Verfahren zur Herstellung von Nl, N2-disubstituierten Carbonsäurehydraziden
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Ni, N2-disubstituierten Carbon säurehydraziden, welches dadurch gekennzeichnet ist, dass man 1,2-Diisopropyl-hydrazin oder ein Salz davon mit einem den Rest R-CO-abgebenden Acylierungsmittel, worin R einen gegebenenfalls durch Halogenatome, Hydroxyl-oder bis zu 4 Kohlenstoffatome enthaltende Acyloxy-, Alkoxyoder Alkylmercaptogruppen substituierten gesättigten oder ungesättigten cycloaliphatischen Kohlenwasserstoffrest, gesättigten oder ungesättigten, geradkettigen oder verzweigten aliphatischen Kohlenwasserstoffrest oder 5gliedrigen heterocyclischen Rest bedeutet, kondensiert.
Falls R die Bedeutung eines acyloxysubstituierten Restes besitzt, kann dieser gegebenenfalls nachträglich zum entsprechenden hydroxyl- substituierten Rest verseift werden. Die erhaltenen Carbonsäurehydrazide können ferner gegebenenfalls in Salze übergeführt werden.
Als Acylierungsmittel verwendet man z. B.
Säurehalogenide, insbesondere das Säurechlorid, oder Säureanhydride. Geeignete Säureanhydride stellen sowohl die aus 2 Molekülen einer Säure der Formel R-COOH gebildeten symmetrischen Anhydride als auch gemischte Anhydride, z. B. mit niederen Alkancarbonsäuren oder mit Kohlensäuremonoestern dar.
Oft ist es vorteilhaft, wenn die Umsetzung mit molaren Mengen Acylierungsmittel und 1,2-Diisopropylhydrazin vorgenommen wird. Im Falle der Verwendung eines Säurehalogenids erfolgt die Umsetzung mit Vorteil in einem inerten Lösungsmittel, wie z. B.
Benzol, unter Zusatz einer tertiären Base, z. B. Pyri- din oder Triäthylamin, oder in Gegenwart von über- schüssigem 1, 2-Diisopropylhydrazin.
Die so erhaltenen Carbonsäurehydrazide weisen die folgende Formel auf :

Die erfindungsgemäss erhältlichen disubstituierten Carbonsäurehydrazide können in an sich bekannter Weise durch Umsetzung mit anorganischen bzw. organischen Säuren, wie z. B. Salzsäure, Phosphorsäure, Oxalsäure, Weinsäure, in die entsprechenden Salze übergeführt werden.
Die erfindungsgemäss hergestellten Nl, N2-di- substituierten Carbonsäurehydrazide üben eine hemmende Wirkung auf die Monoaminoxydase aus ; einzelne Vertreter zeichnen sich durch ihre besonders ausgeprägte antidepressive Wirkung aus und wirken bei Kachexie gewichtssteigernd.
Beispiel 1
23,5 g Acetylchlorid werden bei Raumtemperatur unter gutem Rühren und unter Ausschluss von Feuchtigkeit innert etwa 2 Stunden zu einer Mischung von 34,8 g 1,2-Diisopropyl-hydrazin und 400 ml absolutem Pyridin getropft, worauf noch 2 Stunden bei etwa 600 gerührt wird. Der Hauptteil des Pyridins wird dann im Vakuum abdestilliert, der Rückstand in 500 ml Chloroform aufgenommen und mit einer konzentrierten wässerigen Lösung von 22 g wasserfreiem Kaliumcarbonat portionenweise durchgeschüttelt, die abgetrennte Chloroformschicht über Natriumsulfat getrocknet, im Vakuum eingeengt und der ölige Rückstand durch mehrmalige Fraktionierung im Vakuum gereinigt. Man erhält so 1-Acetyl- 1,2-diisopropyl-hydrazin vom Siedepunkt 88-896l 12 mm.
