Description
Verfahren zur Herstellung eines Glykosides von 5-Fluor-uracil Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 2'-Desoxy-5 fluor-uridin, das heisst von 1- (ss - D - 2'-Desoxy-ribofuranosyl)-5-fluor- uracil, welches dadurch gekennzeichnet ist, dass man ein 2-Desoxy-ribos:
id und 5-Fluor-uracil zwecks Aus tausch des 2'-Desoxy-riboserestes einer enzymati sehen Einwirkung unterwirft. Das erhaltene Produkt kann gewünschtenfalls mit einer Base in ein Salz übergeführt werden.
Als 2-Desoxy-ribosid, das den 2'-Desoxy-ribose- rest liefern kann, verwendet man vorzugsweise Thy- midin. Als Enzymlieferant dient vorteilhaft Strepto- coccus faecalis.
Das erfindungsgemäss erhaltene Glykosid sowie Salze dieser Verbindung sind nützlich als antisep tische Mittel, z. B. gegen grampositive Bakterien, wie Staphylococcus aureus und gegen Pilze, wie Sco- pulariopsis brevicaulis. Sie sind auch verwendbar als Antimetaboliten im Nucleinsäureaufbau und verhin dern so das Wachstum der Zellen, z. B. beim Lacto- bazillus leichmannii.
Das als Ausgangsmaterial verwendete 5 Fluor- uracil kann wie folgt hergestellt werden: Eine Mischung von 200 g (2 Mol) trockenem Natriumsalz der Fluoressigsäure und 442 g (2,86 Mol) Diäthylsulfat werden 31/2 Stunden unter Rückfluss im Ölbad gekocht.
Nach fraktionierter Destillation der Reaktionsmischung erhält man 177,3 g rohen Äthylester der Fluoressigsäure vom Siedebereich 116 bis 120 . Nach nochmaliger Destillation durch eine Fraktionierkolonne erhält man den zwischen 114 und 118 siedenden Äthylester der Fluoressigsäure.
In einen mit Rührer, Tropftrichter und Rück flusskühler versehenen Kolben gibt man 880 ml ab soluten Diäthyläther und 47,6 g in 5 mm Stücke ge schnittenes Kalium. Unter Rühren werden 220 ml absolutes Äthanol zugetropft und bis zur vollständi- gen Auflösung des Kaliums am Rückfluss erhitzt.
In die eisgekühlte Lösung trägt man innerhalb von 21/2 Stunden unter Rühren und Kühlen 135 g (1,22 Mol) des Fluoressigsäureäthylesters und 96,4<B>g</B> (1,3 Mol) von frisch destilliertem Ameisensäureäthylester ein.
Wenn alles eingetragen ist, wird unter Kühlen noch eine Stunde gerührt und dann das Reaktionsgemisch über Nacht stehengelassen. Der gebildete kristalline Rückstand wird abgenutscht, mit Diäthyläther ge waschen und im Vakuum getrocknet. Man erhält das Kaliumsalz des Hydroxymethylen - fluoressigsäure- äthylesters.
Eine Mischung von 103,6 g frisch hergestelltem Kaliumsalz des Hydroxymethylen - fluoressigsäure- äthylesters, 83,4 g (0,3 Mol) des Sulfates von S- Methyl-isothioharnstoff und 32,5 g (0,6 Mol) Na- triummethanolat wird unter Rühren in 1500 ml ab solutem Methanol unter Rückfluss erhitzt.
Nach vor übergehender Lösung wird eine Fällung beobachtet. Nach zweistündigem Erhitzen unter Rückfluss wird das Reaktionsgemisch im Vakuum zur Trockne ein- gedampft und der Rückstand mit 280 ml Wasser be- handelst. Das entstandene Gemisch wird durch Kohle filtriert und das entfärbte Filtrat durch Zusatz von 48 ml 37o/oiger Chlorwasserstoffsäure schwach kongosauer gestellt.
