Verfahren zur Herstellung neuer Äther
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Alkyläthern des d6-Dehydro-des- (acetylamino)-colchicins der Formeln

(Iso-Reihe) worin R einen Alkylrest, wie einen Niederalkylrest, z. B. einen Methyl-, Äthyl-, Propyl-, Butyl-, Pentyloder Hexylrest, bedeutet.
Die neuen Verbindungen besitzen eine cytostatische und mitosehemmende Wirkung. Sie hemmen die Zellteilung, indem sie die Ausbildung der Tei lungsspindel (Metaphase) während des Kernteilungsvorgangs verhindern. Sie können daher als Heilmittel für Mensch und Tier, z. B. zur Behandlung von myeloischen Leukämien, besonders von subakuten und chronischen Verlaufsformen, von Gichtanfällen oder von Präkanzerosen oder Kanzerosen der Haut, z. B. Keratoma senile, Epitheiioma baso- oder spinocellulare, Morbus Bowen oder Rezidiven nach Röntgenbestrahlung von Hauttumoren, Verwendung finden. Die neuen Verbindungen können auch bei der Pflanzenzüchtung Verwendung finden um polyploide Pflanzen herzustellen.
Besonders wirksam sind die Verbindungen der normalen Reihe, welche wesentlich wirksamer sind als Colchicin oder Desacetylmethylcolchicin.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen besteht darin, dass man eine Verbindung der Formel

worin X eine reaktionsfähig veresterte Hydroxylgruppe bedeutet, oder die entsprechende Isoverbindung oder ein Gemisch beider Verbindungen mit einem Alkohol der Formel ROH oder einem Salz davon umsetzt.
Als Salze dieser Alkohole kommen insbesondere Alkalialkoholate in Betracht.
Eine reaktionsfähig veresterte Hydroxylgruppe ist insbesondere eine mit einer starken anorganischen oder organischen Säure veresterte Hydroxylgruppe, wie ein Halogenatom, z. B. Chlor, oder eine Arylsulfonyloxygruppe, wie die Toluolsulfonyloxygruppe.
Die genannten Reaktionen können in an sich bekannter Weise in Ab-oder Anwesenheit von Lösungs- und/oder Verdünnungsmitteln und/oder Kondensationsmitteln bei erniedrigter, normaler oder erhöhter Temperatur und im offenen oder geschlossenen Gefäss durchgeführt werden.
Wenn erwünscht, kann man das gegebenenfalls erhaltene Gemisch der isomeren Alkyläther auftrennen. Diese Trennung kann in an sich bekannter Weise erfolgen, z. B. durch fraktionierte Kristallisation und/oder Adsorption wird zweckmässig als Chromatographie, vorzugsweise an Aluminiumoxyd, durchgeführt. Als Eluierungsmittel verwendet man beispielsweise Essigester-Benzol oder Essigester. Aus den erhaltenen Fraktionen können die Äther durch Kristallisation in reiner Form erhalten werden. Der Gehalt der einzelnen Fraktionen an den isomeren Athern kann bequem durch Dünnschichtchromatographie verfolgt werden.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden gewonnen werden.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, oder als Mittel für die Zucht polyploider Pflanzen Verwendung finden, welche sie in Mischung mit einem organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
Die Präparate zur Verwendung zur Züchtung von polyploiden Pflanzen können z. B. in fester Form oder als Lösungen, Suspensionen oder Emulsionen vorliegen. Als Trägermaterialien kommen beispielsweise Wasser, Agar, Glycerin oder Lanolin in Frage.
Gegebenenfalls können die Präparate noch weitere Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz-oder Emulgiermittel enthalten. Sie können auch andere zur Aufzucht von Pflanzen geeignete Mittel enthalten. Präparate in flüssiger Form können beispielsweise zum Spritzen der Pflanzen verwendet werden. Präparate in Salbenform können der Pflanze direkt verabreicht werden. Die Pflanze kann aber z. B. auch mit einer den Wirkstoff enthaltenden Lösung bepinselt oder in eine solche Lösung eingetaucht werden. Die Dosierung und die Zeitdauer während der man das Präparat einwirken lässt, variiert je nach der Art der Verabreichungsform und Pflanzenart. Da die Toxizität gegenüber Pflanzen im allgemeinen wesentlich geringer ist, als gegenüber Mensch und Tier, können entsprechend höhere Dosen appliziert werden.
In dem folgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.

werden in einem Bombenrohr bei 80-85 4 Stunden lang mit 2 ml einer gesättigten methanolischen Na triummethylatlösung erhitzt. Das alkalische Reaktionsgemisch wird auf Eis gegossen und mit Chloroform ausgezogen. Die neutral gewaschene und getrocknete Chloroformlösung liefert beim Eindampfen 8 mg Neutralteile, welche gemäss dem IR-Spektrum Z-Dehydro-des-(acetylamino)-colchicin enthalten.

Zu einer Lösung von 1,6 g zl6-Dehydro-des- (acetylamino)-colchicin in 310 ml absolutem Benzol werden 0,714 ml Thionylchlorid (rein) zugegeben und 3 Stunden am Rückfluss gekocht. Die Reaktionslösung wird am Rotationsverdampfer zur Trockene eingedampft und der Rückstand wird an Aluminiumoxyd (Aktivität II) chromatographiert. Mit Benzol, Benzol-Methylenchlorid 3 : 1 und 1:1 werden 0, 5100 g Chlortropon der Formel

The new compounds have a cytostatic and mitosis-inhibiting effect. They inhibit cell division by preventing the development of the division spindle (metaphase) during the process of nuclear division. They can therefore be used as a remedy for humans and animals, e.g. B. for the treatment of myeloid leukemia, especially of subacute and chronic forms, of gout attacks or of precancerous or cancerous skin, z.
