  Verfahren zur Herstellung von neuen   Hydrazin-Derivaten   
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Hydrazin-Derivaten der Formel   I,

 worin A eine Alkylengruppe,   Ri    und   R2    niedere Alkylgruppen und   R3    eine Acylgruppe bedeuten, wobei die Gruppen A,   Rt    und   R    zusammen mindestens 5 Kohlenstoffatome enthalten.



   Erfindungsgemäss werden die neuen Hydrazin Derivate hergestellt, indem man aus Isopropylidenhydrazin-Derivaten der Formel   II,   
EMI1.2     
 die Isopropylidengruppe abspaltet.



   Diese Abspaltung kann in der Weise erfolgen, dass man die wässerig alkoholische Lösung des Ausgangsproduktes bei einer Temperatur von 0-40  mit Chlorwasserstoffgas sättigt. Zur Vervollständigung der Reaktion lässt man das Gemisch noch 4 Stunden bei Raumtemperatur stehen, engt darauf ein und macht unter starkem Kühlen alkalisch. Das Endprodukt wird anschliessend aus dem Gemisch mit einem organischen indifferenten Lösungsmittel, vorzugsweise   Diäthyläther,    extrahiert und durch Hochvakuumdestillation gereinigt.



   Die erfindungsgemäss hergestellten neuen Hydrazin-Derivate sind wertvolle Zwischenprodukte für die Herstellung von Medikamenten. Ihre Umsetzung mit reaktionsfähigen mono-oder disubstituierten Malonsäure-Derivaten führt zu   l-Amino-2,    4-dioxoazetidin-Derivaten der Formel   III,

 wobei A,   Ri,      R2    und   R3    die gleiche Bedeutung besitzen wie in Formel   I.    Diese Azetidine sind wertvolle Medikamente. Sie wirken antiphlogistisch und können in der Behandlung rheumatischer Erkrankungen angewendet werden. Einige Azetidin-Derivate übertreffen in ihrer Wirkung das Phenylbutazon um ein Mehrfaches.



   Besonders wichtig für die therapeutische Verwendung dieser Verbindungen ist, dass sie eine basische Gruppe enthalten. Aus diesem Grund können sie mit anorganischen oder organischen Säuren Salze bilden, die in Wasser löslich sind. Dies erleichtert einerseits die Resorption dieser Medikamente bei peroraler Medikation, ermöglicht aber anderseits auch die Herstellung hochkonzentrierter Lösungen, die in vielen Fällen für die parenterale Stosstherapie unentbehrlich sind.



   In den nachfolgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.



   Beispiel   1   
Eine Lösung von 4, 0 g   1-(2'-Diäthylamino-iso-      hexyl-l')-l-acetyl-2-isopropylidenhydrazin    in 28 cm3 86%igem wÏsserigem ¯thanol wird unter Kühlung bei 0  mit Chlorwasserstoffgas gesättigt und anschliessend 4 Stunden bei Zimmertemperatur stehengelassen. Darauf wird die L¯sung im Vakuum bei 30  stark eingeengt, der Rückstand in 20 cm3 Eiswasser aufgenommen und unter starker Kühlung mit   50 böiger wässeriger    Kalilauge alkalisch gemacht.



  Man schüttelt nun portionenweise mit total 100 cm3 Äther aus, trocknet die vereinigten Ätherextrakte über Magnesiumsulfat und entfernt den Äther im Vakuum. Der Rückstand wird im Hochvakuum destilliert, wobei das   1-(2'-Diäthylamino-isohexyl-l')-    1-acetylhydrazin zwischen   126-128     bei 1, 5 mm Hg als farbloses   01     berdestilliert. nD = 1, 4629.



   Das als Ausgangsmaterial verwendete 1-(2'-Di  äthylamino-isohexyl-1')-1-acetyl-2-isopropyliden-    hydrazin wird wie folgt hergestellt :
1,   0 g 1- (2'-Diäthylamino-isohexyl-l')-hydrazin    wird in 5 cm3 Aceton gelöst, wobei Erwärmung eintritt. Man lässt die Reaktionslösung 16 Stunden bei Zimmertemperatur stehen und erhitzt anschliessend noch 1/2 Sunde am Rückfluss zum Sieden. Darauf wird das Aceton abdestilliert, und der Rückstand im Vakuum fraktioniert, wobei das 1-(2'-Diäthyl  amino-isohexyl-1')-2-isopropylidenhydrazin    bei 110  bei 9 mm Hg als farblose Flüssigkeit  berdestilliert. n22 D = 1,4532.



   Zu einer L¯sung von 2,3 g 1-(2'-DiÏthylaminoisohexyl-1')-2-isopropylidenhydrazin in 10 cm3 abs.



  Chloroform lässt man bei 0  unter starkem Kühlen und Rühren langsam eine L¯sung von 0, 83 cm3 Acetylchlorid in 5   cm3    abs. Chloroform zutropfen.



  Anschliessend wird die Reaktionslösung noch 3 Stunden   bei +3    bis   +5  weitergerührt    und sodann 15 Stunden bei Zimmertemperatur stehengelassen. Darauf wird die Reaktionslösung auf 0  abgekühlt und zweimal mit je 25 cm3 eisgekühlter, wässeriger,   gesättig-    ter Sodalösung gewaschen. Die Chloroformlösung wird über Magnesiumsulfat getrocknet, das Chloroform im Vakuum entfernt und der Rückstand im Vakuum destilliert, wobei das   1-(2'-Diäthylamino-      isohexyl-l')-1-acetyl-2-isopropylidenhydrazin    zwischen   119-121     bei   1    mm Hg als leicht gelb ge  färbtes      01     bergeht. n22 D = 1, 4669.