Die gebildeten Kristalle werden abfiltriert, mk Wasser sulfatfrei gewaschen und bei 100 getrocknet. Man erhält,den rohen S-Methyläther von 2-Thio-5-fluor-uracil vom Schmelzbereich 202 bis 221 . Nach dem Lösen in 2035 ml siedendem Äthylacetat und Abkühlen auf - 20 schmilzt das Produkt bei 230-237 .
Nach weiterem Umkristalli- sieren aus Wasser (oder aus Äthylacetat) steigt der Schmelzpunkt auf 241-243 .
Eine Lösung von 10 g S-Methyläther von 2- Thio-5-fluor-uracil vom Schmelzpunkt 230-237 in 150 ml 37o/oiger wässeriger Chlorwasserstoffsäure wird unter Stickstoff vier Stunden unter Rückfluss erhitzt. Das Reaktionsgemisch wird im Vakuum ab gedampft und der bräunliche kristalline Rückstand aus Wasser umkristallisiert. Das Produkt wird durch Sublimation im Vakuum (0,1 mm, 190-200 Bad temperatur) weitergereinigt.
Man erhält 5-Fluor-uracil als farblose oder rötliche Kristalle vom Schmelzpunkt 282-283 (unter Zersetzung). <I>Beispiel 1</I> Zellen von Streptococcus faecalis (ATCC 8043) werden auf einem AOAC Folsäure-Nährboden ge züchtet [Lepper,
Official and Tentative Methods of the Association of Official Agricultural Chemists, Washington, D. C., 7. Auflage (1950), Seite 784], dem 2 mg/1 Thymin zugesetzt wurden; nach den An gaben von Prusoff, Proc. Soc. Exp. Biol. & Med. 85 (1954), Seite 564.
Nach 20stündigem Brüten bei 37 werden die Zellen durch Zentrifugieren gewonnen. Die erhaltenen Zellen werden dreimal mit 4 Volumen einer Kaliumphosphat Pufferlösung gewaschen (n/15 wässrige Monokaliumorthophosphatlösung, die durch Zugabe von 2n Kalilauge auf pH 8,0 gebracht wurde), und die nassen Zellen werden gewogen.
Die Zellen werden schliesslich in der obigen Kaliumphosphat- Pufferlösung suspendiert und in einem Glashomogeni- sator nach Potter verrieben.
900 mg nasse Zellen werden mit der obigen Kaliumphosphat-Pufferlösung zu 25 ml eines Enzym präparates zubereitet. 150 mg (1,15 Millimol) 5- Fluor-uracil und 1,0 g (4,12 Millimol) Thymidin wer den in 15 ml der obigen Kaliumphosphat-Pufferlösung gelöst. Die zwei Lösungen werden vereinigt und wäh rend 18 Stunden bei 37 bebrütet.
Nach dieser Zeit wird die Enzymwirkung durch Zusatz von 4 Volumen Aceton und 1 Volumen peroxydfreiem Diäthyläther gehemmt. Die ausgefallenen festen Teile werden ab filtriert; das Filtrat wird unter Stickstoff und vermin dertem Druck eingedampft, bis die flüchtigen organi schen Lösungsmittel im wesentlichen entfernt sind. Es verbleiben ungefähr 20 ml einer wässrigen Lösung, welche in 100 m_ 1 destilliertem Wasser aufgenommen wird.
10 Microliter dieser Lösung werden auf einem mit 0,2n Monokaliumorthophosphat (pH 7,8) ge- puffertem Papier der absteigenden Chromatographie unterworfen, wobei als Lösungsmittel eine Mischung von Alkohol/Wasserjn-Butyläther (80:13:7) ver wendet wird.
Eine unter ultraviolettem Licht ersicht liche Stelle vom Wert R, = 0,55 wird mit 0,1n Salz säure .ausgelaugt und auf Desoxyribose geprüft [nach Stumpf, J. Biol. Chem.169 (1947), Seite 367]. Diese Analyse zeigt die Anwesenheit von mindestens 85,5 mg (0,35 Millimol) 2'-Desoxy-5-fluor-uridin in dem proteinfreien Reaktionsgemisch.
