = 145-146 C).

In contrast to this, the subsequent stage of hydrogenation of the nitrile to the amino group is carried out at between 20 ° and 600 ° C. after the palladium catalyst has been filtered off with a nickel-chromium catalyst.
The two-stage hydrogenation also offers other advantages. So there is no need for B. the distillation of the crude starting material, such as is obtained in the reaction of cyanobutene with acetylene, carbon monoxide and methyl alcohol. On the other hand, it is possible to distill the hydrogenation product of the first stage, the double bonds of which have already been completely hydrogenated, that is to say the suberone-seminitrile and its ester. Another advantage of this process, in addition to the fact that a more stable product can be distilled, is the possibility of simultaneously isolating a smaller amount of sebacine dinitride, which was obtained by the hydrogenation of dicyanoctadiene, a by-product contained in the starting crude product.
In addition, if a distilled seminitrile is used as the starting point for the second hydrogenation stage, the formation of impurities is reduced to a minimum.
example 1
20 g of methyl ester of 7-cyano-2,5-hepttadienoic acid are dissolved in methanol and, after addition of 10 g of ammonia and 5 g of Raney nickel, hydrogenated in an autoclave.
The hydrogenation already takes place at 500.degree.
The nickel is filtered off and the alcohol is evaporated in vacuo. A residue is obtained which still contains nickel. To remove it, a few drops of a sodium hydroxide solution are added. After extraction with ether, the methyl ester of co-amino-caprylic acid is distilled off at 110 to 120 ° C. (15 mm Hg).
At the same time, the formation of polycondensation products takes place.

Example 2
14 g of methyl ester of 7-cyano-2,5-heptadienoic acid are hydrogenated for 30 minutes at 30 ° C. in a methanol solution under a hydrogen pressure of 30 atmospheres in the presence of 1 g of palladium catalyst on supports.
The catalyst is then filtered off and 5 g of a nickel-chromium catalyst and 0.3 g of NaOH are added. The hydrogenation is then continued for 30 minutes at 300 ° C. under a hydrogen pressure of 30 atmospheres.
The catalyst is then filtered off. The solution appears clear and colorless. Now the alcohol is distilled off and the solution is refluxed for three hours with 360% hydrochloric acid. It is then neutralized with sodium carbonate while heating, and then approximately 0.5 g of a basic oil is extracted with ether.
The solution is then acidified with sulfuric acid, neutralized with ammonia and dried.
The residue is extracted repeatedly with absolute alcohol and the salt is filtered off. The alcohol extract is dried, w-amino caprylic acid being obtained.
The acid hydrochloride melts at 145 to 146 C.
Example 3
50 g of the crude product from the reaction of cyanogen chloride with acetylene, CO and methyl alcohol are hydrogenated in methyl alcohol with 2 g of palladium catalyst.
The product is distilled at 150 = 160 ° C. (18 mm Hg). This gives 45 g of methyl ester of suberoneminitrile; the distillation residue also contains 3 g of sebacin dinitrile as a by-product.

The further processing to the desired end product takes place as described in the previous example. 33 g of co-aminocaprylic acid are obtained.
Example 4
10 g of 7-cyano-2, 5-heptadienoic acid are neutralized with sodium bicarbonate and hydrogenated in aqueous ethanol solution first with palladium and then with nickel-chromium.
The solution is filtered off, saponified with sulfuric acid, neutralized with ammonia and dried.
The dry residue is extracted with alcohol as previously described. This gives 8 g w aminocaprylic acid.