   Beispiel 2
Eine L¯sung von 4, 9 g   l- (-Diäthylaminoäthyl)-    1-acetyl-2-isopropylidenhydrazin in 28 cm3   85 tigern Äthanol    wird unter Kühlung bei 0  mit Chlorwasserstoffgas gesättigt und anschliessend 4 Stunden bei 220 stehengelassen. Darauf wird das Athanol im Vakuum bei 30  entfernt, der ölige Rückstand in 20 cm3 Wasser aufgenommen und unter starkem Kühlen mit 50%iger wässeriger Kalilauge alkalisch gemacht. Man extrahiert mit total 100 cm3 Ather, trocknet die vereinigten   Ätherauszüge    über Magnesiumsulfat, dampft den Ather ab und destilliert den Rückstand im Vakuum, wobei das 1-(¯-DiÏthylaminoÏthyl)-1-acetylhydrazin bei   106     bei 1 mm Hg als farbloses Íl  bergeht. n22 D = 1, 4682.



   Das als Ausgangsmaterial verwendete   l- (jd-Di-      äthylaminoäthyl)-1-acetyl-2-isopropylidenhydrazin    wird wie folgt hergestellt :
3, 4 g   jS-Diäthylaminoäthylhydrazin    werden in 20 cm3 Aceton gelöst, wobei Erwärmung eintritt.



  Man lässt die Reaktionslösung 15 Minuten bei Zimmertemperatur stehen und erhitzt sie anschliessend noch 1/2 Stunde am Rückfluss zum Sieden. Darauf wird das Aceton abdestilliert und der Rückstand im Vakuum fraktioniert, wobei das 1-(¯-DiÏthyl  aminoäthyl)-2-isopropylidenhydrazin    zwischen 86 und 87  bei 9 mm Hg  berdestilliert. n   2 =    1, 4561.



   Zu einer L¯sung von 14, 0 g   1- (/3-Diäthylamino-      äthyl)-2-isopropylidenhydrazin    in 40 cm3 abs. Chloroform lässt man bei 0  unter starkem Kühlen und Rühren langsam eine L¯sung von 6, 5 cm3 Acetylchlorid in 15 cm3 abs. Chloroform zutropfen. Anschliessend wird die Reaktionslösung noch 3 Stunden bei einer Temperatur von + 2 bis   + 5     weitergerührt und sodann 20 Stunden bei Zimmertemperatur stehengelassen. Darauf wird die Reaktionslösung auf 0  abgekühlt und zweimal mit je 25 cm3 wässeriger gesättigter Sodalösung gewaschen.

   Die Chloroformlösung wird über Magnesiumsulfat getrocknet, das Chloroform im Vakuum entfernt, und der Rückstand im Vakuum destilliert, wobei das 1-   (ss-Diäthylaminoäthyl)-1-acetyl-2-isopropyliden-    hydrazin zwischen   103-105     bei 1 mm Hg als farblose Flüssigkeit  bergeht. nD = 1, 4721.



   Beispiel 3
16, 2 g   1-(Dimethylamino-isopropyl)-l-acetyl-2-    isopropylidenhydrazin werden in einem Gemisch aus 72 cm3 Äthanol und 8 cm3 Wasser gelöst. In diese L¯sung leitet man bei einer Temperatur von   30-35     Chlorwasserstoff bis zur Sättigung ein und lässt das Gemisch 3 Stunden bei dieser Temperatur stehen.



  Dann dampft man im Vakuum ein, löst den ¯ligen Rückstand in wenig Wasser und gibt unter gutem Kühlen   50/eige    Kalilauge zu, bis sich ein   01    abscheidet. Dieses wird in Äther aufgenommen, die ätherische L¯sung über Magnesiumsulfat getrocknet und dann eingedampft. Der Rückstand wird bei 11 mm Hg fraktioniert, wobei von   63-68     zuerst etwas   1- (Dimethylamino-isopropyl)-2-isopropyliden-    hydrazin, dann von   117-119       1- (Dimethylamino-iso-      propyl)-l-acetylhydrazin    destilliert.



   Diese Verbindung bildet mit Phenylisothiocyanat 1-   (Dimethylamino-isopropyl)-l-acetyl-4-phenyl-thio-    semicarbazid, das nach Kristallisation aus Essigester bei   159-160     unter Zersetzung schmilzt.



   Das als Ausgangsmaterial verwendete 1-(Di   methylamino-isopropyl)-1-acetyl-2-isopropyliden-    hydrazin wird wie folgt hergestellt :
75 g Dimethylamino-isopropylenhydrazin werden unter Kühlung zu 200 cm3 Aceton gegeben und über Nacht bei Zimmertemperatur stehengelassen. Darauf wird die Reaktionslösung noch   1    Stunde am R ckfluss zum Sieden erhitzt, das überschüssige Aceton im  Vakuum entfernt und der Rückstand im Vakuum destilliert, wobei das   1- (Dimethylamino-isopropyl)-2-    isopropylidenhydrazin zwischen 63-64  bei 10 mm Hg übergeht.



   Dipikrat, Prismen vom Smp.   144-148     (aus Methanol).



   Man lässt zur L¯sung von 33 g 1- (Dimethylamino-isopropyl)-2-isopropylidenhydrazin in 100 cm3 abs. Chloroform unter Rühren bei 0-10  17 cm3 Acetylchlorid zutropfen. Nach 16stündigem Rühren bei Zimmertemperatur werden zu der L¯sung unter gutem Kühlen 150 cm3   30 /aige    Pottaschelösung tropfenweise zugesetzt, dann die Chloroformschicht abgetrennt und die wässerige Phase noch zweimal mit Chloroform ausgeschüttelt. Nach Trocknen der vereinigten Chloroformextrakte über Magnesiumsulfat und Abdampfen des Chloroforms wird der Rückstand im Hochvakuum fraktioniert, wobei 1-(Dimethylamino-isopropyl)-1-acetyl-2-isopropylidenhydrazin bei 0, 2 mm Hg zwischen 76-82  als gelbes   01    übergeht.