Eine Lösung dieser Verbindung in absolutem Äther wird mit äthanolischer Chlorwasserstoffsäure in geringem Überschuss versetzt. Das l-Acetyl-1, 2 diisopropyl-hydrazin-hydrochlorid fällt aus und wird aus Alkohol/Äther umkristallisiert ; Schmelzpunkt 153 bis 155 (Prismen).
Beispiel 2
Eine Lösung von 24,1 g Pivaloylchlorid in 100ml absolutem Benzol wird bei Raumtemperatur unter Rühren zu einer Lösung von 25,5 g 1,2-Diisopropylhydrazin in einer Mischung von 150 ml absolutem Benzol und 150 ml absolutem Pyridin getropft, worauf noch 21/2 Stunden bei 50 weitergerührt wird.
Die erhaltene Suspension wird im Wasserstrahlvakuum bei 45 eingeengt, der Rückstand in Chloroform aufgenommen, die Chloroformlösung mit einer konzentrierten wässerigen Lösung von 15 g Kaliumcarbonat vorsichtig durchgeschüttelt, worauf die Chloroformschicht mit gesättigter Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und im Vakuum eingedampft wird. Der ölige Rückstand wird durch mehrmalige Fraktionierung gereinigt, wobei l-Pivaloyl-1, 2-diisopropyl-hydrazin als farb- loses Ö1 vom Siedepunkt 89-92 113 mm erhalten wird.
Beispiel 3
Eine Lösung von 27,5 g Palmitoylchlorid in 100 ml absolutem Benzol wird unter Rühren zu einer Lösung von 32 g 1,2-Diisopropyl-hydrazin in 150 ml absolutem Benzol getropft, wobei 1,2-Diisopropyl-hydrazin-hydrochlorid ausfällt. Die Mischung wird 1 Stunde unter Rückfluss gekocht, abgekühlt, filtriert und das Filtrat im Vakuum eingedampft. Der feste Rückstand wird aus Methanol umkristallisiert, wobei man 1-Palmitoyl-1, 2-diisopropyl-hydrazin vom Schmelzpunkt 399 erhält.
Beispiel 4
Zu einer Lösung von 70 g 1,2-Diisopropylhydrazin in 1000 ml absolutem Benzol wird unter Rühren bei Raumtemperatur eine Lösung von 33,9 g Chloracetylchlorid in 200 ml absolutem Benzol getropft, wobei sich alsbald 1,2-Diisopropyl-hydrazinhydrochlorid auszuscheiden beginnt. Man rührt noch einige Stunden weiter, lässt die Reaktionsmischung mehrere Stunden stehen, nutscht ab, wäscht das Filtrat mit Wasser, dampft die Benzollösung nach dem Trocknen über Natriumsulfat am Wasserstrahlvakuum zur Trockne ein und destilliert den Rück- stand im Hochvakuum. Dabei erhält man l-Chlor- acetyl-1, 2-diisopropyl-hydrazin als farbloses viskoses Öl vom Siedepunkt 67"1'0, 03 mm.
Beispiel 5
Eine Lösung von 59 g 1,2-Diisopropyl-hydrazin in 1300 ml absolutem Benzol wird bei Raumtemperatur unter Rühren innerhalb 2 Stunden zu einer Lösung von 35 g Acetylmilchsäurechlorid in 200 ml absolutem Benzol getropft. Man rührt 1-2 Stunden weiter, lässt über Nacht stehen und saugt das ausgeschiedene 1,2-Diisopropyl-hydrazin-hydrochlorid ab, wäscht die Benzollösung mit Wasser, trocknet über Natriumsulfat und dampft das Benzol im Vakuum ab. Der zuerst ölige, alsbald kristallin erstarrende Rückstand wird aus hochsiedendem Petroläther umkristallisiert, wobei l-(a-Acetoxy-propionyl)-1, 2-diisopropyl-hydrazin vom Schmelzpunkt 61-64J erhal- ten wird.
