Description
Verfahren zur Herstellung von neuen Glykolen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Glykolen der Formel

worin Rt einen Alkyl-, Alkenyl- oder Cycloalkylrest oder einen gegebenenfalls durch Halogenatome oder niedermolekulare Alkyl- oder Alkoxygruppen substituierten Arylrest, R2 und R3 niedermolekulare Alkylreste, wobei R2 auch mit einem Alkylrest Rj verbunden sein kann, und R4 einen ungesättigten aliphatischen Kohlenwasserstoffrest mit 2-3 Kohlenstoffatomen bedeuten. Wie nun gefunden wurde, besitzen diese Verbindungen sedative, hypnotische bis narkotische Wirksamkeit. Sie wirken auch antikonvulsiv und sind insbesondere als tranquilizers therapeutisch verwendbar.
Ferner potenzieren sie die Wirkung von andern Narkotica und kommen auch als Zwischenprodukte für die Herstellung weiterer pharmakologisch wertvoller Stoffe in Betracht.
Das erfindungsgemässe Verfahren zur Herstellung der oben definierten Verbindungen ist nun dadurch gekennzeichnet, dass man ein a-Hydroxy-keton der Formel

mit einer metallorganischen Verbindung aus Magnesium oder Zink und einem Halogenid der Formel Hal-CH2-R4 III worin Hal Chlor, Brom oder Jod bedeutet, umsetzt und gewünschtenfalls, wenn der Rest R4 des Reaktionsproduktes eine Dreifachbindung enthält, diese zu einer Doppelbindung hydriert. Mit allgemein guten Ausbeuten kann gerechnet werden, wenn man erfindungsgemäss ein Mol eines a-Hydroxy-ketons der Formel II mit ungefähr zwei Mol einer metallorganischen Verbindung aus Magnesium und einem Halogenid der Formel III in einem organischen Lösungsmittel, welches ein oder mehrere Sauerstoffatome zwischen Kohlenwasserstoffresten gebunden enthält, z.
B. ein Äther oder ätherartiges Lösungsmittel, wie Diäthyläther oder Tetrahydrofuran, oder ein Acetal, wie Dimethylformal, umsetzt.
Zu den als Ausgangsstoffe zur Anwendung gelangenden a-Hydroxy-ketonen der Formel II, worin R2 und R3 Methylgruppen bedeuten, gelangt man beispielsweise durch Kondensation von Methylketonen der Formel R-CO-CH3 mit Alkalimetallacetyliden und anschliessender Wasseranlagerung, z. B. in Gegenwart von Quecksilberoxyd und Schwefelsäure, an die so erhaltenen Athinyl-methyl- carbinole. Ferner sei die von Billimoria et al. (J.
Chem. Soc. 1951, 3067) beschriebene Umsetzung von a-Hydroxy-carbonsäuren mit Methyllithium erwähnt; a-Hydroxy-ketone der allgemeinen Formel II sind ferner aus Ketonen der allgemeinen Formel Rt-CO-R2 erhältlich. Diese werden zunächst in die entsprechenden Cyanhydrine übergeführt, deren Hydroxylgruppe zur Verschliessung beispielsweise acyliert oder mit Dihydropyran oder einem Vinyläther umgesetzt wird.
Die so erhaltenen O-Acylderivate, O-Tetrahydropyranylderivate oder O- (a-Alkoxy-äthyl)- derivate von Nitrilen verzweigter a-Hydroxy-alkancarbonsäuren, von a-Hydroxy-a-cyclo alkyl-aceto- nitrilen oder a-Hydroxy-a.-aryl-acetonitrilen lassen sich mit Alkylmagnesiumhalogeniden der Formel R3-Mg-Hal zu Iminen umsetzen, aus welchen durch Hydrolyse die a-Hydroxy-ketone der allgemeinen Formel II erhältlich sind.