Das erfindungsgemäss erhaltene 2'-Desoxy-5- fluor-uridin hat saure Eigenschaften und bildet Salze mit Basen. Durch Reaktion von 2'-Desoxy-5-fluor- uridin reit pharmazeutisch brauchbaren Basen, z. B. Alkali- und Erdalkalihydroxyde, Ammoniak, nicht- toxische organische Basen, z. B. Äthanolamin usw., bilden sich Salze.
2'-Desoxy-5-fluor-uridin und seine pharmazeu tisch brauchbaren Salze sind wertvoll als antibakte rielle Verbindungen, da sie z. B. gegen St. aureus, Sarcina lutea, B. subtilis, E. bacillus und B. simplex wirksam sind; sie sind auch wirksam gegen Pilze, z. B. gegen Scopulariopsis brevicaulis, C. albicans usw.
<I>Beispiel 2</I> Zellen von Streptococcus faecalis (ATCC 8043) werden gemäss Beispiel 1 gezüchtet und aufgearbeitet. 4,06 .g nasse Zellen werden mit der Kaliumphos- phat-Pufferlösung gemäss Beispiel 1 zu 105 ml eines Enzympräparates zubereitet. 200 mg (1,54 Millimol) 5-Fluor-uracil und 1,50 g (6,16 Millimol) Thymidin werden in 15 ml der obigen Kali mphosphat-Puffer- lösung gelöst.
Die zwei Lösungen werden vereinigt und während 18 Stunden bei 37 bebrütet. Nach die ser Zeit wird die Enzymwirkung durch Zusatz von 4 Volumen Aceton und 1 Volumen peroxydfreiem Diäthyläther gehemmt. Die ausgefallenen festen Teile werden abfiltriert; das Filtrat wird unter Stickstoff und vermindertem Druck eingedampft, bis die flüchtigen organischen Lösungsmittel im wesentlichen entfernt sind. Es verbleiben etwa 20 ml einer wässrigen Lösung, welche in 100 ml destilliertem Wasser auf genommen wird.
Die Lösung wird nochmals im Vakuum auf 5 ml eingeengt, und dann durch Zusatz von 20 ml 1n wässriger Natronlauge alkalisch gestellt, wobei eine Mischung erhalten wird, welche Natriumsalze der fol genden Verbindungen enthält:
N-Desoxyribosid von 5-Fluor-uracil, Thymin, Thymidin und 5-Fluor- uracil. Diese Mischung wird gereinigt durch Adsorp- tion an ein Ionenaustauscherharz und nachfolgender Elution mittels einer Pufferlösung von allmählich wachsendem Säuregrad.
Die Pyrimidinverbindungen der Mischung werden in folgender Reihenfolge eluiert: Thymidin, Thymin, 5-Fluor-uracil und 2'-Desoxy-5- fluor-uridin. Zur Reinigung wird die alkalische Mischung durch eine Säule von 2,2 X 27 cm Dowex 1-X4 (Markenprodukt) fliessen gelassen (Anionenaustauscherharz der Dow Chemical Co., Midland,
Michigan, bestehend aus einem querverbun denen Copolymeren von Styrol mit Divinylbenzol [4 Klo des letzteren], welches als funktionelle Gruppen quaternäre Ammoniumgruppen enthält), Maschen abstand 0,075-0,15 mm, welche zuerst in die Formiatform übergeführt wurde. Nach Adsorption wird mit Wasser bis zur neutralen Reaktion ge waschen.
Die Säule wird dann mit 280 ml einer wäss- rigen Ammoniumformiat-Pufferlösung (pH 9,8) ge waschen, welche eine Normalität von<B>0, 1,</B> bezogen auf Formiationen, aufweist. Die Eluate enthalten keine Substanz, die das ultraviolette Licht absorbiert.