Eine Lösung von 10 g 1- (a-Acetoxy-propionyl)1,2-diisopropyl-hydrazin in 50 ml absolutem Methanol wird mit 1 ml 2n absoluter methanolischer Natriummethylat-Lösung versetzt und die Reaktionsmischung über Nacht stehengelassen. Man engt dann im Vakuum ein, nimmt den öligen Rückstand in Äther auf, wäscht mit gesättigter Kochsalzlösung, trocknet die Ätherlösung, dampft den Äther ab und kristallisiert den alsbald erstarrenden öligen Rück- stand aus Petroläther um, wobei man 1- ( (-tHydroxy- propionyl)-1, 2-diisopropyl-hydrazin vom Schmelzpunkt 65-67 erhält.
Beispiel 6
Eine Lösung von 10,8 g Methoxyacetylchlorid in 50 ml absolutem Benzol wird unter Rühren bei Raumtemperatur innert 21/2 Stunden zu einer Lösung von 25,5 g 1,2-Diisopropyl-hydrazin in 350 ml absolutem Benzol getropft, wobei 1,2-Diisopropylhydrazin-hydrochlorid ausfällt. Man rührt während 4 Stunden bei 40 weiter, kühlt ab, filtriert, wäscht das Filtrat mit gesättigter Kochsalzlösung und mit ge sättigter Natriumbicarbonatlösung, trocknet es über Natriumsulfat und destilliert das Lösungsmittel ab.
Der Rückstand wird im Vakuum fraktioniert, wobei man 1-Methoxyacetyl-1, 2-diisopropyl-hydrazin als farbloses Öl vom Siedepunkt 109-110 112 mm erhält ; n24 = 1,4525. Es erstarrt bei Raumtemperatur zu einer tiefschmelzenden Kristallmasse.
Beispiel 7
16,2 g Methoxyessigsäureanhydrid und 25,5 g 1,2-Diisopropyl-hydrazin werden miteinander vermischt, wobei durch Kühlung die Temperatur unter 50 gehalten wird. Man lässt 20 Stunden bei Raumtemperatur stehen, wobei das methoxyessigsaure Salz des 1, 2-Diisopropyl-hydrazins ausfällt. Der entstandene Brei wird zwischen Ather und gesättigter Kochsalzlösung verteilt und die Ätherschicht mit gesättig- ter Kochsalzlosung und mit gesättigter Natriumbicar bonatlösung gewaschen. Man trocknet die ätherische Lösung mit Natriumsulfat und destilliert das Lösungsmittel ab.
Der Rückstand wird im Vakuum destilliert, wobei man 1-Methoxyacetyl-1, 2-diisopropyl-hydrazin als farbloses Öl vom Siedepunkt 109 bis 110, 1112 mm erhält ; n2r) = 1,4522. Es erstarrt bei Raumtemperatur zu einer tiefschmelzenden Kristallmasse.
Beispiel 8
Eine Lösung von 12,5 g Methylmercaptoacetylchlorid in 100 ml absolutem Benzol wird unter Rühren zu einer Lösung von 32 g 1,2-Diisopropylhydrazin in 150 ml absolutem Benzol getropft, wobei 1,2-Diisopropyl-hydrazin-hydrochlorid ausfällt. Die Mischung wird 1 Stunde unter Rückfluss gekocht, abgekühlt, filtriert und das Filtrat im Vakuum eingedampft. Der Rückstand wird im Vakuum destilliert, wobei man 1-Methylmercaptoacetyl-1, 2-diisopropylhydrazin vom Siedepunkt 126-128 112 mm erhält ; nid = 1,4855.
Beispiel 9
Eine Lösung von 10,5 g Cyclopropancarbonsäurechlorid in 100 ml absolutem Benzol wird unter Rühren zu einer Lösung von 32 g 1,2-Diisopropylhydrazin in 150 ml absolutem Benzol getropft, wobei 1,2-Diisopropyl-hydrazin-hydrochlorid ausfällt. Die Mischung wird 1 Stunde unter Rückfluss gekocht, abgekühlt, filtriert und das Filtrat im Vakuum eingedampft. Der Rückstand wird im Vakuum destilliert, wobei man 1-Cyclopropancarbonyl-1, 2-diisopropyl-hydrazin vom Siedepunkt 104-105 113 mm erhält ; nD = 1,4640.