   Beispiel 4
Eine Lösung von 7, 9 g   1- (Diathylamino-isopro-    pyl) -1 - acetyl - 2 - isopropylidenhydrazin in 36 cm3 Athanol und 4 cm3 Wasser wird bei einer Temperatur von 30-35  mit HCl-Gas gesättigt. Dann lässt man 4 Stunden bei Zimmertemperatur stehen, dampft die L¯sung im Vakuum ein, versetzt den Rückstand mit Eiswasser und gibt unter guter Kühlung   500/oigne    wässerige Kalilauge hinzu, bis sich ein   51    ausscheidet. Dieses wird in Ather aufgenommen, die   äthe-    rische L¯sung über Magnesiumsulfat getrocknet und eingedampft. Der Rückstand wird destilliert, wobei das   1- (Diäthylamino-isopropyl)-l-acetylhydrazin    bei einer Luftbadtemperatur von   120-140     bei 12 mm Hg als farbloses   01    übergeht.



   Zur Charakterisierung wird mit Phenylisothiocyanat das Phenylthiosemicarbazid hergestellt. Smp.



  140-141  aus   EssigesterlÄther.   



   Das als Ausgangsmaterial verwendete 1-(Diäthylamino-isopropyl)-1-acetyl-2-isopropylidenhydrazin wird wie folgt hergestellt :
8, 3 g   Diäthylamino-isopropylidenhydrazin    werden in 25 cm3 Aceton gelöst, wobei die Temperatur auf 30  steigt. Man lässt über Nacht stehen, erhitzt darauf die L¯sung 1 Stunde am Rückfluss zum Sieden und dampft das Aceton ab. Der Rückstand wird im Vakuum destilliert, wobei das   1-(Diäthylamino-isopro-    pyl)-2-isopropylidenhydrazin zwischen   77-80     bei 10 mm Hg übergeht.



   Das Dipikrat, umkristallisiert aus Methanol, schmilzt bei   124-128     (Zers.).



   Zu einer L¯sung von 9, 0 g l- (Diäthylamino-isopropyl)-2-isopropylidenhydrazin in 30 cm3 abs.



  Chloroform wird unter Kühlung und Rühren eine L¯sung von 4 cm3 Acetylchlorid in 10   cm3    abs.



  Chloroform zugegeben. Man rührt 3 Stunden bei   5 ,    lässt über Nacht bei Zimmertemperatur stehen, kühlt die Reaktionslösung auf   0     ab und schüttelt mit Pottaschelösung durch. Die Chloroformlösung wird über Magnesiumsulfat getrocknet, das Chloroform abgedampft und der Rückstand destilliert, wobei das    1- (Diäthylamino-isopropyl)-1-acetyl-2-isopropyliden-    hydrazin zwischen 93-95  bei 0, 2 mm Hg übergeht.



   Beispiel 5
In die L¯sung von 19, 4 g   l-(l'-Dimethylamino-    butyl-3')-1-acetyl-2-isopropylidenhydrazin in 100   cm       95  /o igem Äthanol    leitet man bei   15-20     Chlorwasserstoff bis zur Sättigung ein. Nach 4stündigem Stehen bei Zimmertemperatur dampft man die Lösung im Vakuum ein. Der Rückstand wird in wenig Wasser gelöst und unter guter Kühlung mit   piger    Kalilauge versetzt. Das sich ausscheidende 01 wird in Äther aufgenommen, die Atherlösung über Natriumsulfat getrocknet und eingedampft, und der Rückstand fraktioniert destilliert.



      1- (1'-Dimethylaminobutyl-3')-1-acetylhydrazin      destilliert bei 12 mm Hg von 114-115. n    = 1, 4685.



   Das daraus mit Phenylisothiocyanat hergestellte
1-   (1'-Dimethylaminobutyl-3')-1-acetyl-4-phenyl-thio-    semicarbazid schmilzt nach Kristallisation aus Chloro  form/Ather    bei 132  unter Zersetzung.



   Das als Ausgangsmaterial verwendete 1-(1'-Di  methylaminobutyl-3')-1-acetyl-2-isopropyliden-    hydrazin wird wie folgt hergestellt :
Man löst 38, 2 g   1-Dimethylaminobutyl-3-hy-    drazin unter Kühlung in   100    cm3 Aceton, lässt die L¯sung 18 Stunden bei Zimmertemperatur stehen, erhitzt sie darauf   1    Stunde zum Sieden am Rückfluss, dampft das Lösungsmittel ab und destilliert den   Rückstand. 1- (1'-Dimethylaminobutyl-3')-2-isopropy-    lidenhydrazin destilliert bei   12mm    Hg bei   87-89     als farblose   Fliissigkeit.      n2r,    = 1, 4575.



   Das Dipikrat kristallisiert aus Methanol in Prismen, die bei   132-134     unter Zersetzung schmelzen.



   Zu einer L¯sung von 46 g   l-(l'-Dimethylamino-      butyl-3')-2-isopropylidenhydrazin    in 150 cm3 Chloroform lässt man unter Rühren bei 5  20 cm3 Acetylchlorid tropfen. Dann rührt man 16 Stunden bei Zimmertemperatur. Darauf gibt man unter Kühlen 200 cm3   30 /Oige Pottaschelösung    zu, trennt die Chloroformschicht ab und schüttelt die Pottaschelösung nochmals mit Chloroform aus. Nach Trocknen der vereinigten Chloroformextrakte wird das Lösungsmittel abgedampft und der Rückstand fraktioniert destilliert.   l-(l'-Dimethylaminobutyl-3')-1-    acetyl-2-isopropylidenhydrazin destilliert bei 0, 03 mm Hg von   86-87 .      nid =    1, 4690.