Die Säule wird weiter mit einer wässrigen Ammonium- formiat-Pufferlösung (pH 7,4) eluiert, welche eine Normalität von 0,1, bezogen auf Formiationen, auf weist; die Durchflussgeschwindigkeit beträgt 46 ml in der Stunde.
Die Säule wird dann mit einer wässrigen Ammoniumformiat-Pufferlösung (pH 6,5) eluiert, welche eine Normalität von 0,1, bezogen auf Formiat- ionen, aufweist; die Durchflussgeschwindigkeit be- trägt hier 60 ml in der Stunde.
Die Fraktionen werden in Abständen von 30 Minuten gesammelt und einzeln auf Ultraviolettabsorption bei 260 und 280 mcc (pH 14) geprüft.

Die Prüfung der Ultraviolettabsorptionsspektren und die Papierchromatographie der einzelnen Frak tionen haben ergeben, dass die Fraktionen 6 bis 17 nur Thymin und Thymidin, die Fraktionen 34 bis 48 dagegen die entsprechenden Fluorverbindungen ent halten.
Die Fraktionen 34 bis 48 werden deshalb ver einigt und im Vakuum zur Trockne eingedampft. Der erhaltene Rückstand wird in 30 ml der oberen Schicht einer Zweiphasenmischung gelöst, weiche aus 60 Teilen Äthylacetat, 35 Teilen Wasser und 5 Teilen Ameisensäure besteht.
Man erstellt dann eine Ko lonne von 4,4 X 49 cm durch Anfeuchten von 285 g Cellulosepulver (aschefrei, Standard-Qualität) mit der oberen Schicht des erwähnten Äthylacetat/Wasser/ Ameisensäure-Gemisches und Einstampfen der nassen Cellulose in die Absorptionsröhre mit Hilfe eines Stabes.
Die 30 ml der Lösung werden dann durch die Säule fliessengelassen. Die Säule wird mit der oberen Schicht des obigen Äthylacetat/Wasser/Ämei- sensäure-Gemisches eluiert; die Durchflussgeschwin- digkeit beträgt 40 ml in der Stunde und die Frak tionen werden in Abständen von 30 Minuten gesam melt.
Sie werden einzeln auf Ultraviolettabsorption bei 260 mu und 280 mu (pH 14) geprüft.

Die Fraktionen 97 bis 104 werden vereinigt und im Vakuum bei 45 zur Trockne eingedampft. Man erhält 96 mg (25,311/o) 2'-Desoxy-5-fluor-uridin als farblose glasige Substanz. Seine Ultraviolettabsorption ist charakteristisch für ein Desoxyribosid und dem jenigen von Desoxyuridin ähnlich, aber mit einer Verlagerung des Maximums auf .grössere Wellenlän gen.
Wie man es für ein Desoxyribasid erwartet, und im Gegensatz zu dem freien Pyrimidin, gibt es nur eine geringe Verschiebung des Maximums in alkali schem Medium: bei pH 7,2, @,ma,@ = 268 :mu, s = 7570; bei pH 14, Amzx = 270 my, s = 6480.
Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung von 2'-Desoxy-5-fluor- uridin, dadurch gekennzeichnet, dass man ein 2-Des- oxy-ribosid und 5-Fluor-uracil zwecks Austausch des 2'-Desoxy-riboserestes einer enzymatischen Einwir kung unterwirft. UNTERANSPRÜCHE 1.Verfahren nach Patentanspruch, dadurch ge kennzeichnet, dass man das erhaltene 2'-Desoxy-5- fluor-uridin durch Behandeln mit Basen in Salze überführt. 2. Verfahren nach Patentanspruch, dadurch ge kennzeichnet, dass man als 2-Desoxy-ribosid Thy- midin verwendet. 3. Verfahren nach Patentanspruch, dadurch ge kennzeichnet, dass man als Enzymlieferant Strepto- cocous faecalis verwendet. 4.Verfahren nach den Unteransprüchen 2 und 3.