Beispiel 10
Eine Lösung von 14,6 g Cyclohexancarbonsäure- chlorid in 50 ml absolutem Benzol wird unter Rühren bei Raumtemperatur innert 21/2 Stunden zu einer Lösung von 25,5 g 1,2-Diisopropyl-hydrazin in 450 ml absolutem Benzol getropft, wobei 1,2-Diisopropyl-hydrazin-hydrochlorid ausfällt. Man rührt 4 Stunden bei 40 weiter, kühlt ab, filtriert, wäscht das Filtrat mit Wasser und mit gesättigter Natrium bicarbonatlösung, trocknet über Natriumsulfat und engt im Vakuum ein. Der Rückstand wird im Vakuum destilliert, wobei man 1-Cyclohexancarbonyl- 1,2-diisopropyl-hydrazin als farbloses Öl vom Siedepunkt 651, 10, 06 mm erhält.
Es erstarrt bei Raumtemperatur zu einer tiefschmelzenden Kristallmasse.
Beispiel 11
Eine Lösung von 39 g 2-Furoylchlorid in 160 ml absolutem Benzol wird unter Rühren bei Raumtemperatur innert 21/2 Stunden zu einer Mischung von 34,8 g 1,2-Diisopropyl-hydrazin und 400 ml absolutem Pyridin getropft, worauf noch während 3 Stunden weitergerührt wird. Die Aufarbeitung erfolgt nach der in Beispiel 1 angegebenen Weise. Man erhält 1- (2-Furoyl)-1,2-diisopropyl-hydrazin vom Siedepunkt 86-87 /0, 1 mm, Schmelzpunkt 62-66,5 (aus Wasser umkristallisiert).
Beispiel 12
Eine Lösung von 29,3 g 2-Thenoylchlorid in 50 ml absolutem Benzol wird bei Raumtemperatur unter Rühren innert 2 Stunden zu einer Mischung von 200 ml absolutem Benzol, 200 ml absolutem Pyridin und 23,2 g 1,2-Diisopropyl-hydrazin getropft und die erhaltene Suspension 3 Stunden bei 60 weitergerührt, worauf das Benzol und das Pyridin im Wasserstrahlvakuum weitgehend abdestilliert werden. Der Rückstand wird in Chloroform aufge nommen und mit einer Lösung von 20 g Kaliumcarbonat in wenig Wasser geschüttelt.
Die Chloroformschicht wird abgetrennt, über Natriumsulfat getrocknet, eingeengt, der Rückstand mit Wasser verrieben, abgenutscht und aus 50l /o igem Alkohol umkristallisiert, wobei 1- (2-Thenoyl)-1, 2-diisopropyl- hydrazin vom Schmelzpunkt 106-109 erhalten wird.
Das durch Behandlung mit äthanolischer Chlorwasserstoffsäure gebildete 1- (2-Thenoyl)-1, 2-diisopropyl-hydrazin-hydrochlorid kristallisiert nach der Umkristallisation aus AlkohollSither in Form von feinen Nadeln, Schmelzpunkt 132-134 .
Beispiel 13
23,8 g Thiophencarbonsäureanhydrid- (2) werden in 50 ml absolutem Benzol durch Erwärmen gelöst.
Zu dieser Lösung gibt man langsam 25,5 g 1,2-Diisopropyl-hydrazin, wobei dafür gesorgt wird, dass die Temperatur nie 60 übersteigt. Man hält das Reaktionsgemisch noch etwa 1 Stunde zwischen 50 und 60 und lässt dann über Nacht abkühlen. Der Kristallbrei wird zwischen Äther und Wasser verteilt, die Atherschicht mit Wasser und gesättigter Natrium bicarbonatlösung gewaschen, mit Natriumsulfat getrocknet und im Vakuum zur Trockne eingeengt. Der feste Rückstand wird aus Petroläther umkristallisiert, wobei man 1-(2-Thenoyl)-1, 2-diisopropyl-hydrazin vom Schmelzpunkt 106-109 erhält.