   Beispiel 6
In die L¯sung von 29 g   l-(l'-Diäthylaminobutyl-       3')-1-acetyl-2-isopropylidenhydrazin    in 150 cm3 Athanol und 5 cm3 Wasser leitet man bei 20  Chlorwasserstoff bis zur Sättigung ein. Man rührt noch 4 Stunden bei Zimmertemperatur und dampft an schliessend die L¯sung im Vakuum bei   30     ein. Der
Rückstand wird in wenig Wasser gelöst und die Lö- sung unter gutem Kühlen mit   50 feiger wässeriger     Kalilauge stark alkalisch gemacht. Man gibt anschlie ssend noch so viel festes KOH zu, bis sich eine ölige Substanz abgeschieden hat. Alsdann wird von aus  geschiedenem    Kaliumchlorid über hochgereinigter Diatomeenerde filtriert, und der Filterrückstand mit einem Gemisch aus gleichen Teilen Methylenchlorid und Äther gewaschen.

   Das Filtrat wird anschliessend mehrmals mit dem gleichen Lösungsmittelgemisch ausgeschüttelt. Die vereinigten organischen Fraktionen werden nach Trocknen über Pottasche eingedampft und der Rückstand zweimal fraktioniert destilliert.   1- (l'-Diäthylamino-butyl-3')-1-acetylhy-    drazin destilliert bei 0, 08 mm Hg bei   102-105 .      n 24    1, 4711.



   Das als Ausgangsmaterial verwendete   1- (l'-Di-      äthylaminobutyl-3')-l-acetyl-2-isopropylidenhydrazin    wird wie folgt hergestellt :
Man löst 29, 4 g   1-Diäthylaminobutyl-(3)-hydrazin    unter Kühlung in 50 cm3 Aceton, lässt die L¯sung über Nacht stehen, erhitzt dann 1 Stunde zum Sieden am Rückfluss, dampft das überschüssige Aceton ab und destilliert den Rückstand. 1-(1'-DiÏthylamino  butyl-3')-2-isopropylidenhydrazin    destilliert bei 11 mm Hg bei   104-107 .   



   Zu einer L¯sung von 34 g   l-(l'-Diäthylamino-      butyl-3')-2-isopropylidenhydrazin    in   100    cm3 abs.



  Chloroform lässt man unter Rühren bei   5O eine Lö-    sung von 11, 5 cm3 Acetylchlorid in 10 cm3 Chloroform zutropfen. Dann rührt man 17 Stunden bei Zimmertemperatur. Darauf gibt man unter guter Kühlung 100   cm3 30 /Oige wässerige    Kalilauge zu, trennt die Chloroformschicht ab und schüttelt die wässerige alkalische L¯sung noch dreimal mit Chloroform aus.



  Nach Trocknen der vereinigten Chloroformextrakte über Magnesiumsulfat wird das Lösungsmittel eingedampft und der Rückstand fraktioniert destilliert.



      1- (1'-Diäthylaminobutyl-3')-1-acetyl-2-isopropyliden-    hydrazin destilliert bei 0, 1 mm Hg bei   106-110 .   



   Beispiel 7
In die L¯sung von 21 g   l-(l'-Dimethylamino-      pentyl-4')-1-acetyl-2-isopropylidenhydrazin    in 120cm3 Athanol und 4   cm3    Wasser leitet man bei   15-20     Chlorwasserstoff bis zur Sättigung ein. Man lässt die L¯sung noch 4 Stunden bei Zimmertemperatur stehen und dampft die L¯sung anschliessend im Vakuum bei 30  ein. Der Rückstand wird in wenig Wasser gelöst und die L¯sung unter gutem Kühlen mit 50  /o iger wässeriger Kalilauge stark alkalisch gemacht. Man gibt anschliessend noch so viel festes KOH zu, bis sich eine ölige Substanz abgeschieden hat. Alsdann wird vom ausgefallenen Kaliumchlorid über hochgereinigter Diatomeenerde filtriert und der Filterrückstand gut mit einem Gemisch aus gleichen Teilen Methylenchlorid und Ather gewaschen.



  Aus dem Filtrat wird die organische Schicht abgetrennt und die wässerige Phase noch dreimal mit dem gleichen Lösungsmittelgemisch ausgeschüttelt.



  Die vereinigten organischen Fraktionen werden nach Trocknen über Pottasche eingedampft und der   Rück-    stand fraktioniert destilliert.   l-(l'-Dimethylamino-      pentyl-4')-1-acetylhydrazin    siedet unter einem Druck von 0, 15 mm Hg bei   106 .      n2r,    = 1, 4727.



   Das als Ausgangsmaterial verwendete 1-(1'-Dimethylaminopentyl-4')-1-acetyl-2-isopropylidenhydrazin wird wie folgt hergestellt :
Man löst 7, 8 g   1-Dimethylaminopentyl-4-hydrazin    unter Kühlung in 20   cm3    Aceton, lässt die L¯sung über Nacht bei Zimmertemperatur stehen, erhitzt dann   1    Stunde zum Sieden am Rückfluss, dampft das überschüssige Aceton ab und destilliert den Rückstand.   1- (l'-Dimethylaminopentyl-4')-2-iso-      propylidenhydrazin    siedet unter einem Druck von 12 mm Hg bei   104-106 .   



   Zu einer L¯sung von 17, 5 g   1- (I'-Dimethyl-    aminopentyl-4')-2-isopropylidenhydrazin in 50 cm3 abs. Chloroform lässt man unter Rühren bei 0-5  eine L¯sung von 6, 7 cm3 Acetylchlorid in 10 cm3 Chloroform zutropfen. Dann rührt man 17 Stunden bei Zimmertemperatur. Anschliessend versetzt man unter guter Kühlung mit 50 cm3   30",'oiger wässeri-    ger Kalilauge, trennt die Chloroformschicht ab und schüttelt die wässerige Phase noch dreimal mit Chloroform aus. Nach Trocknen der vereinigten Chloroformextrakte über Magnesiumsulfat wird das Lösungsmittel abgedampft und der Rückstand fraktioniert destilliert.   1-(1'-Dimethylaminopentyl-4')-1-    acetyl-2-isopropylidenhydrazin siedet unter einem Druck von 0, 08 mm   Hg bei 100-103 .   