a-Ketole der allgemeinen Formel II, worin R1und R2 zusammen einen Alkylenrest und R3 Methyl bedeuten, erhält man beispielsweise durch Kondensation von cycloaliphatischen Ketonen mit Alkalimetallacetyliden und Wasseranlagerung an die so erhaltenen Äthinylcycloalkanole, oder durch die oben erwähnte Umsetzung von a-Hydroxy-carbonsäuren mit Methyllithium; oder, falls R3 einen Alkylrest mit 2-4 Kohlenstoffatomen verkörpert, durch Umsetzung von Cyanhydrinen geeigneter cycloaliphatischer Ketone (nach Verschliessung der Hydroxylgruppe wie oben beschrieben) mit niedermolekularen Alkylmagnesiumhalogeniden zu Iminen, aus welchen durch Hydrolyse die a-Hydroxy-ketone der allgemeinen Formel II erhältlich sind.
Als Beispiele solcher Verbindungen seien das l-Acetyl-, l-Propionyl-, l-Butyryl- und l-Iso butyryl-cyclo-pentanol,
1 -Acetyl-methylcyclopentanol,
1 -Acetyl-dimethyl-cyclopentanol, l-Acetyl-, l-Propionyl-, l-Butyryl-, l-Isobutyryl-, l-Valeryl- und l-Isovaleryl-cyclohexanol, 1 -Acetyl-2-methyl-cyclohexanol,
1 -Acetyl-4-methylcyclohexanol, die 1 -Acetyl-dimethylcyclohexanole, l-Acetyl- und l-Propionyl-cycloheptanol sowie 1 -Acetyl-cyclooctanol genannt.
Als Beispiele ungesättigter aliphatischer Halogenide der Formel III seien schliesslich Allylchorid, Allylbromid, Allyljodid, Crotylchlorid, Crotylbromid, 4-Brom-buten-(l), Methallylchlorid, Methallylbromid, Propargylbromid und 4-Brom-butin-(l) genannt.
In den nachfolgenden Beispielen bedeuten Teile Gewichtsteile. Diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Zu einer nach Organic Synthese 36, 61 (1956) bereiteten Allylmagnesiumbromid-Lösung (aus 24,3 Teilen Mg, 1 Kriställchen Jod und 60,5 Teilen Allylbromid, unter Verwendung von total 350 Volumteilen abs. Äther) werden innerhalb anderthalb Stunden 28,8 Teile 3,5 - Dimethyl -3 - hydroxyhexanon - (2), Kp6: 650, gelöst in 50 Volumteilen abs. Äther, bei einer Reaktionstemperatur unter 201 zugetropft.
Nach 4stündigem Rühren bei Raumtemperatur werden zur Reaktionsmischung unter Eiskühlung zunächst 100 Teile Wasser zugetropft, worauf sie durch vorsichtige Zugabe von 50 0/o iger Schwefelsäure kongosauer gestellt wird. Nach weiterem einstündigem Rühren bei Raumtemperatur werden nochmals 100 Teile Wasser zugegeben. Der Niederschlag wird abgenutscht und mit Wasser ausgewaschen, die organische Schicht des Filtrats abgetrennt und die wässrige Schicht mit 3mal 100 Volumteilen Äther ausgeschüttelt. Die vereinigte organische Lösung wird neutral gewaschen, über Natriumsulfat getrocknet, und nach dem Entfernen des Lösungsmittels wird der Rückstand durch wiederholte Destillation unter vermindertem Druck gereinigt. Das 4,5,7-Trimethyl- 4,5-dihydroxy-octen-(1) siedet unter 11 mm Druck bei 109-111".
In analoger Weise erhält man unter Verwendung der entsprechenden Ausgangsstoffe: 2-(1'-Hydroxy-cyclopentyl)-2-hydroxy-penten-(4) Kp11: 119,5-121,5", Smp. 35, 5-37O.
4-Methyl-5-p-chlorphenyl-4,5-dihydroxy-hexen-(1) Kp12: etwa 1800. Smp. 98-1000 (aus Äther
Pentan).
4,5-Dimethyl-4,5-dihydroxy-octen-( 1) Kp11: 111". Smp. 41-44) (aus Ather-Pentan) und 4,5,6,6-Tetramethyl-4,5 -dihydroxy-hepten-( 1)
Kp11: 111-112, 5 .
Beispiel 2
12,2 Teile amalgamiertes Magnesium und 200 Volumteile abs. Ather werden in einem trockenen Sulfierkolben vorgelegt und unter Rühren innerhalb einer halben Stunde 65 Teile Propargylbromid in 50 Volumteilen abs. Äther zugetropft, wobei der Äther gelinde unter Rückfluss siedet. Nach beendeter Zugabe wird das Reaktionsgemisch 14 Stunden bei Raumtemperatur gerührt. Dann werden innerhalb einer Stunde 32,5 Teile 3-Methyl-3-hydroxyhexanon-(2), Kp,,: 95O, gelöst in 50 Volumteilen abs.
Ather, bei einer Innentemperatur von etwa 30-35 zugegeben. Nach weiterem vierstündigem Rühren bei Raumtemperatur werden zur Reaktionsmischung unter Eiskühlung zunächst 50 Volumteile Wasser zugegeben, worauf sie mit 50 /aiger Schwefelsäure kongosauer gestellt wird.
Nach Aufarbeitung analog Beispiel 1 erhält man 4,5-Dimethyl-4,5-dihydroxy-octin-(1) vom Kp,: 104-106"; nD: 1,471.
Beispiel 3
Aus 49,0 Teilen Magnesium, 100,0 Teilen frisch destilliertem Methallylchlorid und insgesamt 700 Volumteilen abs. Äther wird das Grignard-Reagens analog Beispiel 1 hergestellt. Die weitere Umsetzung mit 57,6 g 3,5-Dimethyl-3-hydroxy-hexanon-(2) in 100 Volumteilen abs. Äther sowie die Hydrolyse erfolgt analog Beispiel 1, wobei man 2,4,5,7-Tetra- methyl-4,5-dihydroxy-octen-(l) vom Kr12: 119,5"; und: 1,464 erhält.
Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung von neuen Glykolen der Formel EMI3.1 worin Rj einen Alkyl-, Alkenyl- oder Cycloalkylrest oder einen gegebenenfalls durch Halogenatome oder niedermolekulare Alkyl- oder Alkoxygruppen substituierten Arylrest, R2 und R5 niedermolekulare Alkylreste, wobei R2 auch mit einem Alkylrest R, verbunden sein kann, und R, einen ungesättigten aliphatischen Kohlenwasserstoffrest mit 23 Kohlenstoffatomen bedeuten, dadurch gekennzeichnet, dass man ein a-Hydroxy-keton der Formel EMI3.2 mit einer metallorganischen Verbindung aus Magnesium oder Zink und einem Halogenid der Formel Hal-CH2-R4 III worin Hal Chlor, Brom oder Jod bedeutet, umsetzt.UNTERANSPRUCH Verfahren gemäss Patentanspruch, dadurch gekennzeichnet, dass man Verbindungen der Formel I herstellt, worin R4 eine Dreifachbindung enthält, und dass man diese nachträglich zu einer Doppelbindung hydriert.