Beispiel 14
Eine Lösung von 16,1 g 4-Methyl-thiazolcarbon säure-chlorid- (5) in 100 ml absolutem Benzol wird unter Rühren bei Raumtemperatur innert 21/2 Stunden zu einer Lösung von 25,5 g 1,2-Diisopropylhydrazin in 400 ml absolutem Benzol getropft, wobei 1,2-Diisopropyl-hydrazin-hydrochlorid ausfällt. Man rührt 4 Stunden bei 40 weiter, kühlt ab, filtriert, wäscht das Filtrat mit Wasser und mit gesättigter Na triumbicarbonatlösung, trocknet über Natriumsulfat und engt im Vakuum zur Trockne ein. Der feste Rückstand wird aus Petroläther umkristallisiert, wobei man 1- [4-Methyl-thiazol-carbonyl- (5)]-1, 2-diisopropyl-hydrazin vom Schmelzpunkt 52-53 erhält.
Beispiel 15
Eine Lösung von 11,3 g 5-Brom-thiophencarbon säure-chlorid- (2) in 25 ml absolutem Benzol wird unter Rühren bei Raumtemperatur innert 2 Stunden zu einer Lösung von 12,8 g 1, 2-Diisopropyl-hydrazin in 200 ml absolutem Benzol getropft, wobei 1,2-Diisopropyl-hydrazin-hydrochlorid ausfällt. Man rührt während 4 Stunden bei 40 weiter, kühlt ab, filtriert, wäscht das Filtrat mit Wasser und mit gesättigter Natriumbicarbonatlösung, trocknet über Natriumsulfat und engt im Vakuum zur Trockne ein. Der feste Rückstand wird aus SitherlPetroläther umkristallisier, wobei man 1- (5-Brom-2-thenoyl)-1, 2-diisopropyl-hydrazin vom Schmelzpunkt 118-122 erhält.

A solution of this compound in absolute ether is treated with a slight excess of ethanolic hydrochloric acid. The l-acetyl-1,2 diisopropylhydrazine hydrochloride precipitates and is recrystallized from alcohol / ether; Melting point 153 to 155 (prisms).
Example 2
A solution of 24.1 g of pivaloyl chloride in 100 ml of absolute benzene is added dropwise at room temperature with stirring to a solution of 25.5 g of 1,2-diisopropylhydrazine in a mixture of 150 ml of absolute benzene and 150 ml of absolute pyridine, whereupon 21/2 Stirring is continued at 50 hours.

Example 3
A solution of 27.5 g of palmitoyl chloride in 100 ml of absolute benzene is added dropwise with stirring to a solution of 32 g of 1,2-diisopropylhydrazine in 150 ml of absolute benzene, 1,2-diisopropylhydrazine hydrochloride precipitating out. The mixture is refluxed for 1 hour, cooled and filtered, and the filtrate is evaporated in vacuo. The solid residue is recrystallized from methanol, 1-palmitoyl-1,2-diisopropylhydrazine having a melting point of 399 being obtained.
Example 4
To a solution of 70 g of 1,2-diisopropylhydrazine in 1000 ml of absolute benzene, a solution of 33.9 g of chloroacetyl chloride in 200 ml of absolute benzene is added dropwise with stirring at room temperature, and 1,2-diisopropylhydrazine hydrochloride soon begins to separate out. Stirring is continued for a few hours, the reaction mixture is left to stand for several hours, suction filtered, the filtrate is washed with water, the benzene solution is evaporated to dryness after drying over sodium sulfate in a water jet vacuum and the residue is distilled in a high vacuum. Example 5
A solution of 59 g of 1,2-diisopropylhydrazine in 1300 ml of absolute benzene is added dropwise at room temperature with stirring to a solution of 35 g of acetyl lactic acid chloride in 200 ml of absolute benzene over the course of 2 hours. The mixture is stirred for a further 1-2 hours, left to stand overnight and the 1,2-diisopropylhydrazine hydrochloride which has separated out is filtered off with suction, the benzene solution is washed with water, dried over sodium sulfate and the benzene is evaporated off in vacuo. The initially oily residue, which soon solidifies in crystalline form, is recrystallized from high-boiling petroleum ether, l- (α-acetoxypropionyl) -1,2-diisopropylhydrazine with a melting point of 61-64J being obtained.