   Beispiel 8
In die L¯sung von 18, 5 g   1- [I'- (N-Methyl-N-iso-      propyl)-aminobutyl-3']-1-acetyl-2-isopropyliden-    hydrazin in 95 cm3 Athanol und 3, 2 cm3 Wasser leitet man bei 20  Chlorwasserstoff bis zur Sättigung ein. Man rührt noch 4 Stunden bei Zimmertemperatur und dampft die L¯sung anschliessend im Vakuum bei 30  ein. Der Rückstand wird in wenig Wasser gelöst und die L¯sung unter gutem Kühlen mit   50 /0iger wässeriger    Kalilauge stark alkalisch gemacht. Man gibt anschliessend noch so viel festes KOH zu, bis sich eine ölige Substanz abgeschieden hat. Alsdann wird vom ausgeschiedenen Kaliumchlorid filtriert und der Filterrückstand gründlich mit Chloroform gewaschen. Aus dem Filtrat wird die organische Schicht abgetrennt und die wässerige Phase mit Chloroform extrahiert.

   Die vereinigten Chloroformauszüge werden nach Trocknen über Magnesiumsulfat eingedampft und der Rückstand fraktioniert destilliert. 1-   [1'-    (N-Methyl-N-isopropyl)  aminobutyl-3']-1-acetylhydrazin    siedet unter einem Druck von 0, 3 mm Hg bei   95-98 .   



   Das als Ausgangsmaterial verwendete   1- [l'- (N-      Methyl-N-isopropyl)-aminobutyl-3']-1-acetyl-2-iso-    propylidenhydrazin wird wie folgt hergestellt :
Man löst 18, 0 g   l-[l'-(N-Methyl-N-isopropyl)-    aminobutyl-3']-hydrazin in 30 cm3 Aceton, erhitzt die L¯sung 3 Stunden zum Sieden am Rückfluss, dampft das überschüssige Aceton ab und destilliert den Rückstand.   l-[l'-(N-Methyl-N-isopropyl)-amino-    butyl-3']-2-isopropylidenhydrazin destilliert bei 11 mm Hg bei   103-108 .   



   22, 0 g 1-   [1'-    (N-Methyl-N-isopropyl)-amino  butyl-3']-2-isopropylidenhydrazin    werden tropfenweise mit 9, 0 g Acetylchlorid versetzt. Nach dreistündigem Stehen bei Zimmertemperatur wird die Lösung mit 30%iger wÏsseriger Kalilauge stark alkalisch gemacht und anschliessend dreimal mit Chloroform extrahiert. Die vereinigten Chloroformauszüge werden über Magnesiumsulfat getrocknet, das Lösungsmittel verdampft und der Rückstand destilliert.   1- [1'-    (N-Methyl-N-isopropyl)-amino  butyl-3']-1-acetyl-2-isopropylidenhydrazin    siedet unter einem Druck von 0, 3 mm Hg bei   102-106 .  



  



  Process for the preparation of new hydrazine derivatives
The present invention relates to a process for the preparation of new hydrazine derivatives of the formula I,

 where A is an alkylene group, Ri and R2 are lower alkyl groups and R3 is an acyl group, the groups A, Rt and R together containing at least 5 carbon atoms.



   According to the invention, the new hydrazine derivatives are prepared by converting isopropylidene hydrazine derivatives of the formula II,

 splitting off the isopropylidene group.



   This cleavage can be carried out in such a way that the aqueous alcoholic solution of the starting product is saturated with hydrogen chloride gas at a temperature of 0-40. To complete the reaction, the mixture is left to stand for a further 4 hours at room temperature, then concentrated and made alkaline with vigorous cooling. The end product is then extracted from the mixture with an organic inert solvent, preferably diethyl ether, and purified by high vacuum distillation.



   The new hydrazine derivatives prepared according to the invention are valuable intermediates for the manufacture of medicaments. Their reaction with reactive mono- or disubstituted malonic acid derivatives leads to l-amino-2,4-dioxoazetidine derivatives of the formula III,

 where A, Ri, R2 and R3 have the same meaning as in formula I. These azetidines are valuable medicaments. They have an anti-inflammatory effect and can be used in the treatment of rheumatic diseases. Some azetidine derivatives are several times more effective than phenylbutazone.



   It is particularly important for the therapeutic use of these compounds that they contain a basic group. For this reason they can form salts with inorganic or organic acids which are soluble in water. On the one hand, this facilitates the absorption of these drugs in the case of peroral medication, but on the other hand, it also enables the production of highly concentrated solutions, which in many cases are essential for parenteral shock therapy.



   In the following examples, the temperatures are given in degrees Celsius.



   Example 1
A solution of 4.0 g of 1- (2'-diethylamino-isohexyl-l ') - l-acetyl-2-isopropylidene hydrazine in 28 cm3 of 86% aqueous ¯thanol is saturated with hydrogen chloride gas while cooling at 0 and then 4 Left to stand for hours at room temperature. The solution is then strongly concentrated in vacuo at 30, the residue is taken up in 20 cm3 of ice water and made alkaline with gusty aqueous potassium hydroxide solution while being strongly cooled.



  You shake out a total of 100 cm3 of ether in portions, dry the combined ether extracts over magnesium sulphate and remove the ether in vacuo. The residue is distilled in a high vacuum, the 1- (2'-diethylamino-isohexyl-1 ') -1-acetylhydrazine distilling over between 126-128 at 1.5 mm Hg as colorless oil. nD = 1,4629.



   The 1- (2'-diethylamino-isohexyl-1 ') -1-acetyl-2-isopropylidene hydrazine used as starting material is prepared as follows:
1.0 g of 1- (2'-diethylamino-isohexyl-l ') - hydrazine is dissolved in 5 cm3 of acetone, whereupon heating occurs. The reaction solution is left to stand for 16 hours at room temperature and then refluxed for a further 1/2 hour. The acetone is then distilled off and the residue is fractionated in vacuo, the 1- (2'-diethylamino-isohexyl-1 ') -2-isopropylidene hydrazine distilling over at 110 at 9 mm Hg as a colorless liquid. n22 D = 1.4532.



