Description

  

  Verfahren     zur    Herstellung von neuen Estern der     Dehydrocholsäure       Die vorliegende Erfindung     bezieht    sich auf ein  Verfahren zur Herstellung von neuen     Estern        der        De-          hydrocholsäure    mit     einem        Monohydroxyderivat    des       1-Methyl-4-isopropyl-cyclohexans        bzw.    des     1-Methyl-          4-isopropyl-benzols,

          also        auf        die    Herstellung des Car  vomentholesters, des     Mentholesters,        Thymolesters     bzw.     Carvacrolesters    der     Dehydrocholsäure.     



  Die neuen Ester besitzen wertvolle Eigenschaften.  Sie finden Verwendung in der Therapie verschieden  ster     Erkrankungen        der        Leber.        Ein.    Vorteil der neuen  Ester     liegt    im     langanhaltenden,    also     protrahierten     Effekt.  



  Die     neuen    Ester sollen     vorzugsweise    als     Arznei-          mittel    verwendet werden. Die neuen     Ester    werden  durch     Veresterung    der     Dehydrocholsäure    mit einem       Monohydroxyderivat    des     1-Methyl-4-isopropyl-cyclo-          hexans    bzw. des     1-Methyl-4-isopropyl-benzols    gewon  nen.  



  Die     Veresterung    erfolgt z. B. nach     den        für    die  Herstellung von     Dehydrocholsäureestern    üblichen  Methoden, z. B. indem man     Dehydrocholsäure    und  den     entsprechenden    Alkohol in     Gegenwart        eines    sau  ren     Katalysators    erhitzt, oder aber indem     man    ein       Dehydrocholsäurehalogenid,    vorzugsweise das Chlo  rid, mit dem entsprechenden Alkohol umsetzt.  



       Gemäss    der ersten     Variante    kann man beispiels  weise eine Schmelze von     Dehydrocholsäure    und Men  thol bzw.     Carvomenthol    bzw.     Thymol        bzw.        Carvaerol     herstellen; in diese     Schmelze    kann man     zwecks    Er  leichterung der Wasserabspaltung     gasförmige    Salz  säure einleiten. Man     kann    aber auch katalytische  Mengen einer     organischem        Sulfonsäure,    z.

   B.     p-To-          luo,Isulfons;äure,    zusetzen. Das Wasser     kann    fortlau  fend aus dem Gleichgewicht entfernt werden, z. B.  durch Zusatz eines     Entrainers    wie Benzol,     usf.     



       Gemäss    der zweiten Variante ist es beispielsweise         möglich,        Dehydrocholsäurechlorid    mit einem der ge  nannten     Alkohole    zur     Umsetzung    zu bringen. Hier  bei kann man in An- oder Abwesenheit von Lö  sungsmitteln arbeiten. Als     Lösungsmittel    kann man  z.

   B.     verwenden:        Benzol,        Äther,        Dioxan,        usf.    Man       kann    den     abgespaltenen    Chlorwasserstoff durch Er  wärmen     wegtreiben.        oder    aber durch Zusatz eines       basischen    Kondensationsmittels binden.

   Als basische  Kondensationsmittel kann man     verwenden:        tertiäre     Basen wie     Triäthylamin,        Pyridin,        Äthyldüsopropyl-          amin,        Alkalialkoholate    wie     Na-methanolat,        Na-äthan-          olat,        usf.     



       Selbstverständlich    kann jede andere für diesen  Zweck     brauchbare        Veresterungsmethode    verwendet  werden. So kann     man    z. B. ein gemischtes     Anhydrid     der     Dehydrocholsäure,    z.

   B. ein     Schwefelsäureanhy-          drid,        Phosphorsäure-anhydrid    oder auch     ein        Anhydrid     mit einem     Kohlensäurehalbester    mit     einem        der   <B>ge-</B>  nannten     Alkohole    zur Umsetzung     bringen.     



  Die Herstellung     solcher    gemischter     Anhydride    er  folgt z. B. nach bekannten     Methoden.    Sie sind z. B.  für die Herstellung     gemischter        Säureanhydride    in der       Patentliteratur    der letzten Jahre in     allen        Einzelheiten          beschrieben    worden.  



  Es sei noch     erwähnt,    dass man     selbstverständlich     auch ein     funktionelles    Derivat von Menthol,     Carvo-          menthol,        Thymol    bzw.     Carvacrol,    z. B. einen     organi-          schen        Sulfonsäureester    wie das,     Mesylat    oder     Tosylat     mit     Dehydrocholsäure    oder einem Salz der     Dehydro-          cholsäure,    umsetzen     kann.     



  Man kann     auch    auf eine Lösung oder     Suspension     der     Dehydrocholsäure    und     eine    der genannten     Hy-          droxykörper    ein     wasserabspaltendes    Mittel wie z. B.

         ein,        Carbodiimid,        beispielsweise        Di-cyclohexyl-carbo-          dürrdd,    einwirken     lassen.         <I>Ausführungsbeispiel</I>       Äquimolekulare    Mengen     Dehydrocholsäure    und       Menthol    werden in einem     Schliffkolben,    versehen mit       Rückflusskühler    und zwei     seitlichen        Ansätzen    für  Thermometer und     Gaseinleitungsrohr,

  Der     so    erhaltene     Dehydrocholsäure-mentholester     schmilzt bei 197,5-199  C;     er    ist in Wasser praktisch  nicht löslich, gut     löslich        dagegen,    in Alkohol,     Benzol     und     Chloroform.     



       Die    übrigen Ester, der     Carvomentholester,    der       Thymolester    oder der     Carvacrolester    können in ähn  licher Weise, wie oben im einzelnen     beschrieben,    her  gestellt werden.

PATENTANSPRUCH Verfahren zur Herstellung von neuen Estern der Dehydrocholsäure mit einem Monohydroxyderivat des 1-Methyl-4-isopropyl-cyclohexans bzw. des 1-Methyl- 4-isopropyl-benzols, dadurch gekennzeichnet, dass man Dehydrocholsäure mit einem Monohydroxyderi- vat des 1-Methyl-4-isopropyl-cyclohexans bzw. des 1-Methyl-4-iso#propyl-benzols verestert. UNTERANSPRÜCHE 1.