A solution of 10 g of 1- (a-acetoxypropionyl) 1,2-diisopropylhydrazine in 50 ml of absolute methanol is mixed with 1 ml of 2N absolute methanolic sodium methylate solution and the reaction mixture is left to stand overnight. It is then concentrated in vacuo, the oily residue is taken up in ether, washed with saturated sodium chloride solution, the ether solution is dried, the ether is evaporated and the oily residue, which soon solidifies, is recrystallized from petroleum ether, using 1- ((-thydroxy- propionyl) -1, 2-diisopropylhydrazine with a melting point of 65-67.
Example 6
A solution of 10.8 g of methoxyacetyl chloride in 50 ml of absolute benzene is added dropwise with stirring at room temperature within 21/2 hours to a solution of 25.5 g of 1,2-diisopropylhydrazine in 350 ml of absolute benzene, 1.2- Diisopropylhydrazine hydrochloride precipitates. The mixture is stirred for 4 hours at 40, cooled, filtered, the filtrate is washed with saturated sodium chloride solution and with saturated sodium bicarbonate solution, dried over sodium sulfate and the solvent is distilled off.
The residue is fractionated in vacuo, 1-methoxyacetyl-1,2-diisopropylhydrazine being obtained as a colorless oil with a boiling point of 109-110 112 mm; n24 = 1.4525. It solidifies at room temperature to form a deep-melting crystal mass.
Example 7
16.2 g of methoxyacetic anhydride and 25.5 g of 1,2-diisopropylhydrazine are mixed with one another, the temperature being kept below 50 by cooling. The mixture is left to stand for 20 hours at room temperature, the methoxyacetic acid salt of 1,2-diisopropylhydrazine precipitating out. The resulting paste is distributed between ether and saturated saline solution and the ether layer is washed with saturated saline solution and with saturated sodium bicarbonate solution. The ethereal solution is dried with sodium sulfate and the solvent is distilled off.
The residue is distilled in vacuo, 1-methoxyacetyl-1,2-diisopropylhydrazine being obtained as a colorless oil with a boiling point of 109 to 110, 1112 mm; n2r) = 1.4522. It solidifies at room temperature to form a deep-melting crystal mass.
Example 8
A solution of 12.5 g of methyl mercaptoacetyl chloride in 100 ml of absolute benzene is added dropwise with stirring to a solution of 32 g of 1,2-diisopropylhydrazine in 150 ml of absolute benzene, 1,2-diisopropylhydrazine hydrochloride precipitating. The mixture is refluxed for 1 hour, cooled and filtered, and the filtrate is evaporated in vacuo. The residue is distilled in vacuo, 1-methylmercaptoacetyl-1,2-diisopropylhydrazine having a boiling point of 126-128 112 mm; nid = 1.4855.
Example 9
A solution of 10.5 g of cyclopropanecarboxylic acid chloride in 100 ml of absolute benzene is added dropwise with stirring to a solution of 32 g of 1,2-diisopropylhydrazine in 150 ml of absolute benzene, 1,2-diisopropylhydrazine hydrochloride precipitating out. The mixture is refluxed for 1 hour, cooled and filtered, and the filtrate is evaporated in vacuo. The residue is distilled in vacuo, giving 1-cyclopropanecarbonyl-1,2-diisopropylhydrazine with a boiling point of 104-105 113 mm; nD = 1.4640.