   To a solution of 2.3 g of 1- (2'-DiÏthylaminoisohexyl-1 ') -2-isopropylidene hydrazine in 10 cm3 abs.



  A solution of 0.83 cm3 of acetyl chloride in 5 cm3 of abs is slowly allowed to chloroform at 0 with vigorous cooling and stirring. Add chloroform dropwise.



  The reaction solution is then stirred for a further 3 hours at +3 to +5 and then left to stand for 15 hours at room temperature. The reaction solution is then cooled to 0 and washed twice with 25 cm3 each of ice-cooled, aqueous, saturated soda solution. The chloroform solution is dried over magnesium sulfate, the chloroform is removed in vacuo and the residue is distilled in vacuo, the 1- (2'-diethylamino-isohexyl-1 ') -1-acetyl-2-isopropylidene hydrazine between 119-121 at 1 mm Hg as slightly yellow colored 01 passes over. n22 D = 1,4669.



   Example 2
A solution of 4.9 g of l- (-diethylaminoethyl) -1-acetyl-2-isopropylidene hydrazine in 28 cm3 of 85 tiger ethanol is saturated with hydrogen chloride gas while cooling at 0 and then left to stand at 220 for 4 hours. The ethanol is then removed in vacuo at 30, the oily residue is taken up in 20 cm3 of water and made alkaline with 50% strength aqueous potassium hydroxide solution while cooling vigorously. It is extracted with a total of 100 cm3 of ether, the combined ether extracts are dried over magnesium sulfate, the ether is evaporated and the residue is distilled in vacuo, the 1- (¯-diethylaminoethyl) -1-acetylhydrazine passing over as colorless oil at 106 at 1 mm Hg. n22 D = 1,4682.



   The l- (jd-diethylaminoethyl) -1-acetyl-2-isopropylidene hydrazine used as starting material is prepared as follows:
3.4 g of iS-diethylaminoethylhydrazine are dissolved in 20 cm3 of acetone, whereupon heating occurs.



  The reaction solution is left to stand for 15 minutes at room temperature and then refluxed for a further 1/2 hour. The acetone is then distilled off and the residue is fractionated in vacuo, the 1- (¯-diethylaminoethyl) -2-isopropylidene hydrazine distilling over between 86 and 87 at 9 mm Hg. n 2 = 1.4561.



   To a solution of 14.0 g of 1- (/ 3-diethylaminoethyl) -2-isopropylidene hydrazine in 40 cm3 abs. A solution of 6.5 cm3 acetyl chloride in 15 cm3 abs is slowly allowed to chloroform at 0 with vigorous cooling and stirring. Add chloroform dropwise. The reaction solution is then stirred for a further 3 hours at a temperature of + 2 to + 5 and then left to stand for 20 hours at room temperature. The reaction solution is then cooled to 0 and washed twice with 25 cm3 of aqueous saturated soda solution each time.

   The chloroform solution is dried over magnesium sulfate, the chloroform is removed in vacuo, and the residue is distilled in vacuo, the 1- (ss-diethylaminoethyl) -1-acetyl-2-isopropylidene hydrazine between 103-105 at 1 mm Hg as a colorless liquid passes over. nD = 1.4721.



   Example 3
16.2 g of 1- (dimethylamino-isopropyl) -l-acetyl-2-isopropylidene hydrazine are dissolved in a mixture of 72 cm3 of ethanol and 8 cm3 of water. At a temperature of 30-35 hydrogen chloride is passed into this solution until it is saturated and the mixture is left to stand at this temperature for 3 hours.



  Then it is evaporated in a vacuum, the oily residue is dissolved in a little water and 50% potassium hydroxide solution is added with good cooling until an oil separates out. This is taken up in ether, the ethereal solution dried over magnesium sulphate and then evaporated. The residue is fractionated at 11 mm Hg, some 1- (dimethylaminoisopropyl) -2-isopropylidene hydrazine from 63-68, then from 117-119 1- (dimethylamino-isopropyl) -l-acetylhydrazine distilling.



   With phenyl isothiocyanate, this compound forms 1- (dimethylamino-isopropyl) -l-acetyl-4-phenyl-thiosemicarbazide, which, after crystallization from ethyl acetate, melts at 159-160 with decomposition.



   The 1- (dimethylamino-isopropyl) -1-acetyl-2-isopropylidene hydrazine used as starting material is prepared as follows:
75 g of dimethylaminoisopropylene hydrazine are added to 200 cm3 of acetone with cooling and left to stand overnight at room temperature. The reaction solution is then refluxed for 1 hour, the excess acetone is removed in vacuo and the residue is distilled in vacuo, the 1- (dimethylamino-isopropyl) -2-isopropylidene hydrazine passing over between 63-64 at 10 mm Hg.



   Dipicrate, prisms of m.p. 144-148 (from methanol).



   To dissolve 33 g of 1- (dimethylamino-isopropyl) -2-isopropylidene hydrazine in 100 cm3 of abs. Add chloroform dropwise with stirring at 0-10 17 cm3 acetyl chloride. After stirring for 16 hours at room temperature, 150 cm3 of 30 / ae potash solution are added dropwise to the solution with good cooling, then the chloroform layer is separated off and the aqueous phase is extracted twice more with chloroform. After the combined chloroform extracts have been dried over magnesium sulfate and the chloroform has been evaporated, the residue is fractionated in a high vacuum, with 1- (dimethylamino-isopropyl) -1-acetyl-2-isopropylidene hydrazine at 0.2 mm Hg between 76-82 as yellow oil.



