Description
Verfahren zur Herstellung eines gegen Erkrankungen der Mundhöhle wirkenden Mittels
Gegenstand der Erfindung ist ein mikrobiologisches Verfahren zur Herstellung eines gegen Erkrankungen der Mundhöhle, besonders gegen Aphthen wirkenden Mittels.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man die bei Parodontose, Gingivitis, Stomatitis und Aphthen vorhandenen Mundbakterien, die beispielsweise in Form eines Abstriches oder auf andere Weise aus der Mundhöhle entnommen wurden, bei pH 5 bis 7 in einem pulverisierten Rohkaffee, Kohlehydrate und Stickstoffquellen enthaltenden Nährmedium unter nichtsterilen Bedingungen durch Oberflächen- oder Submerskultur fermentiert, das Kulturmedium filtriert und das Filtrat und gegebenenfalls auch das Mycel aufarbeitet.
Nach entsprechend langem Wachstum bei vorzugsweise etwa 18 bis 24 C, gewöhnlich nach etwa 3 Wochen, ist genügend Perkolat erzeugt, worauf das Kulturmedium nach dem Filtrieren in übliche flüssige oder feste Verarbeitungsformen übergeführt werden kann. Das beim Filtrieren zurückbleibende Mycel kann zu einer Salbe verarbeitet werden.
Eine besonders vorteilhafte Ausführungsform besteht darin, dass man Wasser mit pulverisiertem Rohkaffee und danach mit Haferflocken versetzt.
Zum besseren Aufschluss dieser Rohstoffe, insbesondere des Rohkaffees, sollte während der Zugabe dieser Stoffe das Wasser auf einer Temperatur von bis zu etwa 1000 C gehalten werden. Darauf stellt man den pH-Wert, z. B. mit Weinsäure, auf 5 bis 7 ein und beimpft das so hergestellte Nährmedium nach dem Abkühlen auf etwa 70" C oder darunter mit den Bakterien.
Zur Durchführung des Verfahrens können die angegebenen Stoffe in etwa den folgenden Mengenverhältnissen verwendet werden: 2 1 Wasser, 250 g Haferflocken, 100 g Rohkaffee, 15 g Weinsäure.
Der so hergestellte, physiologisch einwandfreie, d. h. keine unerwünschten Nebenwirkungen hervor rufende Wirkstoff stellt ein ausgezeichnetes Mittel zur Bekämpfung von Krankheiten der Mundhöhle, insbesondere von Aphthen, Parodontose, Gingivitis, Stomatitis und jeglichen Arten von Ulcerationen dar.
Er besitzt gleichzeitig analgetische Eigenschaften und kann intern oder extern angewandt werden.
Klinische Untersuchungen haben gezeigt, dass sich der nach dem erfindungsgemässen Verfahren erzeugte Wirkstoff zur Behandlung aller entzündlichen Prozesse an der Mundschleimhaut, einschliesslich sämtlicher Zahlfleischentzündiingen, insbesondere der Gingivitis ulcerosa, und bei dem fast bei allen Paror dontosen vorhandenen Entzündungszustand der marginalen Gingiva eignet. Die Behandlung mit dem Wirkstoff führt bei den genannten entzündlichen Zuständen zu einer weitestgehenden Besserung bzw., falls eine Wiederherstellung überhaupt möglich ist, zu einer völligen Heilung. Ganz hervorragend aber bewährte sich seine Anwendung zur Bekämpfung aller Fälle von Aphthen.
Der nach dem erfindungsgemässen Verfahren erzeugte Wirkstoff zeigt ausserdem überraschende desodorierende Eigenschaften gegenüber den verschiedenartigsten, als unangenehm empfundenen Ge rüchen, z. B. Mundgerüchen. Falls die desodorierende Wirkung gesteigert werden soll, kann man dem Nährmedium vor der Fermentation noch Chlorophyll oder Chlorophyll enthaltende Substanzen zugeben.
Man hat schon ein Rohkaffee und Stärke enthaltendes Medium einer Fermentation unter nichtsterilen Bedingungen ausgesetzt. Dieses enthielt jedoch ausserdem Schwefel und war vor der Einwirkung der von aussen spontan zutretenden Mikroorganismen nicht mit Bakterien beimpft worden. Als Fermentationsprodukt wurde dementsprechend ein völlig anderer, nämlich rheumaspezifischer Wirkstoff mit pharmakologischem Histamingehalt erhalten, der mit dem erfindungsgemäss erhaltenen Wirkstoff nicht vergleichbar ist; seine Herstellung konnte im Hinblick auf das differenzierte Verhalten von Mikroorganismen unter wechselnden Bedingungen auch nicht aus dem bekannten Verfahren abgeleitet werden.

The subject of the invention is a microbiological process for the production of an agent which is effective against diseases of the oral cavity, especially against aphthous ulcers.
The method according to the invention is characterized in that the oral bacteria present in periodontal disease, gingivitis, stomatitis and aphthae, which were taken from the oral cavity, for example in the form of a smear or in some other way, at pH 5 to 7 in a pulverized green coffee, carbohydrates and nitrogen sources containing nutrient medium fermented under non-sterile conditions by surface or submerged culture, the culture medium filtered and the filtrate and optionally also the mycelium worked up.
After a correspondingly long period of growth at preferably about 18 to 24 C, usually after about 3 weeks, sufficient percolate has been produced, whereupon the culture medium can be converted into conventional liquid or solid processing forms after filtering. The mycelium that remains after the filtration can be processed into an ointment.
A particularly advantageous embodiment consists in adding pulverized raw coffee to water and then adding oat flakes.
For better digestion of these raw materials, in particular the raw coffee, the water should be kept at a temperature of up to about 1000 C while these substances are being added. The pH value is then set, e.g. B. with tartaric acid, to 5 to 7 and inoculate the nutrient medium produced in this way after cooling to about 70 ° C or below with the bacteria.
To carry out the process, the specified substances can be used in approximately the following proportions: 2 liters of water, 250 g of oat flakes, 100 g of green coffee, 15 g of tartaric acid.
The physiologically flawless, d. H. Active ingredient that does not cause undesirable side effects is an excellent means of combating diseases of the oral cavity, in particular aphthae, periodontal disease, gingivitis, stomatitis and all kinds of ulcerations.
It also has analgesic properties and can be used internally or externally.
Clinical investigations have shown that the active ingredient produced by the method according to the invention is suitable for the treatment of all inflammatory processes on the oral mucosa, including all inflammation of the flesh, in particular ulcerative gingivitis, and in the inflammation of the marginal gingiva that is present in almost all periodontoses. Treatment with the active ingredient leads to the most extensive improvement in the mentioned inflammatory conditions or, if recovery is at all possible, to complete healing. However, its use for combating all cases of aphthae has proven to be very good.
The active ingredient produced by the process according to the invention also shows surprising deodorant properties against the most varied of odors that are perceived as unpleasant Ge, eg. B. Bad breath. If the deodorizing effect is to be increased, chlorophyll or substances containing chlorophyll can also be added to the nutrient medium before fermentation.
A medium containing green coffee and starch has already been subjected to fermentation under non-sterile conditions. However, this also contained sulfur and had not been inoculated with bacteria prior to the action of the microorganisms spontaneously entering from the outside. A completely different, namely rheumatoid-specific active ingredient with a pharmacological histamine content, which is not comparable with the active ingredient obtained according to the invention, was accordingly obtained as the fermentation product; its production could not be derived from the known process with regard to the differentiated behavior of microorganisms under changing conditions.