Example 10
A solution of 14.6 g of cyclohexanecarboxylic acid chloride in 50 ml of absolute benzene is added dropwise with stirring at room temperature within 21/2 hours to a solution of 25.5 g of 1,2-diisopropylhydrazine in 450 ml of absolute benzene, where 1, 2-Diisopropylhydrazine hydrochloride precipitates. The mixture is stirred for a further 4 hours at 40, cooled, filtered, the filtrate is washed with water and with saturated sodium bicarbonate solution, dried over sodium sulfate and concentrated in vacuo. The residue is distilled in vacuo, 1-cyclohexanecarbonyl-1,2-diisopropylhydrazine being obtained as a colorless oil with a boiling point of 651.10.06 mm.
It solidifies at room temperature to form a deep-melting crystal mass.
Example 11
A solution of 39 g of 2-furoyl chloride in 160 ml of absolute benzene is added dropwise with stirring at room temperature within 21/2 hours to a mixture of 34.8 g of 1,2-diisopropylhydrazine and 400 ml of absolute pyridine, followed by a further 3 hours is stirred further. Work-up is carried out in the manner indicated in Example 1. 1- (2-Furoyl) -1,2-diisopropylhydrazine with a boiling point of 86-87 / 0.1 mm, melting point 62-66.5 (recrystallized from water) is obtained.
Example 12
A solution of 29.3 g of 2-thenoyl chloride in 50 ml of absolute benzene is added dropwise to a mixture of 200 ml of absolute benzene, 200 ml of absolute pyridine and 23.2 g of 1,2-diisopropylhydrazine at room temperature with stirring within 2 hours the suspension obtained is stirred for a further 3 hours at 60, after which the benzene and pyridine are largely distilled off in a water jet vacuum. The residue is taken up in chloroform and shaken with a solution of 20 g of potassium carbonate in a little water.
The chloroform layer is separated off, dried over sodium sulfate, concentrated, the residue triturated with water, suction filtered and recrystallized from 50 l / o alcohol, 1- (2-thenoyl) -1, 2-diisopropylhydrazine of melting point 106-109 being obtained .
The 1- (2-thenoyl) -1,2-diisopropylhydrazine hydrochloride formed by treatment with ethanolic hydrochloric acid crystallizes after recrystallization from AlcoholSither in the form of fine needles, melting point 132-134.
Example 13
23.8 g of thiophenecarboxylic anhydride (2) are dissolved in 50 ml of absolute benzene by heating.
25.5 g of 1,2-diisopropylhydrazine are slowly added to this solution, taking care that the temperature never exceeds 60. The reaction mixture is kept between 50 and 60 for about 1 hour and then left to cool overnight. The crystal pulp is distributed between ether and water, the ether layer is washed with water and saturated sodium bicarbonate solution, dried with sodium sulfate and concentrated to dryness in vacuo. A solution of 16.1 g of 4-methyl-thiazolecarbonic acid chloride (5) in 100 ml of absolute benzene is converted into a solution of 25.5 g of 1,2-diisopropylhydrazine in 400 ml at room temperature within 21/2 hours while stirring dropped into absolute benzene, whereby 1,2-diisopropylhydrazine hydrochloride precipitates. The mixture is stirred for a further 4 hours at 40, cooled, filtered, the filtrate is washed with water and with saturated sodium bicarbonate solution, dried over sodium sulfate and concentrated to dryness in vacuo. The solid residue is recrystallized from petroleum ether, 1- [4-methyl-thiazol-carbonyl- (5)] -1, 2-diisopropylhydrazine having a melting point of 52-53.
Example 15
A solution of 11.3 g of 5-bromo-thiophene carbonic acid chloride (2) in 25 ml of absolute benzene is stirred at room temperature within 2 hours to a solution of 12.8 g of 1,2-diisopropylhydrazine in 200 ml dropped into absolute benzene, whereby 1,2-diisopropylhydrazine hydrochloride precipitates. The mixture is stirred for 4 hours at 40, cooled, filtered, the filtrate is washed with water and with saturated sodium bicarbonate solution, dried over sodium sulfate and evaporated to dryness in vacuo. The solid residue is recrystallized from Sitherl petroleum ether, 1- (5-bromo-2-thenoyl) -1, 2-diisopropylhydrazine having a melting point of 118-122.