   Example 4
A solution of 7.9 g of 1- (diethylamino-isopropyl) -1 - acetyl - 2 - isopropylidene hydrazine in 36 cm3 of ethanol and 4 cm3 of water is saturated with HCl gas at a temperature of 30-35. The mixture is then left to stand for 4 hours at room temperature, the solution is evaporated in vacuo, the residue is treated with ice water and 500% aqueous potassium hydroxide solution is added with good cooling until a 51 separates out. This is taken up in ether, the ethereal solution dried over magnesium sulphate and evaporated. The residue is distilled, the 1- (diethylamino-isopropyl) -l-acetylhydrazine passing over as colorless oil at an air bath temperature of 120-140 at 12 mm Hg.



   For characterization, phenylthiosemicarbazide is produced with phenyl isothiocyanate. M.p.



  140-141 from ethyl acetate ether.



   The 1- (diethylamino-isopropyl) -1-acetyl-2-isopropylidene hydrazine used as starting material is prepared as follows:
8.3 g of diethylaminoisopropylidene hydrazine are dissolved in 25 cm 3 of acetone, the temperature rising to 30. It is left to stand overnight, then the solution is refluxed for 1 hour and the acetone is evaporated off. The residue is distilled in vacuo, the 1- (diethylamino-isopropyl) -2-isopropylidene hydrazine passing over between 77-80 at 10 mm Hg.



   The dipicrate, recrystallized from methanol, melts at 124-128 (dec.).



   To a solution of 9.0 g of l- (diethylamino-isopropyl) -2-isopropylidene hydrazine in 30 cm3 of abs.



  Chloroform is a solution of 4 cm3 acetyl chloride in 10 cm3 abs.



  Chloroform added. The mixture is stirred for 3 hours at 5, left to stand overnight at room temperature, the reaction solution is cooled to 0 and shaken with Potash solution. The chloroform solution is dried over magnesium sulfate, the chloroform is evaporated and the residue is distilled, the 1- (diethylamino-isopropyl) -1-acetyl-2-isopropylidene hydrazine passing over between 93-95 at 0.2 mm Hg.



   Example 5
The solution of 19.4 g of 1- (l'-dimethylamino-butyl-3 ') -1-acetyl-2-isopropylidene hydrazine in 100 cm of 95% ethanol is introduced at 15-20 g of hydrogen chloride until saturation . After standing for 4 hours at room temperature, the solution is evaporated in vacuo. The residue is dissolved in a little water, and pigerous potassium hydroxide solution is added with good cooling. The oil which separates out is taken up in ether, the ether solution is dried over sodium sulfate and evaporated, and the residue is fractionally distilled.



      1- (1'-Dimethylaminobutyl-3 ') -1-acetylhydrazine distilled at 12 mm Hg from 114-115. n = 1,4685.



   The one made from it with phenyl isothiocyanate
1- (1'-Dimethylaminobutyl-3 ') -1-acetyl-4-phenyl-thiosemicarbazide melts after crystallization from chloro form / ether at 132 with decomposition.



   The 1- (1'-dimethylaminobutyl-3 ') -1-acetyl-2-isopropylidene hydrazine used as starting material is prepared as follows:
38.2 g of 1-dimethylaminobutyl-3-hydrazine are dissolved in 100 cm3 of acetone with cooling, the solution is left to stand at room temperature for 18 hours, then heated to reflux for 1 hour, the solvent is evaporated and the distilled Residue. 1- (1'-Dimethylaminobutyl-3 ') -2-isopropylidene hydrazine distills at 12 mm Hg at 87-89 as a colorless liquid. n2r, = 1.4575.



   The dipicrate crystallizes from methanol in prisms which melt at 132-134 with decomposition.



   To a solution of 46 g of 1- (l'-dimethylaminobutyl-3 ') - 2-isopropylidene hydrazine in 150 cm3 of chloroform, while stirring at 5, 20 cm3 of acetyl chloride are added dropwise. The mixture is then stirred for 16 hours at room temperature. 200 cm3 of 30% potash solution are then added with cooling, the chloroform layer is separated off and the potash solution is shaken out again with chloroform. After the combined chloroform extracts have been dried, the solvent is evaporated off and the residue is fractionally distilled. 1- (l'-Dimethylaminobutyl-3 ') -1-acetyl-2-isopropylidene hydrazine distilled at 0.03 mm Hg from 86-87. nid = 1,4690.



   Example 6
The solution of 29 g of l- (l'-diethylaminobutyl-3 ') -1-acetyl-2-isopropylidene hydrazine in 150 cm3 of ethanol and 5 cm3 of water is introduced at 20 cm of hydrogen chloride until it is saturated. The mixture is stirred for a further 4 hours at room temperature and the solution is then evaporated in vacuo at 30. Of the
The residue is dissolved in a little water and the solution is made strongly alkaline with good cooling with 50% aqueous potassium hydroxide solution. You then add enough solid KOH until an oily substance has separated out. The precipitated potassium chloride is then filtered through highly purified diatomaceous earth, and the filter residue is washed with a mixture of equal parts of methylene chloride and ether.

   The filtrate is then extracted several times with the same solvent mixture. After drying over potash, the combined organic fractions are evaporated and the residue is fractionally distilled twice. 1- (l'-Diethylamino-butyl-3 ') -1-acetylhydrazine distilled at 0.08 mm Hg at 102-105. n 24 1, 4711.



   The 1- (l'-diethylaminobutyl-3 ') - l-acetyl-2-isopropylidene hydrazine used as starting material is prepared as follows:
29.4 g of 1-diethylaminobutyl (3) hydrazine are dissolved in 50 cm3 of acetone with cooling, the solution is left to stand overnight, then heated to reflux for 1 hour, the excess acetone is evaporated and the residue is distilled. 1- (1'-DiÏthylamino butyl-3 ') -2-isopropylidene hydrazine distilled at 11 mm Hg at 104-107.



   To a solution of 34 g of l- (l'-diethylaminobutyl-3 ') - 2-isopropylidene hydrazine in 100 cm3 abs.



  Chloroform is allowed to drop in a solution of 11.5 cm3 of acetyl chloride in 10 cm3 of chloroform with stirring at 50. The mixture is then stirred for 17 hours at room temperature. 100 cm3 of 30% aqueous potassium hydroxide solution are then added with good cooling, the chloroform layer is separated off and the aqueous alkaline solution is shaken out three more times with chloroform.



  After the combined chloroform extracts have been dried over magnesium sulfate, the solvent is evaporated and the residue is fractionally distilled.



      1- (1'-Diethylaminobutyl-3 ') -1-acetyl-2-isopropylidene hydrazine distilled at 0.1 mm Hg at 106-110.



   Example 7
Into the solution of 21 g of l- (l'-dimethylaminopentyl-4 ') -1-acetyl-2-isopropylidene hydrazine in 120 cm3 of ethanol and 4 cm3 of water, 15-20 g of hydrogen chloride are introduced until saturation. The solution is left to stand for 4 hours at room temperature and the solution is then evaporated in vacuo at 30. The residue is dissolved in a little water and the solution is made strongly alkaline with 50% aqueous potassium hydroxide solution while cooling well. You then add enough solid KOH until an oily substance has separated out. The precipitated potassium chloride is then filtered through highly purified diatomaceous earth and the filter residue is washed thoroughly with a mixture of equal parts of methylene chloride and ether.



  The organic layer is separated off from the filtrate and the aqueous phase is extracted three more times with the same solvent mixture.



  After drying over potash, the combined organic fractions are evaporated and the residue is fractionally distilled. l- (l'-Dimethylaminopentyl-4 ') -1-acetylhydrazine boils at 106 under a pressure of 0.15 mm Hg. n2r, = 1.4727.



   The 1- (1'-dimethylaminopentyl-4 ') - 1-acetyl-2-isopropylidene hydrazine used as starting material is prepared as follows:
7.8 g of 1-dimethylaminopentyl-4-hydrazine are dissolved in 20 cm3 of acetone while cooling, the solution is left to stand overnight at room temperature, then heated to reflux for 1 hour, the excess acetone is evaporated and the residue is distilled. 1- (1'-Dimethylaminopentyl-4 ') -2-isopropylidene hydrazine boils at 104-106 under a pressure of 12 mm Hg.



   To a solution of 17.5 g of 1- (I'-dimethylaminopentyl-4 ') -2-isopropylidene hydrazine in 50 cm3 abs. A solution of 6.7 cm3 of acetyl chloride in 10 cm3 of chloroform is added dropwise to chloroform while stirring at 0-5. The mixture is then stirred for 17 hours at room temperature. Then 50 cm3 of 30 "aqueous potassium hydroxide solution is added with good cooling, the chloroform layer is separated off and the aqueous phase is extracted three times with chloroform. After the combined chloroform extracts have been dried over magnesium sulfate, the solvent is evaporated and the residue is fractionally distilled 1- (1'-Dimethylaminopentyl-4 ') -1-acetyl-2-isopropylidene hydrazine boils under a pressure of 0.08 mm Hg at 100-103.



   Example 8
In the solution of 18.5 g of 1- [I'- (N-methyl-N-isopropyl) aminobutyl-3 '] -1-acetyl-2-isopropylidene hydrazine in 95 cm3 of ethanol and 3, 2 cm3 of water is passed in at 20 hydrogen chloride until it is saturated. The mixture is stirred for a further 4 hours at room temperature and the solution is then evaporated at 30 in a vacuum. The residue is dissolved in a little water and the solution is made strongly alkaline with good cooling with 50/0 aqueous potassium hydroxide solution. You then add enough solid KOH until an oily substance has separated out. The precipitated potassium chloride is then filtered off and the filter residue is washed thoroughly with chloroform. The organic layer is separated off from the filtrate and the aqueous phase is extracted with chloroform.

   The combined chloroform extracts are evaporated after drying over magnesium sulfate and the residue is fractionally distilled. 1- [1'- (N-Methyl-N-isopropyl) aminobutyl-3 '] -1-acetylhydrazine boils at 95-98 under a pressure of 0.3 mm Hg.



   The 1- [l'- (N-methyl-N-isopropyl) -aminobutyl-3 '] -1-acetyl-2-isopropylidene hydrazine used as starting material is prepared as follows:
18.0 g of 1- [l '- (N-methyl-N-isopropyl) - aminobutyl-3'] - hydrazine are dissolved in 30 cm3 of acetone, the solution is heated to reflux for 3 hours, and the excess acetone is evaporated and the residue is distilled off. 1- [l '- (N-methyl-N-isopropyl) -aminobutyl-3'] - 2-isopropylidene hydrazine distilled at 11 mm Hg at 103-108.



   22.0 g of 1- [1'- (N-methyl-N-isopropyl) -amino butyl-3 '] -2-isopropylidene hydrazine are added dropwise with 9.0 g of acetyl chloride. After standing for three hours at room temperature, the solution is made strongly alkaline with 30% strength aqueous potassium hydroxide solution and then extracted three times with chloroform. The combined chloroform extracts are dried over magnesium sulphate, the solvent is evaporated and the residue is distilled. 1- [1'- (N-Methyl-N-isopropyl) -amino butyl-3 '] -1-acetyl-2-isopropylidene hydrazine boils under a pressure of 0.3 mm Hg at 102-106.

 

Claims (1)

PATENTANSPRUCH Verfahren zur Herstellung von neuen Hydrazin Derivaten der Formel I, EMI5.1 worin A eine Alkylengruppe, Ri und R2 niedere Alkylgruppen und R3 eine Acylgruppe bedeuten, wobei die Gruppen A, Ri und R2 zusammen mindestens 5 Kohlenstoffatome enthalten, dadurch gekennzeichnet, dass man aus Isopropylidenhydrazin- Derivaten der Formel II, EMI5.2 die Isopropylidengruppe abspaltet.
